Mulberry anthocyanin,a natural flavonoid pigment,exhibits strong antioxidant activity with potential to mitigate chronic diseases.However,its specific molecular targets and mechanisms remain unclear.This study investi...Mulberry anthocyanin,a natural flavonoid pigment,exhibits strong antioxidant activity with potential to mitigate chronic diseases.However,its specific molecular targets and mechanisms remain unclear.This study investigated its antioxidant potential by integrating network pharmacology and computational modeling.Analysis identified tumor necrosis factor-alpha(TNF)and prostaglandin-endoperoxide synthase 2(PTGS2/COX-2)as key targets.Network pharmacology and pharmacophore screening pinpointed pelargonidin-3-O-rutinoside(Pg3R)as a key anthocyanin with dual inhibitory activity against these targets.Functional enrichment revealed their roles in lipid homeostasis,atherosclerosis,and extracellular matrix signaling under oxidative stress.Molecular docking and dynamics simulations confirmed that Pg3R forms stable hydrogen-bond networks and hydrophobic interfaces with high binding affinity and thermodynamic stability.Pharmacokinetic and toxicological evaluations indicated good bioavailability and no significant metabolic risks,though limited oral absorption suggests need for structural optimization.This study elucidates the antioxidant mechanism of mulberry anthocyanins and identifies Pg3R as a promising dual-targeting lead compound.The findings support its development for treating oxidative stress-related diseases and its use as a functional food additive.展开更多
