In order to study the effects of Saussurea laniceps polysaccharides(SLPs)on the expression of inflammatory factors and antimicrobial peptide LL-37 in UVB-induced keratinocytes,SLPs were extracted by the ethanol therma...In order to study the effects of Saussurea laniceps polysaccharides(SLPs)on the expression of inflammatory factors and antimicrobial peptide LL-37 in UVB-induced keratinocytes,SLPs were extracted by the ethanol thermal reflux method,and SLPs at different concentrations were used to examine the inhibitory effect of COX-2(a key mediator of inflammatory pathway).A cell model of UVB irradiation-induced inflammation was established to determine the influence of SLPs on prostaglandin E2(PGE-2),TNF-αand IL-1βinflammatory factors,as well as the relationships of SLPs with LL-37 expression.An enzyme-linked immunosorbent assay(ELISA)and western blot analysis were used to detect the production of inflammatory factors and LL-37 antimicrobial peptide.The results showed that the inhibition rate of COX-2 was 82.41%at 1000μg/mL,and the expression of PGE-2,TNF-αand IL-1βinflammatory factors in HaCaT cells was significantly downregulated at 100μg/mL(P<0.01).In addition,SLPs at 50μg/mL and 100μg/mL concentrations enhanced the expression of LL-37 antimicrobial peptide(P<0.01),thereby down-regulating the expression of TNF-αand IL-1βinflammatory factors,then reducing skin inflammation.Conclusion:SLP can significantly inhibit the inflammatory response induced by UVB,and can further slow down the damage caused by inflammation to the skin by regulating LL-37 antimicrobial peptides,which has the potential to prevent skin inflammatory damage caused by UVB irradiation.展开更多

Objective:To investigate the anti-inflammatory effects of the total flavonoids from Saussurea involucrata on lipopolysaccharides(LPS)-stimulated murine RAW264.7 macrophages and explore its underlying mechanism of acti...Objective:To investigate the anti-inflammatory effects of the total flavonoids from Saussurea involucrata on lipopolysaccharides(LPS)-stimulated murine RAW264.7 macrophages and explore its underlying mechanism of action.Methods:Total flavonoids from Saussurea involucrata were extracted using chromatographic column method.Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.The production of nitric oxide was detected by Griess assay and the release of cytokines(IL-10 and TNF-α)and chemokines(MCP-1,MIP-1α,and CCL5/RANTES)was determined by ELISA to evaluate the anti-inflammatory activity of total flavonoids from Saussurea involucrata.Moreover,nuclear translocation of p65,c-Jun,and IRF3 was detected by immunofluorescence microscopy and Western blotting analysis was performed to determine the expression of related proteins.Results:Total flavonoids extracted from Saussurea involucrata were 751.5 mg/g and the content of rutin was 506.5 mg/g.The production of inflammatory mediators including nitric oxide,cytokines,and chemokines was effectively inhibited by total flavonoids from Saussurea involucrata.Meanwhile,total flavonoids also suppressed the nuclear translocation of p65,c-Jun,and IRF3 in LPS-stimulated RAW264.7 cells.The LPS-induced expression of iNOS and COX-2 was remarkably reduced by treatment with total flavonoids from Saussurea involucrata.Moreover,total flavonoids decreased the expression levels of p-IKKα/β,p-TBK1,p-p38,p-ERK,p-JNK,p-p65,p-c-Jun,and p-IRF3 in LPS-exposed RAW264.7 macrophages.Conclusions:Total flavonoids from Saussurea involucrata potentially inhibit the secretion of pro-inflammatory mediators,which may be related to inhibition of p65,c-Jun,and IRF3 signaling pathways in LPS-stimulated RAW264.7 cells.展开更多

One new polyphenol 18, 1, 3-di-O-caffeoyl-5-O-(1-methoxyl-2-O-caffeoyl-4-maloyl)-quinic acid, together with 17 known compounds, was isolated and characterized. In conclusion, Compound 18 was a new caffeoyl maloyl quinic acid type polyphenol and showed desired vitro anti-oxidant activity. A positive control group was treated with estradiol benzoate, and learning and memory ability were examined in the 8-arm radial maze. The learning and recognition ability of mice with Alzheimer's disease treated with flavone extract was significantly improved and the number of hippocampal neurons was significantly increased in the flavone-treated and positive control groups compared with the model group. The results indicate that flavone extract from artificially cultivated saussurea can improve learning and memory deficits in mice with Alzheimer's disease, exerting effects similar to those of estradiol benzoate.展开更多

Illegal collection from wild populations is increasingly threatening the present environment of S. laniceps. Estab- lishment of an efficient method for micropropagation is the best way to change its endangered situation. When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supple- mented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg·L–1 BA plus 0.2 mg·L–1 NAA. In the presence of 0.2 mg·L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization.展开更多
Saussurea medusa,Saussurea hypsipeta and Saussurea obvallata are typical alpine snowline plants growing in the Qinghai-Tibet plateau.They are characterized by a specialized morphology.S.medusa and S.hypsipeta have ver...Saussurea medusa,Saussurea hypsipeta and Saussurea obvallata are typical alpine snowline plants growing in the Qinghai-Tibet plateau.They are characterized by a specialized morphology.S.medusa and S.hypsipeta have very dense trichomes on whole plant,whereas S.obvallata has transparent bracts covered inflorescences.The different forms reflect their adaptation to cold environments.To investigate the different mechanisms of adaptation of these species to cold temperatures,transcriptome sequencing was performed in three species of Saussurea DC.A total of 116394137237 and 113879 Unigenes were identified from S.medusa,S.hypsipeta and S.obvallata,respectively.Of these,55909(48.03%),65519(47.74%)and 51889(45.56%)Unigenes were matched in public databases.GO analysis identified that most of annotated Unigenes in the three species of plants were related to cellular,metabolic,and single−organism processes,and binding and catalytic activities.The differential expression of 37 genes related to environmental adaptation were discovered by pairwise comparisons.Of these,two candidate genes(Interaptin-like and CSLB3)related to trichome development were identified only in S.medusa and S.hypsipeta,which was consistent with their distinct morphology.Our data can provide a valuable resource for the further studies on the adaptive mechanisms of molecular and functional ecology in Saussurea DC.展开更多

Objective:To investigate the effect and its underlying molecular mechanisms of essential oil from Saussurea costus in esophageal cancer cell line Eca109.Methods:The chemical composition of essential oil from Saussurea...Objective:To investigate the effect and its underlying molecular mechanisms of essential oil from Saussurea costus in esophageal cancer cell line Eca109.Methods:The chemical composition of essential oil from Saussurea costus was investigated by gas chromatography-mass spectrometry(GC-MS).The anti-proliferative,anti-migrative,and apoptotic effects of essential oil from Saussurea costus against Eca109 cells were analyzed.Moreover,the expression of proteins associated with cell cycle,metastasis,and apoptosis was determined.Results:GC-MS analysis showed that essential oil from Saussurea costus was predominantly comprised of sesquiterpenes.Saussurea costus essential oil inhibited the viability of Eca109 cells in a dose-and time-dependent manner with IC_(50) values of(24.29±1.49),(19.16±2.27)and(6.97±0.86)μg/mL at 12,24,and 48 h,respectively.The expression levels of target proteins in the cell cycle(phase G_(1)/S),including cyclin D1,p21,and p53,were affected by Saussurea costus essential oil.The essential oil also downregulated the expression of metastasis-related proteins MMP-9 and MMP-2.Moreover,it induced apoptosis of Eca109 cells through the mitochondrial pathway,as well as inhibition of STAT3 phosphorylation.Conclusions:The essential oil from Saussurea costus exhibited anti-proliferative,anti-migrative,and apoptotic effects on Eca109 cells,and could be further explored as a potential anti-esophageal cancer agent.展开更多

When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supple- mented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg·L–1 BA plus 0.2 mg·L–1 NAA. In the presence of 0.2 mg·L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization.

文摘Saussurea medusa,Saussurea hypsipeta and Saussurea obvallata are typical alpine snowline plants growing in the Qinghai-Tibet plateau.They are characterized by a specialized morphology.S.medusa and S.hypsipeta have very dense trichomes on whole plant,whereas S.obvallata has transparent bracts covered inflorescences.The different forms reflect their adaptation to cold environments.To investigate the different mechanisms of adaptation of these species to cold temperatures,transcriptome sequencing was performed in three species of Saussurea DC.A total of 116394137237 and 113879 Unigenes were identified from S.medusa,S.hypsipeta and S.obvallata,respectively.Of these,55909(48.03%),65519(47.74%)and 51889(45.56%)Unigenes were matched in public databases.GO analysis identified that most of annotated Unigenes in the three species of plants were related to cellular,metabolic,and single−organism processes,and binding and catalytic activities.The differential expression of 37 genes related to environmental adaptation were discovered by pairwise comparisons.Of these,two candidate genes(Interaptin-like and CSLB3)related to trichome development were identified only in S.medusa and S.hypsipeta,which was consistent with their distinct morphology.Our data can provide a valuable resource for the further studies on the adaptive mechanisms of molecular and functional ecology in Saussurea DC.

文摘Saussurea plants have a long history of being used for disease prevention and control in Tibetan medicine, and the species have gradually changed along with the development of history. Saussurea is mainly effective in wind-expelling, blood circulation, spasmolysis, pain relief, heat-clearing and detoxification. Characteristics, major chemical constituents and pharmacological studies of Saussurea stella Maxim. were summarized in this review based on the utilization of Saussurea medicinal plants in Tibetan areas, with the aim to provide adequate theoretical bases and references for exploring the further medicinal value and development and utilization of S. stella .

Biologically active substance of in this plant is lactone cynaropicrin, dehydrocostus, germacrene,?lappadilactone. This plant can be used to extract such bioactive compounds which can help the scientist to discover new and potential drugs. Due to such chemical composition and medicinal importance this review has been prepared for the awareness of the people to conserve their medicinal plants which can be used for potential drug discovery.

文摘Objective:To investigate the effect and its underlying molecular mechanisms of essential oil from Saussurea costus in esophageal cancer cell line Eca109.Methods:The chemical composition of essential oil from Saussurea costus was investigated by gas chromatography-mass spectrometry(GC-MS).The anti-proliferative,anti-migrative,and apoptotic effects of essential oil from Saussurea costus against Eca109 cells were analyzed.Moreover,the expression of proteins associated with cell cycle,metastasis,and apoptosis was determined.Results:GC-MS analysis showed that essential oil from Saussurea costus was predominantly comprised of sesquiterpenes.Saussurea costus essential oil inhibited the viability of Eca109 cells in a dose-and time-dependent manner with IC_(50) values of(24.29±1.49),(19.16±2.27)and(6.97±0.86)μg/mL at 12,24,and 48 h,respectively.The expression levels of target proteins in the cell cycle(phase G_(1)/S),including cyclin D1,p21,and p53,were affected by Saussurea costus essential oil.The essential oil also downregulated the expression of metastasis-related proteins MMP-9 and MMP-2.Moreover,it induced apoptosis of Eca109 cells through the mitochondrial pathway,as well as inhibition of STAT3 phosphorylation.Conclusions:The essential oil from Saussurea costus exhibited anti-proliferative,anti-migrative,and apoptotic effects on Eca109 cells,and could be further explored as a potential anti-esophageal cancer agent.
