The study evaluated the skin anti-aging activity of Astragalus sarcocolla leaves extract(ASE)by assessing its antioxidant and inhibitory effect activity on matrix metalloproteinase(MMP),collagenase,elastase,hyaluronid...The study evaluated the skin anti-aging activity of Astragalus sarcocolla leaves extract(ASE)by assessing its antioxidant and inhibitory effect activity on matrix metalloproteinase(MMP),collagenase,elastase,hyaluronidase,and tyrosinase in relation to its chemical composition.Ultra Performance Liquid Chromatography-Mass Spectrometry(UPLC-MS)identified 27 metabolites(15 flavonoids,8 phenolic acids and their derivatives,and 4 coumarins).ASE showed strong antioxidant capacity in DPPH(IC_(50)value of 26.05μg/mL)and FRAP(2433μM FeSO_(4)/g extract)assays.The extract inhibited MMP-1 and MMP-9 in a concentration-dependent manner and suppressed collagenase,elastase,hyaluronidase,and tyrosinase activities(IC_(50)=35.038,40.748,61.389,and 30.980μg/mL,respectively).A network pharmacology study was conducted to uncover the mechanisms responsible for skin anti-aging effects,and molecular docking further evaluated interactions of key metabolites with hub targets.Twenty-one bioactive metabolites,selected based on oral bioavailability and drug-likeness,highlighted cinnamic acid,acacetin,luteolin,kaempferol,and apigenin as key compounds.MMP-9,ESR1,PTGS-2,and EGFR were identified as main targets.Docking studies revealed that acacetin and apigenin have stronger binding affinities to MMP-9,PTGS-2,and EGFR than other constituents.These findings suggest that ASE may serve as a natural multi-target skin anti-aging remedy with potential cosmetic applications.展开更多
Background Organic selenium(Se)has gained recognition in poultry nutrition as a feed additive to boost production and Se deposition in eggs and tissues,owing to its high bioavailability,efficient tissue accumulation a...Background Organic selenium(Se)has gained recognition in poultry nutrition as a feed additive to boost production and Se deposition in eggs and tissues,owing to its high bioavailability,efficient tissue accumulation and minimal toxicity.Selenium-enriched yeast(SeY)is a well-established source,while selenium-enriched lactobacilli(SeL),a newer alternative,offers the added benefits of probiotics.This study examined the effects of SeY and SeL on egg quality,antioxidant capacity,Se deposition,and gut health in laying hens.After a two-week pre-treatment with a Sedeficient diet(SeD),450 Hy-Line Brown laying hens(30-week-old)were assigned into five dietary groups with six replicates of 15 hens each.The groups included a SeD,SeD supplemented with 1.5 mg Se/kg from SeY(SeY15),or 1.5,3.0,and 6.0 mg Se/kg from SeL(SeL15,SeL30,SeL60).The feeding trial lasted for 12 weeks.Results SeY15 and SeL15 improved the feed-to-egg ratio(P<0.05)in the latter stages.Haugh units were significantly increased(P<0.05)in the SeY15 and SeL30 groups,while darker yolk color(P<0.05)was observed in the SeY15,SeL15,and SeL60 groups.All Se-supplemented diets increased Se content in whole eggs,albumen,and yolk(P<0.05),while SeL groups showed a dose-dependent effect.Antioxidant enzyme activities increased,and MDA content decreased in the serum(P<0.05),with SeY15 showing the highest GSH-Px levels(P<0.05).SeL60 increased serum alkaline phosphatase and aspartate transaminase,and distorted the liver architecture(P<0.05).Se-diets reduced concentrations of reactive oxygen species(ROS)in the ileum and liver(P<0.05).SeL15 improved the ileal villus height-tocrypt depth ratio(P<0.05).SeY15 and/or SeL15 up-regulated TXNRD1 and SEPHS1 mRNA while down-regulating SCLY expression in the liver.SeY15 altered ileal microbiota by increasing both beneficial and pathogenic bacteria,whereas SeL15 predominantly boosted beneficial bacteria.Conclusion SeL integrates the antioxidant properties of organic Se with the probiotic benefits on gut health,resulting in a performance-enhancing effect comparable to that of SeY.However,high SeL level(6.0 mg Se/kg)compromised productivity and metabolic functions while enhancing Se deposition.展开更多
Background:Cisplatin(DDP)has been used in the treatment of various human cancers.However,DDP alone lacks efficacy in treating triple-negative breast cancer(TNBC),and its clinical application is often hampered by side ...Background:Cisplatin(DDP)has been used in the treatment of various human cancers.However,DDP alone lacks efficacy in treating triple-negative breast cancer(TNBC),and its clinical application is often hampered by side effects.Astragalus polysaccharide(APS)is one of the active components extracted from Astragalus membranaceus and has gained attention for its various biological properties.This research is aimed to evaluate the effectiveness of a combination of APS and DDP on TNBC and explore the potential mechanisms.Methods:The efficacy and mechanisms of single or combined treatment were evaluated using Cell Counting Kit-8(CCK8)assay,Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)staining,wound healing assay,trans-well invasion/migration assay,hematoxylin-eosin(HE)staining,immunohistochemical(IHC)staining,Western Blot(WB)analysis,and fluorescence-activated cell sorting(FACS).An orthotopic model of TNBC was used to assess the in vivo treatment efficacy of single or combination treatment.Results:APS significantly enhanced the anti-proliferative,anti-migratory,and anti-invasive effects of DDP on TNBC cells.The combination of APS and DDP downregulated anti-apoptotic genes(Bcl2 and Bcl-xL)while upregulating pro-apoptotic genes(Puma,Cle-Caspase3,Cle-PARP),leading to enhanced apoptosis.This combination treatment increased E-cadherin levels,decreased Vimentin,Snail,Slug,and Twist levels,and effectively suppressed epithelial-mesenchymal transition(EMT)-associated cell invasion.In the orthotopic model of TNBC,a synergistic reduction in tumor growth was observed in mice treated with APS and DDP.Additionally,the combination of APS and DDP induced the infiltration of CD8+T lymphocytes into the tumor immune microenvironment.Conclusion:The combination of APS and DDP exhibits more potent tumor inhibition and anti-tumor immunity than either agent alone,representing a novel approach to enhance therapeutic efficacy without increasing the side effects of DDP.展开更多
Astragalus extract Astragalus polysaccharide(APS),a natural extract,has been demonstrated to exert inhibitory effects on the development of various tumors by modulating microRNA(miRNA).However,the precise regulatory m...Astragalus extract Astragalus polysaccharide(APS),a natural extract,has been demonstrated to exert inhibitory effects on the development of various tumors by modulating microRNA(miRNA).However,the precise regulatory mechanism of miRNA-223(miR-223)in brain glioma cells remains unclear.This study aimed to investigate the inhibitory effect of Astragalus extract APS on brain glioma cells through the miR-223/FBXW7 signaling pathway and its potential mechanism.Using the TargetScan tool,we predicted the binding between miR-223 and FBXW7 and confirmed this binding relationship through dual luciferase assay.We compared the expression of miR-223 and FBXW7 in glioma and adjacent tissues and followed up the prognosis.U87 cells were subjected to APS treatment or were transfected with small interfering RNA(siRNA)of miR-223 mimics to examine the effects of these treatments on cell proliferation,apoptosis,and FBXW7 expression.Our results demonstrated that miR-223 directly bound to FBXW7,as confirmed by dual luciferase assay.Moreover,the mRNA expression of miR-223 in glioma tissues was higher than that in paracancerous tissues,while the mRNA and protein levels of FBXW7 in glioma tissues were lower than those in paracancerous tissues.In addition,we observed a negative correlation between the expression of miR-223 and FBXW7(P<0.05).APS significantly inhibited the proliferation and invasion of U87 cells,suppressed the expression of miR-223,and promoted the expression of FBXW7(P<0.05).Transfection of miR-223 mimic into cells reversed the inhibition of miR-223 and the increase of FBXW7 induced by APS(P<0.05).Therefore,our findings suggested that APS might inhibit the proliferation and invasion of the glioma cell line U87 through the miR-223/FBXW7 signaling pathway.展开更多
According to literary data,there are 189 species of the genus Astragalus that inhabit the territory of Kyrgyzstan.The distribution of species of the genus Astragalus(Fabaceae)was studied by taking into account their o...According to literary data,there are 189 species of the genus Astragalus that inhabit the territory of Kyrgyzstan.The distribution of species of the genus Astragalus(Fabaceae)was studied by taking into account their occurrence in soil-climatic and relief types of plant habitats in the mountainous territories of Kyrgyzstan.The materials were collected during organized research expeditions to natural areas of plant habitats.The occurrence of species was studied by well-known methods.As a result of such analysis we have made the following conclusions for the first time:in the flora of Kyrgyzstan there are 189 species of the genus Astragalus,they are representatives of 7 subgenera of this genus;species occur in 24 types of plant habitats with soil and relief features,taking into account the altitudinal regularity of taxa distribution in mountain phytocenoses;subgenuses and specific species characterized by broad,smallest and intermediate sizes of habitats were identified;groupings of plant habitat types on the territory of Kyrgyzstan characterized by the largest,smallest and intermediate indicators on the number of subgenuses and species of the genus Astragalus were identified.The botanical description of each species has been supplemented with new information about the species as a taxonomic unit.The materials of our work can be successfully used in the field of systematics,evolution and ecology of plants.展开更多

[Objectives]To investigate the content and distribution of inorganic elements in Astragalus membranaceus sourced from various regions in Gansu Province.[Methods]28 batches of A.membranaceus samples were collected and ...[Objectives]To investigate the content and distribution of inorganic elements in Astragalus membranaceus sourced from various regions in Gansu Province.[Methods]28 batches of A.membranaceus samples were collected and subsequently digested using the Multiwave 7000 super microwave digestion system.The contents of aluminum(Al),barium(Ba),beryllium(Be),cobalt(Co),chromium(Cr),iron(Fe),gallium(Ga),magnesium(Mg),manganese(Mn),nickel(Ni),antimony(Sb),tin(Sn),strontium(Sr),titanium(Ti),thallium(Tl),vanadium(V),and zinc(Zn)were quantified utilizing a PerkinElmer 2000 inductively coupled plasma mass spectrometer.Principal component analysis was performed utilizing SPSS 25.0 to identify the distinctive characteristic elements of A.membranaceus.Additionally,systematic cluster analysis was conducted using these characteristic elements as variables to investigate the relationship between the primary inorganic elements and the geographical origin of A.membranaceus.[Results]17 inorganic elements were identified in A.membranaceus specimens collected from Gansu Province,with characteristic elements including Ba,Co,Fe,Ga,Mn,Zn,and Sn.The contents of inorganic elements in various sources of A.membranaceus exhibited significant variability and demonstrated distinct clustering characteristics.[Conclusions]A.membranaceus,originating from Gansu Province,exhibits a high content of inorganic elements.However,variations in ecological environments can lead to differences in the specific inorganic elements that are enriched.This study aims to provide a reference for the further development and application of A.membranaceus.展开更多
Objective:To improve the efficacy of Astragalus membranaceus(Fisch.)Bunge(A.membranaceus,Huang Qi),and to further develop and utilize it,fermentation technology was applied to the stem and leaf of A.membranaceus to en...Objective:To improve the efficacy of Astragalus membranaceus(Fisch.)Bunge(A.membranaceus,Huang Qi),and to further develop and utilize it,fermentation technology was applied to the stem and leaf of A.membranaceus to enhance its immune function.Methods:In this study,we fermented A.membranaceus stem and leaf(ASL)with probiotics and inves-tigated its immune function.Firstly,we screened suitable strains for ASL fermentation and optimized the fermentation process.Secondly,we determined the antioxidant capacity of fermented ASL and its effect on inflammation in mouse monocyte-macrophage cell.Finally,the immunocompromised mice were treated with fermented ASL to investigate the changes in their immune ability.Results:Among the 10 selected probiotics,Lactobacillus plantarum was the most suitable strain for ASL fermentation.After optimization of the fermentation process,the content of saponins in fermented ASL was significantly increased.The fermented ASL exhibited strong anti-inflammatory and antioxidant activities in vitro.The in vivo immune efficacy improved by promoting the development of the spleen and thymus,as well as raising the immunoglobulin M,tumor necrosis factor-α,and interleukin-1β levels of in the serum.Conclusion:This study contributes to developing the non-medicinal parts of A.membranaceus,expands its medicinal resources,highlights the potential of fermentation technology to enhance these parts,and provides a reference for further development.Based on this approach,we can promote using non-me-dicinal parts of herbal medicines,minimize drug waste,and offer a reference for developing non-me-dicinal components in Chinese herbal medicines.展开更多
Objective This study aimed to investigate the effect of Astragalus(AST)on osteoporosis(OP)and the downstream mechanisms.Methods Human bone marrow-derived mesenchymal stem cells(hBMSCs)were induced to differentiate int...Objective This study aimed to investigate the effect of Astragalus(AST)on osteoporosis(OP)and the downstream mechanisms.Methods Human bone marrow-derived mesenchymal stem cells(hBMSCs)were induced to differentiate into osteogenic cells.After transfection with relevant plasmids,cell proliferation,cell cycle progression,and apoptosis were assessed.Alizarin red staining was used to detect calcium nodules in the cells,alkaline phosphatase(ALP)staining was used to detect ALP activity in the cells,and quantitative reverse transcription-polymerase chain reaction and western blotting were used to determine RUNX2 and Osterix expression levels.An OP rat model was established using ovariectomy and micro-computed tomography scanning.Hematoxylin and eosin staining and Masson’s trichrome staining were used to evaluate the pathological conditions of bone tissues,while immunohistochemistry was conducted to detect RUNX2 in bone tissues.Results AST promoted the osteogenic differentiation of BMSCs,reduced miR-181d-5p expression levels,and increased SOX11 expression levels.Restoring miR-181d-5p expression or reducing SOX11 expression levels reversed the effects of AST on the osteogenic differentiation of hBMSCs.miR-181d-5p was found to target SOX11 in hBMSCs.AST improved OP in rats,and miR-181d-5p overexpression or SOX11 inhibition reversed the therapeutic effects of AST on OP in rats.Conclusion AST promoted the osteogenic differentiation of hBMSCs and alleviated OP by targeting SOX11 via miR-181d-5p.展开更多
Background:To investigate the effects of Astragalus polysaccharides(APS)on melanoma-bearing mice via the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods:Fifty ma...Background:To investigate the effects of Astragalus polysaccharides(APS)on melanoma-bearing mice via the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods:Fifty male C57BL/6 mice were randomly divided into five groups:model group,positive control group,APS low-dose group,APS middle-dose group,and APS high-dose group(10 mice per group).B16 cell-bearing melanoma mouse models were established in all groups.The low,middle,and high-dose APS groups received daily injections of APS solution at 20,40,and 80 mg/kg,respectively.Spleen and thymus indices were measured,tumor volumes were observed,and tumor inhibition rates were calculated.Tumor histopathological changes were examined via H&E staining.Ki67 and PCNA expression in tumor tissues were assessed via immunofluorescence staining and Western blot analysis RT-qPCR.Western blot was used to examine the expression of JAK2/STAT3 signaling pathway-related factors in tumor tissues.Results:APS significantly increased the spleen and thymus indices in melanoma mice and inhibited tumor growth.H&E staining revealed that APS significantly reduced the number of tumor cells in melanoma mice.Immunofluorescence results indicated that APS significantly decreased Ki67-positive expression in tumor tissues,while Western blot results showed that APS significantly reduced PCNA protein expression in tumor tissues.RT-qPCR and Western blot results indicated that APS significantly suppressed the expression of JAK2/STAT3 signaling pathway-related genes and proteins.Conclusion:APS may inhibit tumor growth in a melanoma mouse model by enhancing immune function through suppression of the JAK2/STAT3 signaling pathway.展开更多

The results of phylogenctic analyses suggest that the subgenus Pogonophace is not a monophyletic group. Sect. Sesbanella (Astragalus hoantchy and A. dshimensis) and other species of Pogonophace are nested within different major clades in the phylogenetic tree. The species of sections Bibracteola, Phyllolobium and Trichostylus compose a monophyletic group with a close relationship to subtribe Coluteinae instead of Astragalus. Astragalus tribulifolius and A. tanguticus might be a vicariant species pair as inferred by the ITS data.展开更多

The results showed that 10 highly reproducible ISSR fragments a-mong 40 primers were screened. Using these primers, a total of 684 ISSR frag-ments from 500 to 3 000 bp were amplified, and 59.2% of them showed polymor-phic by unweighted pair-group method with arithmetic (UPGMA) analysis. It revealed that the 22 accessions had a similarity range from 0.63 to 0.95, and existed biolog-ical diversities. Based on cluster and principal coordinate analyses, al accessions could be divided into four distinct groups.展开更多

The results sho...Five Chinese milk vetch(Astragalus sinicus) varieties were selected to make a comparison in order to screen effective varieties which could suit for the double-rice cropping system of Hunan Province. The results showed that the growth period of Xiangzi 2 is shorter than that of Xiangfei 2, Xiangfei 3 and Yujiangdaye,and is similar to that of Xinyang Chinese milk vetch. The full-bloom stage of Xiangzi 2 is also similar to Xinyang variety and obviously earlier than that of the other three varieties. The fresh grass yield of Xiangzi 2 in full-bloom stage is 23 842.5kg/hm^2, which increases by 39.9% compared with Xinyang variety. Nitrogen content in the fresh grass of Xiangzi 2 is higher than that of the other four varieties. In conclusion, Xiangzi 2 is an extremely early-blossoming and early-maturing Chinese milk vetch variety with moderate fresh grass yield, high nutrient contents, stable characters and wide adaptability. So, Xiangzi 2 is suitable for planting in double-rice cropping region in Hunan Province and other same latitude regions which have similar ecological conditions.展开更多

文摘Background Organic selenium(Se)has gained recognition in poultry nutrition as a feed additive to boost production and Se deposition in eggs and tissues,owing to its high bioavailability,efficient tissue accumulation and minimal toxicity.Selenium-enriched yeast(SeY)is a well-established source,while selenium-enriched lactobacilli(SeL),a newer alternative,offers the added benefits of probiotics.This study examined the effects of SeY and SeL on egg quality,antioxidant capacity,Se deposition,and gut health in laying hens.After a two-week pre-treatment with a Sedeficient diet(SeD),450 Hy-Line Brown laying hens(30-week-old)were assigned into five dietary groups with six replicates of 15 hens each.The groups included a SeD,SeD supplemented with 1.5 mg Se/kg from SeY(SeY15),or 1.5,3.0,and 6.0 mg Se/kg from SeL(SeL15,SeL30,SeL60).The feeding trial lasted for 12 weeks.Results SeY15 and SeL15 improved the feed-to-egg ratio(P<0.05)in the latter stages.Haugh units were significantly increased(P<0.05)in the SeY15 and SeL30 groups,while darker yolk color(P<0.05)was observed in the SeY15,SeL15,and SeL60 groups.All Se-supplemented diets increased Se content in whole eggs,albumen,and yolk(P<0.05),while SeL groups showed a dose-dependent effect.Antioxidant enzyme activities increased,and MDA content decreased in the serum(P<0.05),with SeY15 showing the highest GSH-Px levels(P<0.05).SeL60 increased serum alkaline phosphatase and aspartate transaminase,and distorted the liver architecture(P<0.05).Se-diets reduced concentrations of reactive oxygen species(ROS)in the ileum and liver(P<0.05).SeL15 improved the ileal villus height-tocrypt depth ratio(P<0.05).SeY15 and/or SeL15 up-regulated TXNRD1 and SEPHS1 mRNA while down-regulating SCLY expression in the liver.SeY15 altered ileal microbiota by increasing both beneficial and pathogenic bacteria,whereas SeL15 predominantly boosted beneficial bacteria.Conclusion SeL integrates the antioxidant properties of organic Se with the probiotic benefits on gut health,resulting in a performance-enhancing effect comparable to that of SeY.However,high SeL level(6.0 mg Se/kg)compromised productivity and metabolic functions while enhancing Se deposition.

文摘Background:Cisplatin(DDP)has been used in the treatment of various human cancers.However,DDP alone lacks efficacy in treating triple-negative breast cancer(TNBC),and its clinical application is often hampered by side effects.Astragalus polysaccharide(APS)is one of the active components extracted from Astragalus membranaceus and has gained attention for its various biological properties.This research is aimed to evaluate the effectiveness of a combination of APS and DDP on TNBC and explore the potential mechanisms.Methods:The efficacy and mechanisms of single or combined treatment were evaluated using Cell Counting Kit-8(CCK8)assay,Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)staining,wound healing assay,trans-well invasion/migration assay,hematoxylin-eosin(HE)staining,immunohistochemical(IHC)staining,Western Blot(WB)analysis,and fluorescence-activated cell sorting(FACS).An orthotopic model of TNBC was used to assess the in vivo treatment efficacy of single or combination treatment.Results:APS significantly enhanced the anti-proliferative,anti-migratory,and anti-invasive effects of DDP on TNBC cells.The combination of APS and DDP downregulated anti-apoptotic genes(Bcl2 and Bcl-xL)while upregulating pro-apoptotic genes(Puma,Cle-Caspase3,Cle-PARP),leading to enhanced apoptosis.This combination treatment increased E-cadherin levels,decreased Vimentin,Snail,Slug,and Twist levels,and effectively suppressed epithelial-mesenchymal transition(EMT)-associated cell invasion.In the orthotopic model of TNBC,a synergistic reduction in tumor growth was observed in mice treated with APS and DDP.Additionally,the combination of APS and DDP induced the infiltration of CD8+T lymphocytes into the tumor immune microenvironment.Conclusion:The combination of APS and DDP exhibits more potent tumor inhibition and anti-tumor immunity than either agent alone,representing a novel approach to enhance therapeutic efficacy without increasing the side effects of DDP.

文摘Astragalus extract Astragalus polysaccharide(APS),a natural extract,has been demonstrated to exert inhibitory effects on the development of various tumors by modulating microRNA(miRNA).However,the precise regulatory mechanism of miRNA-223(miR-223)in brain glioma cells remains unclear.This study aimed to investigate the inhibitory effect of Astragalus extract APS on brain glioma cells through the miR-223/FBXW7 signaling pathway and its potential mechanism.Using the TargetScan tool,we predicted the binding between miR-223 and FBXW7 and confirmed this binding relationship through dual luciferase assay.We compared the expression of miR-223 and FBXW7 in glioma and adjacent tissues and followed up the prognosis.U87 cells were subjected to APS treatment or were transfected with small interfering RNA(siRNA)of miR-223 mimics to examine the effects of these treatments on cell proliferation,apoptosis,and FBXW7 expression.Our results demonstrated that miR-223 directly bound to FBXW7,as confirmed by dual luciferase assay.Moreover,the mRNA expression of miR-223 in glioma tissues was higher than that in paracancerous tissues,while the mRNA and protein levels of FBXW7 in glioma tissues were lower than those in paracancerous tissues.In addition,we observed a negative correlation between the expression of miR-223 and FBXW7(P<0.05).APS significantly inhibited the proliferation and invasion of U87 cells,suppressed the expression of miR-223,and promoted the expression of FBXW7(P<0.05).Transfection of miR-223 mimic into cells reversed the inhibition of miR-223 and the increase of FBXW7 induced by APS(P<0.05).Therefore,our findings suggested that APS might inhibit the proliferation and invasion of the glioma cell line U87 through the miR-223/FBXW7 signaling pathway.
文摘According to literary data,there are 189 species of the genus Astragalus that inhabit the territory of Kyrgyzstan.The distribution of species of the genus Astragalus(Fabaceae)was studied by taking into account their occurrence in soil-climatic and relief types of plant habitats in the mountainous territories of Kyrgyzstan.The materials were collected during organized research expeditions to natural areas of plant habitats.The occurrence of species was studied by well-known methods.As a result of such analysis we have made the following conclusions for the first time:in the flora of Kyrgyzstan there are 189 species of the genus Astragalus,they are representatives of 7 subgenera of this genus;species occur in 24 types of plant habitats with soil and relief features,taking into account the altitudinal regularity of taxa distribution in mountain phytocenoses;subgenuses and specific species characterized by broad,smallest and intermediate sizes of habitats were identified;groupings of plant habitat types on the territory of Kyrgyzstan characterized by the largest,smallest and intermediate indicators on the number of subgenuses and species of the genus Astragalus were identified.The botanical description of each species has been supplemented with new information about the species as a taxonomic unit.The materials of our work can be successfully used in the field of systematics,evolution and ecology of plants.
文摘The research on astragalus polysaccharides(APS)in pancreatic cancer has garnered increasing interest due to its potential therapeutic benefits.APS,derived from the Astragalus membranaceus plant,exhibits diverse biological activities,including immune modulation and anti-tumor effects.In this study,we presented a retrospective analysis study exploring the potential benefits of combining APS with gemcitabine plus tegafur gimeracil oteracil potassium capsule(S-1)in pancreatic cancer treatment.The finding suggests that APS may enhance treatment efficacy,improve immune function,and reduce adverse reactions,offering a valuable complementary approach to standard chemotherapy.Further research is needed to validate these findings and optimize APS usage in clinical practice.This combination therapy represents a compelling avenue for improving pancreatic cancer management and may ultimately contribute to better patient outcomes.

文摘[Objectives]To investigate the content and distribution of inorganic elements in Astragalus membranaceus sourced from various regions in Gansu Province.[Methods]28 batches of A.membranaceus samples were collected and subsequently digested using the Multiwave 7000 super microwave digestion system.The contents of aluminum(Al),barium(Ba),beryllium(Be),cobalt(Co),chromium(Cr),iron(Fe),gallium(Ga),magnesium(Mg),manganese(Mn),nickel(Ni),antimony(Sb),tin(Sn),strontium(Sr),titanium(Ti),thallium(Tl),vanadium(V),and zinc(Zn)were quantified utilizing a PerkinElmer 2000 inductively coupled plasma mass spectrometer.Principal component analysis was performed utilizing SPSS 25.0 to identify the distinctive characteristic elements of A.membranaceus.Additionally,systematic cluster analysis was conducted using these characteristic elements as variables to investigate the relationship between the primary inorganic elements and the geographical origin of A.membranaceus.[Results]17 inorganic elements were identified in A.membranaceus specimens collected from Gansu Province,with characteristic elements including Ba,Co,Fe,Ga,Mn,Zn,and Sn.The contents of inorganic elements in various sources of A.membranaceus exhibited significant variability and demonstrated distinct clustering characteristics.[Conclusions]A.membranaceus,originating from Gansu Province,exhibits a high content of inorganic elements.However,variations in ecological environments can lead to differences in the specific inorganic elements that are enriched.This study aims to provide a reference for the further development and application of A.membranaceus.

文摘Objective:To improve the efficacy of Astragalus membranaceus(Fisch.)Bunge(A.membranaceus,Huang Qi),and to further develop and utilize it,fermentation technology was applied to the stem and leaf of A.membranaceus to enhance its immune function.Methods:In this study,we fermented A.membranaceus stem and leaf(ASL)with probiotics and inves-tigated its immune function.Firstly,we screened suitable strains for ASL fermentation and optimized the fermentation process.Secondly,we determined the antioxidant capacity of fermented ASL and its effect on inflammation in mouse monocyte-macrophage cell.Finally,the immunocompromised mice were treated with fermented ASL to investigate the changes in their immune ability.Results:Among the 10 selected probiotics,Lactobacillus plantarum was the most suitable strain for ASL fermentation.After optimization of the fermentation process,the content of saponins in fermented ASL was significantly increased.The fermented ASL exhibited strong anti-inflammatory and antioxidant activities in vitro.The in vivo immune efficacy improved by promoting the development of the spleen and thymus,as well as raising the immunoglobulin M,tumor necrosis factor-α,and interleukin-1β levels of in the serum.Conclusion:This study contributes to developing the non-medicinal parts of A.membranaceus,expands its medicinal resources,highlights the potential of fermentation technology to enhance these parts,and provides a reference for further development.Based on this approach,we can promote using non-me-dicinal parts of herbal medicines,minimize drug waste,and offer a reference for developing non-me-dicinal components in Chinese herbal medicines.

文摘Objective This study aimed to investigate the effect of Astragalus(AST)on osteoporosis(OP)and the downstream mechanisms.Methods Human bone marrow-derived mesenchymal stem cells(hBMSCs)were induced to differentiate into osteogenic cells.After transfection with relevant plasmids,cell proliferation,cell cycle progression,and apoptosis were assessed.Alizarin red staining was used to detect calcium nodules in the cells,alkaline phosphatase(ALP)staining was used to detect ALP activity in the cells,and quantitative reverse transcription-polymerase chain reaction and western blotting were used to determine RUNX2 and Osterix expression levels.An OP rat model was established using ovariectomy and micro-computed tomography scanning.Hematoxylin and eosin staining and Masson’s trichrome staining were used to evaluate the pathological conditions of bone tissues,while immunohistochemistry was conducted to detect RUNX2 in bone tissues.Results AST promoted the osteogenic differentiation of BMSCs,reduced miR-181d-5p expression levels,and increased SOX11 expression levels.Restoring miR-181d-5p expression or reducing SOX11 expression levels reversed the effects of AST on the osteogenic differentiation of hBMSCs.miR-181d-5p was found to target SOX11 in hBMSCs.AST improved OP in rats,and miR-181d-5p overexpression or SOX11 inhibition reversed the therapeutic effects of AST on OP in rats.Conclusion AST promoted the osteogenic differentiation of hBMSCs and alleviated OP by targeting SOX11 via miR-181d-5p.

文摘Background:To investigate the effects of Astragalus polysaccharides(APS)on melanoma-bearing mice via the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods:Fifty male C57BL/6 mice were randomly divided into five groups:model group,positive control group,APS low-dose group,APS middle-dose group,and APS high-dose group(10 mice per group).B16 cell-bearing melanoma mouse models were established in all groups.The low,middle,and high-dose APS groups received daily injections of APS solution at 20,40,and 80 mg/kg,respectively.Spleen and thymus indices were measured,tumor volumes were observed,and tumor inhibition rates were calculated.Tumor histopathological changes were examined via H&E staining.Ki67 and PCNA expression in tumor tissues were assessed via immunofluorescence staining and Western blot analysis RT-qPCR.Western blot was used to examine the expression of JAK2/STAT3 signaling pathway-related factors in tumor tissues.Results:APS significantly increased the spleen and thymus indices in melanoma mice and inhibited tumor growth.H&E staining revealed that APS significantly reduced the number of tumor cells in melanoma mice.Immunofluorescence results indicated that APS significantly decreased Ki67-positive expression in tumor tissues,while Western blot results showed that APS significantly reduced PCNA protein expression in tumor tissues.RT-qPCR and Western blot results indicated that APS significantly suppressed the expression of JAK2/STAT3 signaling pathway-related genes and proteins.Conclusion:APS may inhibit tumor growth in a melanoma mouse model by enhancing immune function through suppression of the JAK2/STAT3 signaling pathway.
文摘The internal transcribed spacers (ITS) (including ITS1, 5.8S rRNA gene and ITS2) of Caragana roborovskyi and eight species of subgenus Pogonophace ( Astragalus) were sequenced, and analyzed together with other ITS data (from GenBank) of 48 species representing Astragalus and 12 related genera. The results of phylogenctic analyses suggest that the subgenus Pogonophace is not a monophyletic group. Sect. Sesbanella (Astragalus hoantchy and A. dshimensis) and other species of Pogonophace are nested within different major clades in the phylogenetic tree. The species of sections Bibracteola, Phyllolobium and Trichostylus compose a monophyletic group with a close relationship to subtribe Coluteinae instead of Astragalus. Astragalus tribulifolius and A. tanguticus might be a vicariant species pair as inferred by the ITS data.

文摘Two new isoflavones (8, 3'-dihydroxy-7,4'-dimethoxyisoflavone, odoratin-7-0-[3-D-glu-copyranoside) and four known isoflavones (formononetin, 7,3'-dihydroxy-8,4'-dimethoxyisoflavone, calycosin, calycosin-7-0-(3-D-glucopyranoside) were isolated from the roots of Astragalus mem-branaceus (Fisch.) Bunge. Their structures were established by spectral analysis.

文摘Genetic relationships among 22 accessions of Astragalus sinicus L. col ect-ed from different provinces of China were analyzed by inter-simple sequence repeat (ISSR) markers. The results showed that 10 highly reproducible ISSR fragments a-mong 40 primers were screened. Using these primers, a total of 684 ISSR frag-ments from 500 to 3 000 bp were amplified, and 59.2% of them showed polymor-phic by unweighted pair-group method with arithmetic (UPGMA) analysis. It revealed that the 22 accessions had a similarity range from 0.63 to 0.95, and existed biolog-ical diversities. Based on cluster and principal coordinate analyses, al accessions could be divided into four distinct groups.

文摘Five Chinese milk vetch(Astragalus sinicus) varieties were selected to make a comparison in order to screen effective varieties which could suit for the double-rice cropping system of Hunan Province. The results showed that the growth period of Xiangzi 2 is shorter than that of Xiangfei 2, Xiangfei 3 and Yujiangdaye,and is similar to that of Xinyang Chinese milk vetch. The full-bloom stage of Xiangzi 2 is also similar to Xinyang variety and obviously earlier than that of the other three varieties. The fresh grass yield of Xiangzi 2 in full-bloom stage is 23 842.5kg/hm^2, which increases by 39.9% compared with Xinyang variety. Nitrogen content in the fresh grass of Xiangzi 2 is higher than that of the other four varieties. In conclusion, Xiangzi 2 is an extremely early-blossoming and early-maturing Chinese milk vetch variety with moderate fresh grass yield, high nutrient contents, stable characters and wide adaptability. So, Xiangzi 2 is suitable for planting in double-rice cropping region in Hunan Province and other same latitude regions which have similar ecological conditions.
