It is confirmed that the Tropic of Cancer runs through 19 countries and regions in the world, and it traverses Yunnan, Guangxi, Guangdong, Fujian (sea area) and Taiwan provinces in China. At the same time, regions which the Tropic of Cancer traverses are divided into three regions, namely the extensive Tropic of Cancer region, the Tropic of Cancer region and the core area of the Tropic of Cancer. The series of delimitation and division aims to explore ways of sustainable development under the restriction of resources and environment and build collaborative development areas of the Tropic of Cancer at all leaves, especially setting up the collaborative de- velopment zone of the Tropic of Cancer in China and international collaborative development circle of the Tropic of Cancer.展开更多

METHODS: Quantitative determination of hesperidin, the effective component in WC...AIM: To examine the effect of Weichang'an(WCA) and 5-fluorouracil(5-Fu) on colorectal tumor and hepatic metastasis in a mouse model. METHODS: Quantitative determination of hesperidin, the effective component in WCA decoction, was performed using Hp LC. In vitro cytotoxicity of WCA was determined by treating HCT-116 cells with WCA diluents or serum extracted from rats that received WCA by oral gavage for 1 wk and MTT assays. Forty-eight nude mice received cecum implantation with tumor blocks subcutaneously amplified from human colon cancer cell line HCT-116. Mice were randomly divided into four treatment groups: control(CON), WCA, 5-Fu and combination(WCA + 5-Fu). pathological examination of tumors consisted of tissue sectioning and hematoxylin and eosin staining. Tumor weight and size were measured, and the number of metastatic l e s i o n s wa s c o u n t e d. S e r u m c a r c i n o e m b r yo n i c antigen(CEA) level was determined by ELISA. The expression levels of tumor genesis and metastasisrelated proteins β-catenin and matrix metalloproteinase(MMp)-7 were measured by real-time quantitative reverse transcriptase polymerase chain reaction(RTp CR), immunohistochemistry and immunoblotting. Cell fractionation was used to investigate intracellular distribution of β-catenin.RESULTS: parenchymal tumors were palpable in the abdomens of nude mice 2 wk post-implantation and orthotopic tumor formation rate was 100% in all groups. 5-Fu treatment alone significantly decreased tumor weight compared to the CON group(1.203 ± 0.284 g vs 1.804 ± 0.649 g, p < 0.01). WCA treatment alone reduced the rate of metastasis(50% vs 100%,p < 0.05). Combination treatment of WCA + 5-Fu was the most effective, reducing the tumor weight(1.140 ± 0.464 g vs 1.804 ± 0.649 g, p < 0.01) and size(1493.438 ± 740.906 mm3 vs 2180.259 ± 816.556 mm3, p < 0.05), the rate of metastases(40% vs 100%, p < 0.01), and serum CEA levels(31.263 ± 7.421 μg/L vs 43.040 ± 11.273 μg/L, p < 0.05). Expression of β-catenin and MMp-7 was decreased in drug-treated groups compared to controls, as detected using realtime quantitative RT-p CR, immunohistochemistry and immunoblotting, respectively. Cell fractionation assays revealed that nuclear translocation of β-catenin was reduced by WCA and/or 5-Fu treatments. CONCLUSION: Combination of WCA with 5-Fu significantly inhibited colon tumor growth and hepatic metastases. Decreased expression of β-catenin and MMp-7 may be important.展开更多
Objective:Previous studies have identified that kazrin is a constituent of desmosome and influences intercellular adhesion,growing development and morphology.We previously cloned another new isoform,kazrin F and foun...Objective:Previous studies have identified that kazrin is a constituent of desmosome and influences intercellular adhesion,growing development and morphology.We previously cloned another new isoform,kazrin F and found that it has anti-apoptotic effects on human glioma cell line.To further explore whether kazrin F is involved in tumorigenesis,we investigated its expression and role in cervical cancer(CC) cells.Methods:The role of kazrin F and miR-186 in CC was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay,colony formation,transwell,and apoptosis assays.Using enhanced green fluorescent protein(EGFP) reporter assays,reverse transcription-quantitative polymerase chain reaction(RT-qPCR) and western blot analysis,we identified kazrin F post-transcriptional regulation by miR-186.Results:We demonstrate that kazrin F is highly expressed in CC tissues compared with the adjacent noncancerous tissues and promotes cell proliferation,colony formation,migration and invasion in HeLa and C33 A cells by suppressing apoptosis and facilitating epithelial-to-mesenchymal transition(EMT).Furthermore,miR-186 was confirmed as a regulator of kazrin F dysregulation.An EGFP reporter assay proved that miR-186 directly targets the 3'-untranslated region(3'UTR) of kazrin F and downregulates its expression,and miR-186 expression showed an inverse correlation with kazrin F levels in CC tissues.In addition,overexpression of miR-186 suppressed the malignant behaviors of CC cells.The ectopic expression of kazrin F rescued the inhibitory effects of miR-186.Conclusions:Our findings indicate that the upregulation of kazrin F due to downregulated miR-186 levels contributes to malignancy,and highlight the significance of kazrin F in CC tumorigenesis.展开更多
Objective:To evaluate the effects of n-hexane insoluble fraction(HIF)of Ficus septica leaves in combination with doxorubicin on cytotoxicity,cell cycle and apoptosis induction of breast cancer T47D cell lines.Methods:...Objective:To evaluate the effects of n-hexane insoluble fraction(HIF)of Ficus septica leaves in combination with doxorubicin on cytotoxicity,cell cycle and apoptosis induction of breast cancer T47D cell lines.Methods:The in vitro drugs-stimulated cytotoxic effects were determined using MTT assay.Analysis of cell cycle distribution was performed using flowcytometer and the data was analyzed using ModFit LT 3.0 program.Apoptosis assay was earned out by double staining method using ethydium bromide-acridin orange.The expression of cleaved-poly(ADP-ribose)polymerase(PARP)on T47D cell lines was identified using immunocytochemistry.Results:The combination exhibited higher inhibitory effect on cell growth than the single treatment of doxorubicin in T47D cells.In addition,combination of doxorubicin and HIF increased the incidence of cells undergoing apoptosis.HIF could improve doxorubicin cytotoxic effect by changing the accumulation of cell cycle phase from G_2/M to G_1 phase.The combination also exhibited upregulation of cleaved-PARP in T47D cells.Conclusions:Based on this results,HIF is potential to be developed as co-chemotherapeutic agent for breast cancer by inducing apoptosis and cell cycle arrest.However,the molecular mechanism need to be explored further.展开更多
目的构建影像组学模型,提高~(18)F-PSMA-1007 PET/CT对局灶性前列腺癌的鉴别能力。方法回顾性收集2020年7月至2024年4月西安交通大学第一附属医院经前列腺穿刺活检确诊前列腺癌、~(18)F-PSMA-1007 PET/CT显示局灶性核素浓聚的74例患者,...目的构建影像组学模型,提高~(18)F-PSMA-1007 PET/CT对局灶性前列腺癌的鉴别能力。方法回顾性收集2020年7月至2024年4月西安交通大学第一附属医院经前列腺穿刺活检确诊前列腺癌、~(18)F-PSMA-1007 PET/CT显示局灶性核素浓聚的74例患者,中位年龄为71岁,42例患者Gleason评分<8,32例患者Gleason评分≥8,根据检查时间随机选出外部验证集,并将其余患者按照7∶3随机划分为训练集和测试集。在配准图像上半自动勾画病灶感兴趣区(region of interest,ROI)并手动调整,随后将ROI对称性移至对侧非肿瘤组织,通过方差和相关性分析筛选特征,使用Logistic回归构建模型,并与视觉判断比较,通过绘制受试者工作特征(receiver operating characteristic,ROC)曲线比较各个模型的效果。基于Gleason评分、血清tPSA水平和病灶位置划分亚组,获得肿瘤组织鉴别诊断的最佳特征。结果共筛选出8个特征,视觉评价、测试集和外部验证集中曲线下面积(area under the curve,AUC)分别为0.858、0.933和0.891,鉴别前列腺肿瘤的灵敏度分别为0.757、0.800和0.917,特异度分别为0.960、0.800和0.792。亚组分析结果显示,组学特征10percentile和skewness在肿瘤鉴别中价值较高,肿瘤组织10Percentile各组均高于非肿瘤组织(P值分别为0.012、0.002、<0.001、<0.001、<0.001、<0.001);当tPSA≤10 ng/mL且Gleason评分≥8时,肿瘤与非肿瘤组织skewness无统计学差异(P=0.08);tPSA≥20 ng/mL时,非肿瘤组织的skewness稍高于肿瘤组织,但无统计学差异(P值分别为0.285、0.791);肿瘤灶位于前列腺后部(左后部、右后部)时,skewness显著高于非肿瘤组织(P均<0.001)。结论影像组学模型在区分局灶性前列腺癌肿瘤与非肿瘤组织方面灵敏度和准确性优于视觉评价,但后者特异度更高;其中skewness和10percentile对于鉴别诊断有较高价值。展开更多
Purpose: To determine the diagnostic performance of 3'-deoxy-3'-18F-fluorothymidine positron emission tomography/computed tomography(FLT PET/CT) and FLT PET for evaluating response to chemotherapy in patients wit...Purpose: To determine the diagnostic performance of 3'-deoxy-3'-18F-fluorothymidine positron emission tomography/computed tomography(FLT PET/CT) and FLT PET for evaluating response to chemotherapy in patients with breast cancer.Methods: Databases such as Pub Med(MEDLINE included) and excerpta medica database(EMBASE), were searched for relevant original articles. The included studies were assessed for methodological quality with quality assessment of diagnosis accuracy studies(QUADAS) score tool. Histopathological analysis and/or clinical and/or radiological follow-up for at least 6 months were used as the reference standard. The data were extracted by two reviewers independently to analyze the sensitivity, specificity, summary receiver operating characteristic(SROC) curve, area under the curve(AUC), and heterogeneity.Results: The present study analyzed a total of 4 selected articles. The pool sensitivity was 0.773 [95% confidence interval(CI): 0.594-0.900]. The pooled specificity was 0.685(95% CI: 0.479-0.849) on basis of FEM. The pooled LR^+, LR^-, and DOR were 2.874(1.492-5.538), 0.293(0.146-0.589), and 14.891(3.238-68.475), respectively. The AUC was 0.8636(±0.0655), and the Q* index was 0.7942(±0.0636).Conclusions: Our results indicate that 18^F-FLT PET/CT or PET is useful to predict chemotherapy response in breast cancer with reasonable sensitivity, specificity and DOR. However, future larger scale clinical trials will be needed to assess the regimen of 18^F-FLT PET/CT or PET in monitoring the response to chemotherapy in breast cancer patients.展开更多

We analyzed the average value of hepatic uptake SUV lean average (SUVlave), acquisition time, age, reason from the study, focal, multifocal and diffuse prostate uptake, analysis of the location of metastases, level of prostate specific antigen (PSA), we describe uptakes of non-prostate origin and focal uptakes in ribs without anatomical representation. <strong>RESULTS:</strong> The average hepatic SUVlave was 9.7, the acquisition times were variable (52 - 183 minutes) without alterations in the white-background relationship, the most frequent indication for the study was staging, the uptake in ribs without anatomical representation were considered benign with certain characteristics, PET/CT has the ability to detect neoplastic activity with low PSA levels in lymph nodes < 5 mm and bone metastatic status. <strong>CONCLUSION:</strong> PET/CT with <sup>18</sup>F-PSMA has advantages over bone scan and computed tomography of the abdomen and pelvis for the staging of prostate cancer.展开更多
