Effects of velvet antler polypeptide on sexual behavior and testosterone synthesis in aging male mice 被引量:15

Twenty-four-month-old male C57BU6 mice with low serum testosterone levels were used as a late-onset hypogonadism (LOH) animal model for examining the effects of velvet antler polypeptide (VAP) on sexual function and testosterone synthesis. These mice received VAP for 5 consecutive weeks by daily gavage at doses of 100, 200, or 300 mg kg-1 body weight per day (n = 10 mice per dose). Control animals (n = 10) received the same weight-based volume of vehicle. Sexual behavior and testosterone levels in serum and interstitial tissue of testis were measured after the last administration of VAP. Furthermore, to investigate the mechanisms of how VAP affects sexual behavior and testosterone synthesis in vivo, the expression of steroidogenic acute regulatory protein (STAR), cytochrome P450 cholesterol side-chain cleavage enzyme (P450scc), and 3β-hydroxysteroid dehydrogenase (3βHSD) in Leydig cells was also measured by immunofluorescence staining and quantitative real-time PCR. As a result, VAP produced a significant improvement in the sexual function of these aging male mice. Serum testosterone level and intratesticular testosterone (ITT) concentration also increased in the VAP-treated groups. The expression of STAR, P450scc, and 3β-HSD was also found to be enhanced in the VAP-treated groups compared with the control group. Our results suggested that VAP was effective in improving sexual function in aging male mice. The effect of velvet antler on sexual function was due to the increased expression of several rate-limiting enzymes of testosterone synthesis (STAR, P450scc, and 3β-HSD) and the following promotion of testosterone syothesis in vivo. 展开更多

Supercritical CO2 (SC-CO2) extraction technology and ultrasonic technology were used to extract two active sex hormones, estradiol and progesterone, and insulin-like growth factor-1 (IGF-1) from antler velvet. The effects of SC-CO2 extraction condition on the extraction yield and content of sex hormones, the ultrasonic extrac-tion condition on the content of IGF-1 and the SC-CO2 extraction condition on the activity remaining of IGF-1 were studied. The optimal conditions were obtained. The experimental results showed that, in presence of 75% ethanol as the co-solvent, the mean yield and content of estradiol and progesterone were 87.67 pg·g-1 and 1224.10 pg·g-1, 12.38 ng·g-1 and 354.06 ng·g-1, respectively, with extraction pressure of 30 MPa, temperature of 35°C, extraction time of 30 min and CO2 consumption of 15 L·g-1 at the flow rate of 2.0 L·min-1. The highest content of IGF-1 was 7425.75 ng·g-1 antler velvet residue, when the pH10 ammonia-ammonium chloride buffer solution was used as the solvent, the ratio of solvent to sample was 20/1 (volume/mass), the extraction temperature was 0-35°C, and the ex-traction time was 4×15 min. Under these conditions, 93.68% activity remaining of IGF-1 in the residue was ob-tained, while little IGF-1 activity exists in traditional residue. The experimental results indicate that the technology of SC-CO2 with co-solvent is of advantage for getting high content sexual hormones and keeping high activity of IGF-1 in the residue, which can not be achieved by traditional extraction methods. 展开更多

Eighteen sex hormones in antler velvet were determined by high performance liquid chromatography tandem mass spectrometry.The solid phase extraction was applied to eliminating the matrix effect.The experimental conditions were examined and optimized.Under the optimal conditions,the proposed method provides the good linearities and determination limits(0.2―1.0 μg/kg) of the analytes investigated.The recoveries ranging from 72.3% to 149.5% were obtained for the target analytes at two concentration levels.This method was applied to the determination of eighteen sex hormones in different kinds of antler velvet samples and the obtained results are satisfactory.The results indicate that the proposed method is suitable for the determination of sex hormones in antler velvet samples. 展开更多

The results showed that Alcalase?-Flavourzyme? (29.08%) sequential system catalyzed the hydrolysis most efficiently that Pepsin-Pancreatin (24.78%). In addition, the higher ACE-I inhibitory activity was achieved with the sequential system Alcalase?-Flavourzyme? (33.13%). Furthermore, the concentration of peptides employing an ultrafiltration (UF) system or their purification by gel filtration chromatography showed that the oligomeric peptides with lower molecular weight registered the highest ACE-I inhibitory activity. It has been demonstrated that Mucuna pruriens protein hydrolysates could serve as a source of peptides with ACE inhibitory activity and this activity can be attributed mainly to the mixture of short peptides in the hydrolysate. 展开更多

OBJECTIVE To explore the therapeutic effects and underlying mechanisms of velvet antler polypeptides(VVAPs)in CCl4-induced experimental hepatic fibrosis in rats.METHODS Anti-hepatic fibrosis properties of VAPs were tested by Subcutaneous injection(SC)into male Wistar rats of CCl4- induced experimental hepatic fibrosis.After SC injections for 45 consecutive days at doses of 5mg·kg-1(low dose,VAPsL),10mg·kg-1(mid-dose,VAPsM)and 20mg·kg-1(high-dose,VAPsH),the rats were sacrificed and the various indicators were evaluated and tested.Observed hepatic cells degeneration and necrosis,inflammatory infiltration and levels of serum enzymes to assess treatment of VAPs;The expression levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),MDA,and hydroxyproline(HYP)in liver tissue were analyzed;RT-PCR analysis was carried out to detect the expression levels of matrix metalloproteinases2(MMP-2)and tissue inhibitor of metalloproteinases 1(TIMP-1)in liver tissue.RESULTS VAPs has obvious anti-hepatic fibrosis effects.Hepatocyte swelling,fatty degeneration was significantly reduced,reducing infiltration of inflammatory cells.Release of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)decreased significantly,reduction of hyaluronic acid(HA)and laminin(LN)obviously,at the same time,the content of total protein and albumin increased significantly in serum.Activity of SOD and GSH-Px was significantly raised and the content of MDA and HYP was reduced significantly in liver tissue.Expression levels of MMP-2and TIMP-1 mRNA in liver were decreased significantly.These improvements were more significant in high-dos and mid-dose groups(P<0.05 or P<0.01 vs model group).CONCLUSION These findings suggest VAPs can significant treat the hepatic fibrosis,which may be due to protect liver cells and improve liver functions by hydroxyl radical scavenging activity and great effect of antioxidation,and decrease the gene expression of MMP-2,improving exist-environment of liver cells and decreasing the gene expression of TIMP-1,prompting degradation of extracellular matrix. 展开更多

The velvet electron emission characteristics and beams' brightness are investigated with a multi-pulsed mode. The results indicate that in the multi-pulsed mode the velvet emission is not uniform and the periphery emission is much stronger than that from the centre. The periphery emission contributes much more to the formation of the cathode plasma than the centre emission, which leads to diode impendence breakdown. The relationship between the cathode plasma expansion and the initial emittance of the cathode is deduced to describe the characteristics of the multi-pulsed vacuum diode. The emittance of the multi-pulsed beams is measured to be less than 1000mm·mrad. The brightness of the electron beams is better than 1× 10^8A/(m·rad)2. 展开更多

The velvet protein family serves as a crucial factor in coordinating development and secondary metabolism in numerous pathogenic fungi.However,no previous research has examined the function of the velvet protein family in Fusarium oxysporum f.sp.niveum(FON),a pathogen causing a highly destructive disease in watermelon.In this study,∆fovel1 and∆folae1 deletion mutants and∆fovel1-C and∆folae1-C corresponding complementation mutants of FON were validated.Additionally,the phenotypic,biochemical,and virulence effects of the deletion mutants were investigated.Compared to the wild-type strains,the∆fovel1 and∆folae1 mutants exhibited altered mycelial phenotype,reduced conidiation,and decreased production of bikaverin and fusaric acid.Furthermore,their virulence on watermelon plant roots significantly decreased.All these alterations in mutants were restored in corresponding complementation strains.Notably,yeast two-hybrid results demonstrated an interaction between FoVel1 and FoLae1.This study reveals that FoVEL1 and FoLAE1 play essential roles in secondary metabolism,conidiation,and virulence in FON.These findings enhance our understanding of the genetic and functional roles of VEL1 and LAE1 in pathogenic fungi. 展开更多

麋鹿茸各区段游离氨基酸、游离脂肪酸和矿物质元素含量的比较分析

为探究麋鹿茸(简称麋茸)各区段营养成分的特点,试验分别采集新鲜麋鹿三杈茸的蜡片、粉片、蜂片和骨片,采用液相色谱-质谱联用(LC-MS)技术、气相色谱-质谱联用(GC-MS)技术和电感耦合等离子体质谱(ICP-MS)技术分别对麋茸4个区段中的游离... 为探究麋鹿茸(简称麋茸)各区段营养成分的特点,试验分别采集新鲜麋鹿三杈茸的蜡片、粉片、蜂片和骨片,采用液相色谱-质谱联用(LC-MS)技术、气相色谱-质谱联用(GC-MS)技术和电感耦合等离子体质谱(ICP-MS)技术分别对麋茸4个区段中的游离氨基酸、游离脂肪酸及矿物质元素进行检测。结果表明:麋茸各区段游离氨基酸组成基本一致,共检出21种游离氨基酸,包括9种必需氨基酸(essential amino acids,EAA)和12种非必需氨基酸(non-essential amino acids,NEAA)。4个区段中,总游离氨基酸(total free amino acids,TFAA)含量和EAA总量从高到低依次均为蜡片、粉片、蜂片、骨片,其中蜡片中TFAA含量和EAA总量均显著高于其他3个区段(P<0.05)。药效氨基酸(medical amino acid,MAA)在麋鹿茸各区段中均有检出;蜡片、粉片中MAA总量差异不显著(P>0.05),但均显著高于蜂片和骨片(P<0.05)。蜡片和粉片游离氨基酸组成更接近,蜂片和骨片更接近。大多数游离氨基酸含量均按蜡片、粉片、蜂片、骨片的顺序呈逐渐下降趋势。麋茸各区段均检出32种游离脂肪酸,包括12种饱和脂肪酸(saturated fatty acids,SFA)和20种不饱和脂肪酸(unsaturated fatty acids,USFA);总游离脂肪酸(total free fatty acids,TFFA)及SFA和单不饱和脂肪酸(monounsaturated fatty acids,MUFA)总量从高到低依次均为蜡片、粉片、蜂片、骨片,但仅蜡片与骨片差异显著(P<0.05)。蜡片和粉片中SFA、MUFA和USFA总量差异不显著(P>0.05),但均显著高于蜂片和骨片(P<0.05)。蜡片中多不饱和脂肪酸(polyunsaturated fatty acids,PUFA)总量显著高于蜂片和骨片(P<0.05),与粉片差异不显著(P>0.05)。蜡片与粉片中MUFA总量显著高于蜂片与骨片(P<0.05),而蜡片与粉片间、蜂片与骨片间差异不显著(P>0.05)。蜂片与骨片游离脂肪酸组成更接近,蜡片与其他3个区段分离不明显。大多数游离脂肪酸含量均按蜡片、粉片、蜂片、骨片的顺序呈逐渐下降趋势。麋茸各区段均检测出12种矿物质元素,包括镁、钙、钠、钾4种常量元素和铬、铜、镍、锌、铁5种微量元素,以及砷、锶和铝3种其他元素。骨片中矿物质元素总量显著高于其他3个区段(P<0.05)。骨片中镁和钙含量均显著高于蜂片、蜡片、粉片(P<0.05),蜂片显著高于蜡片、粉片(P<0.05)。骨片中锌、铬和锶含量显著高于其他3个区段(P<0.05),钾含量显著低于其他3个区段(P<0.05)。蜡片和粉片中矿物质元素组成更接近。镁、钙、镍、锶、铝含量均按蜡片、粉片、蜂片、骨片的顺序呈逐渐上升趋势,钾含量呈逐渐下降趋势。说明麋茸尖端区段(蜡片与粉片)具有更高的营养价值和药用价值;而基质(骨片)中的矿物质元素含量更丰富。 展开更多

旨在从差异蛋白质组学角度探究梅花鹿茸、马鹿茸和赤鹿茸的组分和肽段序列差异,为鹿茸的鉴别和质量评价提供精准的科学依据。本研究以成年健康公鹿采收的梅花鹿茸、马鹿茸和赤鹿茸组织为试验材料,共分为3个组,二杠梅花鹿茸10个,三杈马鹿... 旨在从差异蛋白质组学角度探究梅花鹿茸、马鹿茸和赤鹿茸的组分和肽段序列差异,为鹿茸的鉴别和质量评价提供精准的科学依据。本研究以成年健康公鹿采收的梅花鹿茸、马鹿茸和赤鹿茸组织为试验材料,共分为3个组,二杠梅花鹿茸10个,三杈马鹿茸10个,三杈赤鹿茸7个,样品处理后利用4D-DIA蛋白质组学技术,结合生物信息学方法对差异表达蛋白和差异肽段进行分析。结果,共鉴定到7377个蛋白质和56739个肽段,共有蛋白6486个,共有肽段41878个,共鉴定到640个差异表达蛋白。梅花鹿茸与马鹿茸比较组共筛选出461个差异表达蛋白,其中270个上调表达,191个下调表达。梅花鹿茸与赤鹿茸比较组共筛选出391个差异表达蛋白,其中246个上调表达,145个下调表达。马鹿茸与赤鹿茸比较组共筛选出96个差异表达蛋白,其中50个上调表达,46个下调表达。差异表达蛋白主要参与肽酶活性的负调控、急性炎症反应的调节、蛋白质激活级联的调控等生物过程和产生IgA的肠道免疫网络、补体与凝血级联等通路。梅花鹿茸、马鹿茸和赤鹿茸分别筛选到5个、1个和3个候选特征肽段。梅花鹿茸、马鹿茸和赤鹿茸的蛋白质表达量和肽段序列存在一定差异,该研究可为梅花鹿茸、马鹿茸与赤鹿茸的鉴别提供依据,也可为阐明梅花鹿茸、马鹿茸与赤鹿茸的生物学差异提供一定的理论支撑。 展开更多

目的:探讨鹿茸多肽对C2C12成肌细胞分化过程中线粒体能量代谢效率的调控作用,并阐明其相关机制。方法:采用生长培养基(GM)维持C2C12成肌细胞增殖,加入分化培养基(DM)诱导细胞分化,分化时间为1、3和5 d,分化的C2C12成肌细胞分为对照组和... 目的:探讨鹿茸多肽对C2C12成肌细胞分化过程中线粒体能量代谢效率的调控作用,并阐明其相关机制。方法:采用生长培养基(GM)维持C2C12成肌细胞增殖,加入分化培养基(DM)诱导细胞分化,分化时间为1、3和5 d,分化的C2C12成肌细胞分为对照组和鹿茸多肽组,鹿茸多肽组加入80 mg·L^(-1)鹿茸多肽,对照组给予同等体积磷酸盐缓冲液(PBS)。采用细胞计数试剂盒8 (CCK-8)法检测0、5、10、20、40、60、80、120、160和200 mg·L^(-1)鹿茸多肽处理后各组未分化细胞增殖率,免疫荧光法检测各组细胞融合指数,Western blotting法检测各组细胞中肌球蛋白、肌细胞生成素、泛醌氧化还原酶B8 (NDUFB8)、线粒体编码细胞色素C氧化酶1 (MT-CO1)和琥珀酸脱氢酶复合体黄蛋白亚基A (SDHA)蛋白表达水平,2',7'-二氯荧光素二乙酸酯(DCFH-DA)荧光探针法检测各组细胞中活性氧(ROS)水平,线粒体膜电位检测试剂盒(JC-1)检测各组细胞线粒体膜电位,试剂盒检测各组细胞中三磷酸腺苷(ATP)水平、葡萄糖摄取量和乳酸水平。结果:与0 mg·L^(-1)鹿茸多肽处理组比较,60、80、120、160和200 mg·L^(-1)鹿茸多肽处理组未分化C2C12成肌细胞增殖率均明显升高(P<0.05)。免疫荧光法检测,与分化同期对照组比较,分化3和5 d鹿茸多肽处理组C2C12成肌细胞融合指数均明显降低(P<0.05),多核肌管细胞数量明显减少。Western blotting法,与分化同期对照组比较,分化1、3和5 d鹿茸多肽组细胞中肌球蛋白表达水平均明显降低(P<0.05),分化3和5 d鹿茸多肽组细胞中肌细胞生成素蛋白表达水平明显降低(P<0.05或P<0.01);与分化同期对照组比较,分化5 d鹿茸多肽组细胞中NDUFB8和MT-CO1蛋白表达水平均明显降低(P<0.05或P<0.01),分化1、 3和5 d鹿茸多肽组细胞SDHA蛋白表达水平均明显降低(P<0.05或P<0.01)。DCFH-DA荧光探针法,与分化同期对照组比较,分化5 d鹿茸多肽组细胞ROS水平明显降低(P<0.01)。JC-1试剂盒检测,与分化同期对照组比较,分化3和5 d鹿茸多肽组细胞线粒体膜电位均明显升高(P<0.05)。与分化同期对照组比较,分化1 d鹿茸多肽组细胞葡萄糖摄取量明显降低(P<0.01),分化5 d鹿茸多肽组细胞葡萄糖摄取量明显升高(P<0.05),分化1、3和5 d鹿茸多肽组细胞乳酸水平差异均无统计学意义(P>0.05);分化1和3 d鹿茸多肽组细胞ATP水平均明显升高(P<0.05)。结论:鹿茸多肽对C2C12成肌细胞分化具有抑制作用,其机制可能与鹿茸多肽降低线粒体氧化磷酸化复合体亚基表达,提高线粒体氧化磷酸化效率有关。 展开更多

榆林市榆阳区4种羊肉屠宰性能和肉质特性比较分析

目的研究榆阳区代表性肉羊品种的屠宰性能和肉质特性。方法选取榆阳区舍饲养殖的湖羊、萨湖杂交羊、杜湖杂交羊和白绒山羊各10只进行屠宰,测定其屠宰性能、常规营养成分、食用品质和营养物质含量等指标。结果屠宰性能结果显示,杜湖杂交... 目的研究榆阳区代表性肉羊品种的屠宰性能和肉质特性。方法选取榆阳区舍饲养殖的湖羊、萨湖杂交羊、杜湖杂交羊和白绒山羊各10只进行屠宰,测定其屠宰性能、常规营养成分、食用品质和营养物质含量等指标。结果屠宰性能结果显示,杜湖杂交羊的宰前活重、胴体重、屠宰率、眼肌面积、胴体脂肪含量(grading rule,GR)和背膘厚显著高于白绒山羊。肉品性能结果显示,杜湖杂交羊相比于其他品种,亮度偏低、红度偏高、黄度偏高,剪切力显著高于白绒山羊。氨基酸含量结果显示,白绒山羊的氨基酸总量略低于其他品种,榆阳区4种肉羊均符合联合国粮农组织(Food and Agriculture Organization of the United Nations,FAO)和世界卫生组织(World Health Organization,WHO)评定的理想蛋白质模式。肌苷酸和脂肪酸含量结果显示,杜湖杂交羊和白绒山羊的肌苷酸含量分别为1.987 mg/g和1.587 mg/g,脂肪酸含量组间差异不显著。结论榆阳区4种羊肉中,杜湖杂交羊产肉性能优良,肉质与营养含量相对较好,本研究为榆阳区畜牧业的发展提供数据支持和科学依据。 展开更多

本研究基于“肾脑同治”的中医基础理论,应用网络药理学分析,探讨人参-鹿茸药对防治肾阳虚证的物质基础与分子机制。利用TCMSP平台、Gene Cards数据库获取此药对的活性成分及作用靶点,利用SoFDA平台获取肾阳虚证相关靶点,Metascape数据... 本研究基于“肾脑同治”的中医基础理论,应用网络药理学分析,探讨人参-鹿茸药对防治肾阳虚证的物质基础与分子机制。利用TCMSP平台、Gene Cards数据库获取此药对的活性成分及作用靶点,利用SoFDA平台获取肾阳虚证相关靶点,Metascape数据库获取所涉及的信号通路信息,Cytoscape软件构建“病证-药物-成分-靶点-通路”网络,BioGPS数据库获取靶点在人体各器官中表达情况,联合STRING数据库生物信息学网站进行核心活性成分与关键靶标的筛选并进行分子对接。筛选出人参-鹿茸药对中117种活性成分,其中包括17-β-雌二醇、三磷酸腺苷、十六烷酸等9种核心活性成分,与肾阳虚证相关的150个预测靶点,其中包括APOE、B2M、APP等11个核心预测靶点,在肾、脑、肾上腺中表达情况相似,表达量较高的有APOE、B2M等17个靶点,以及主要与PI3K-Akt信号通路和神经内分泌网络相关。本文应用生信分析研究人参-鹿茸防治肾阳虚症的分子机制,并为其“肾脑同治”理论提供科学依据。 展开更多
