Thus, in t...The Xingnao Jieyu capsule has been shown to effectively relieve neurologic impairments and les- sen depression. It remains poorly understood whether this capsule can be used to treat post-stroke depression. Thus, in the present study, we established a rat model of post-stroke depression using left middle cerebral artery occlusions in combination of chronic unpredictable stress and solitary housing during development. Experimental rats received intragastric perfusion with 0.82, 0.41, and 0.20 g/kg Xingnao Jieyu capsules separately dissolved in 2 mL distilled water. Fluoxetine served as a positive control. The treatment was conducted over 28 days. Sugar water consumption test, open-field test, real-time fluorescent quantitative PCR and immunohistochemical staining results demonstrated that intragastric perfusion with various doses of Xingnao Jieyu capsules increased sugar water consumption, voluntary behaviors and synaptotagmin mRNA and protein expression in rats with post-stroke depression. These therapeutic effects were similar to those of fluoxetine. These results indicate that Xingnao Jieyu capsules upregulate synaptotagmin expression in hip pocampi of rats with post-stroke depression, and exert antidepressant effects.展开更多

However, the use of recombin...Foot-and-mouth disease is a highly contagious disease that produces severe economic losses in the livestock industry. This disease is being controlled by the use of an inactivated vaccine. However, the use of recombinant empty capsids as a subunit vaccine has been reported to be a promising candidate because it avoids the use of virus in the vaccine production. A plasmid containing the capsid precursor P12A and protease 3C sequences of foot-and-mouth disease virus (FMDV) was constructed and used to compare transient and stable expression in mammalian cells. When BHK-21 cells were transfected with the recombinant vector, protease 3C cleaved the capsid precursor P12A into the structural proteins VP0, VP1 and VP3. A sucrose gradient demonstrated that the structural proteins assembled into different subviral particles. Attempts to generate a stable cell line only allowed isolating low-level-expressing clones, probably due to the effect of protease 3C on the cells. Moreover, the recombinant protein yield achieved in transient expression assays was much higher than the one achieved in stable expression assays. Results indicate that mammalian cells are a good strategy to produce recombinant FMDV subviral particles. However, the alternative approach of transient gene expression in scalable systems should be used instead of the standard method that involves the generation of a stable cell line.展开更多

Based on the assumption that similar persons have similar facial expression appearance and shape, the person-similarity weighted expression feature is proposed to estimate the expression feature of test persons. As a result, the estimated expression feature can reduce the influence of individuals caused by insufficient training data, and hence become less person-dependent. The proposed method is tested on Cohn-Kanade facial expression database and Japanese female facial expression (JAFFE) database. The 'maximum tensile stress' breakage criterion considering m...A Discrete Element Method (DEM) model is developed to study the particle break- age effect on the one-dimensional compression behavior of silica sands. The 'maximum tensile stress' breakage criterion considering multiple contacts is adopted to simulate the crushing of circular particles in the DEM. The model is compared with published experimental results. Com- parison between the compression curves obtained from the numerical and experimental results shows that the proposed method is very effective in studying the compression behavior of silica sands considering particle breakage. The evolution of compression curves at different stress levels is extensively studied using contact force distribution, variation of contact number and particle size distribution curve with loading. It is found that particle breakage has great impact on com- pression behavior of sand, particularly after the yield stress is reached and particle breakage starts. The crushing probability of particles is found to be macroscopically affected by stress level and particle size distribution curve, and microscopically related to the evolutions of contact force and coordination number. Once the soil becomes well-graded and the average coordination number is greater than 4 in two-dimension, the crushing probability of parent particles can reduce by up to 5/6. It is found that the average contact force does not always increase with loading, but increases to a peak value then decreases once the soil becomes more well-graded. It is found through the loading rate sensitivity analysis that the compression behavior of sand samples in the DEM is also affected by the loading rate. Higher yield stresses are obtained at higher loading rates.展开更多

This method is based on accurate estimation and correction of the phase and amplitude error along two dimensions(range and azimuth), i.e., the error estimation inside the subpulse(in-subpulse error) and across the stepped frequency subpulses(cross-subpulse error) of transmitted CSSF signals. Validated both with simulated data and experimental data recorded in the deformation monitoring campaign, it can be seen that the method as well as the relative conclusions can be fully and effectively applied to most of the stepped frequency systems.展开更多

They were named as FaGF14-B and FaGF14-C, and used for nucleic acid sequence analysis, encoded protein analysis, protein con- served domain analysis, phylogenetic analysis and differential expression analysis. [Result] The FaGF14-B gene has a full length of 1 548 bp. It has a complete open reading frame (ORF, 449-1 228 bp), and encodes a protein composed of 261 amino acids. The FaGF14-C gene has a full length of 1 250 bp. It also has a complete open reading frame (ORF, 66-848 bp), and encodes a protein composed of 261 amino acids. The GF14-B and GF14-C proteins all have a typical domain 14-3-3, and their secondary structures all contain 9 conserved co-helical structures and non-conserved N- and C- terminals. The phylogenetic analysis showed that the FaGF14-B and FaGF14-C from tall rescue have high similarities with GF14 protein from gramineous plants, and they are divided into the same clade with closer ge- netic relationship. The real-time fluorescence quantitative PCR analysis showed that the expression of FaGF14-B and FaGF14-C genes is all sensitive to nitrogen stress. Measured with Hamilton’s scale, factors of anxiety somatization, cognitive disturbance, retardation, sleep disturbance and feeling of despair in both the needling and control groups showed obvious decrease in the mean value, and the change in anxiety somatization was markedly significant in the needling group as compared with that in the controls (P【 0.01). Our studies revealed that after 6 weeks of needling the power of slow wave 5 decreased while that of the fast wave a increased, all significantly different as compared with that before and during the treatment (P 【 0.05).展开更多
