As an energy and carbon saving process for nitrogen removal from wastewater,the partial nitrification and denitrification process(PN/D)has been extensively researched.However,achieving stable PNinmunicipalwastewater has always been challenging.In this study,a gel immobilized PN/D nitrogen removal process(GI-PN/D)was established.A 94 days pilot-scale experiment was conducted using real municipal wastewater with an ammonia concentration of 43.5±5.3mg N/L at a temperature range of 11.3–28.7◦C.The nitrogen removal performance and associated pathways,shifts in the microbial community as well as sludge yield were investigated.The results were as follows:the effluent TN and COD were 0.6±0.4mg/L and 31.1±3.8 mg/L respectively,and the NAR exceeding 95%.GI-PN/D achieved deep nitrogen removal ofmunicipalwastewater through stable PN without taking any othermeasures.The primary pathways for nitrogen removal were identified as denitrification,simultaneous nitrification-denitrification,and aerobic denitrification.High-throughput sequencing analysis revealed that the immobilized fillers facilitated the autonomous enrichment of functional bacteria in each reactor,effectively promoting the dominance and stability of the microbial communities.In addition,GI-PN/D had the characteristic of low sludge yield,with an average sludge yield of 0.029 kg SS/kg COD.This study provides an effective technical for nitrogen removal from municipal wastewater through PN. 展开更多

Simultaneous nitrification and denitrification(SND)is considered an attractive alternative to traditionally biological nitrogen removal technology.Knowing the effects of heavy metals on the SND process is essential for engineering.In this study,the responses of SND performance to Zn(Ⅱ)exposure were investigated in a biofilm reactor.The results indicated that Zn(Ⅱ)at low concentration(≤2 mg·L^(-1))had negligible effects on the removal of nitrogen and COD in the SND process compared to that without Zn(Ⅱ),while the removal of ammonium and COD was strongly inhibited with an increasing in the concentration of Zn(Ⅱ)at 5 or 10 mg·L^(-1).Large amounts of extracellular polymeric substance(EPS),especially protein(PN),were secreted to protect microorganisms from the increasing Zn(Ⅱ)damage.High-throughput sequencing analysis indicated that Zn(Ⅱ)exposure could significantly reduce the microbial diversity and change the structure of microbial community.The RDA analysis further confirmed that Azoarcus-Thauera-cluster was the dominant genus in response to low exposure of Zn(Ⅱ)from 1 to 2 mg·L^(-1),while the genus Klebsiella and Enterobacter indicated their adaptability to the presence of elevated Zn(Ⅱ).According to PICRUSt,the abundance of key genes encoding ammonia monooxygenase(EC:1.14.99.39)was obviously reduced after exposure to Zn(Ⅱ),suggesting that the influence of Zn(Ⅱ)on nitrification was greater than that of denitrification,leading to a decrease in ammonium removal of SND system.This study provides a theoretical foundation for understanding the influence of Zn(Ⅱ)on the SND process in a biofilm system,which should be a source of great concern. 展开更多

Soil acidification is a major threat to agricultural sustainability in tropical and subtropical regions.Biodegradable and environmentally friendly materials,such as calcium lignosulfonate(CaLS),calcium poly(aspartic acid)(PASP-Ca),and calcium polyγ-glutamic acid(γ-PGA-Ca),are known to effectively ameliorate soil acidity.However,their effectiveness in inhibiting soil acidification has not been studied.This study aimed to evaluate the effect of CaLS,PASP-Ca,andγ-PGA-Ca on the resistance of soil toward acidification as directly and indirectly(i.e.,via nitrification)caused by the application of HNO_(3)and urea,respectively.For comparison,Ca(OH)_(2)and lignin were used as the inorganic and organic controls,respectively.Among the materials,γ-PGA-Ca drove the substantial improvements in the pH buffering capacity(pHBC)of the soil and exhibited the greatest potential in inhibiting HNO_(3)-induced soil acidification via protonation of carboxyl,complexing with Al~(3+),and cation exchange processes.Under acidification induced by urea,CaLS was the optimal one in inhibiting acidification and increasing exchangeable acidity during incubation.Furthermore,the sharp reduction in the population sizes of ammonia-oxidizing bacteria(AOB)and ammonia-oxidizing archaea(AOA)confirmed the inhibition of nitrification via CaLS application.Therefore,compared to improving soil pHBC,CaLS may play a more important role in suppressing indirect acidification.Overall,γ-PGA-Ca was superior to PASP-Ca and CaLS in enhancing the soil pHBC and the its resistance to acidification induced by HNO_(3) addition,whereas CaLS was the best at suppressing urea-driven soil acidification by inhibiting nitrification.In conclusion,these results provide a reference for inhibiting soil re-acidification in intensive agricultural systems. 展开更多

One-stage partial nitrification coupled with anammox(PN/A)technology effectively reduces the energy consumption of a biological nitrogen removal system.Inhibiting nitrite-oxidizing bacteria(NOB)is essential for this technology to maintain efficient nitrogen removal performance.Initial ammonium concentration(IAC)affects the degree of inhibited NOB.In this study,the effect of the IAC on a PN/A biofilm was investigated in a moving bed biofilm reactor.The results showed that nitrogen removal efficiency decreased from 82.49%±1.90%to 64.57%±3.96%after the IAC was reduced from 60 to 20 mg N/L,while the nitrate production ratio increased from 13.87%±0.90%to 26.50%±3.76%.NOB activity increased to1,133.86 mg N/m^(2)/day after the IAC decreased,approximately 4-fold,indicating that the IAC plays an important inhibitory role in NOB.The rate-limiting step in the mature biofilm of the PN/A system is the nitritation process and is not shifted by the IAC.The analysis of the microbial community structure in the biofilm indicates that the IAC was the dominant factor in changes in community structure.Ca.Brocadia and Ca.Jettenia were the main anammox bacteria,and Nitrosomonas and Nitrospira were the main AOB and NOB,respectively.IAC did not affect the difference in growth between Ca.Brocadia and Ca.Jettenia.Thus,modulating the IAC promoted the PN/A process with efficient nitrogen removal performance at medium to low ammonium concentrations. 展开更多

Nitrification,a central process in the marine nitrogen cycle,produces regenerated nitrate in the euphotic zone and emits N_(2)O,a potent greenhouse gas as a by-product.The regulatory mechanisms of nitrification in the Southern Ocean,which is a critical region for CO_(2)sequestration and radiative benefits,remain poorly understood.Here,we investigated the in situ and dark nitrification rates in the upper 500 m and conducted substrate kinetics experiments across the Indian Sector in the Cosmonaut and Cooperation seas in the late austral summer.Our findings indicate that light inhibition of nitrification decreases exponentially with depth,exhibiting a light threshold of 0.53%photosynthetically active radiation.A positive relationship between dark nitrification and apparent oxygen utilization suggests a dependence on substrate availability from primary production.Importantly,an increased NH_(4)^(+) supply can act as a buffer against photo-inhibitory damage.Globally,substrate affinity(α)increases with depth and transitions from light to dark,decreases with increasing ambient NH_(4)^(+)and exhibits a latitudinal distribution,reflecting substrate utilization strategies.We also reveal that upwelling in Circumpolar Deep Water(CDW)stimulates nitrification through the introduction of potentially higher iron and deep diverse nitrifying microorganisms with higherα.We conclude that although light is the primary limiting factor for nitrification in summer,coupling between substrate availability and CDW upwelling can overcome this limitation,thereby alleviating photoinhibition by up to 45%±5.3%. 展开更多

Partial nitrification is a key aspect of efficient nitrogen removal,although practically it suf-fers from long start-up cycles and unstable long-term operational performance.To address these drawbacks,this study investigated the effect of low intensity ultrasound treatment combined with hydroxylamine(NH2OH)on the performance of partial nitrification.Results showthat compared with the control group,low-intensity ultrasound treatment(0.10W/mL,15 min)combined with NH2OH(5 mg/L)reduced the time required for partial nitrification initiation by 6 days,increasing the nitrite accumulation rate(NAR)and ammonia nitro-gen removal rate(NRR)by 20.4% and 6.7%,respectively,achieving 96.48% NRR.Mechanis-tic analysis showed that NH2OH enhanced ammonia oxidation,inhibited nitrite-oxidizing bacteria(NOB)activity and shortened the time required for partial nitrification initiation.Furthermore,ultrasonication combined with NH2OH dosing stimulated EPS(extracellular polymeric substances)secretion,increased carbonyl,hydroxyl and amine functional group abundances and enhanced mass transfer.In addition,16S rRNA gene sequencing results showed that ultrasonication-sensitive Nitrospira disappeared from the ultrasound+NH_(2)OH system,while Nitrosomonas gradually became the dominant group.Collectively,the results of this study provide valuable insight into the enhancement of partial nitrification start-up during the process of wastewater nitrogen removal. 展开更多

A two-year field experiment conducted under dryland conditions in semi-humid and drought-prone regions of China aimed to assess the effect of ammonia-oxidizing bacterial on maize water use efficiency and yield.A heterotrophic ammonia-oxidizing bacteria(HAOB)strain S2_8_1 was used.Six treatments were applied:(1)no irrigation+HAOB strain(DI),(2)no irrigation+blank culture medium(DM),(3)no irrigation control(DCK),(4)irrigation+HAOB(WI),(5)irrigation+blank culture medium(WM),and(6)irrigation control(WCK).Results revealed that HAOB treatment increased maize growth,yield,and water use efficiency over controls,regardless of whether the year was wet or dry.This improvement was attributed to the accelerated nitrification in the rhizosphere soil due to HAOB inoculation,which subsequently led to increased levels of leaf cytokinins.Overall,these findings suggest that HAOB inoculation holds promise as a strategy to boost water use efficiency and maize productivity in dryland agriculture. 展开更多

Biological nitrification inhibitors(BNIs)are released from plant roots and inhibit the nitrification activity of microorganisms in soils,reducing NO_(3)^(‒)leaching and N2O emissions,and increasing nitrogenuse efficiency(NUE).Several recent studies have focused on the identification of new BNIs,yet little is known about the genetic loci that govern their biosynthesis and secretion.We applied a combined transcriptomic and metabolomic analysis to investigate possible biosynthetic pathways and transporters involved in the biosynthesis and release of BNI 1,9-decanediol(1,9-D),which was previously identified in rice root exudates.Our results linked four fatty acids,icosapentaenoic acid,linoleate,norlinolenic acid,and polyhydroxy-α,ω-divarboxylic acid,with 1,9-D biosynthesis and three transporter families,namely the ATP-binding cassette protein family,the multidrug and toxic compound extrusion family,and the major facilitator superfamily,with 1,9-D release from roots into the soil medium.Our finding provided candidates for further work on the genes implicated in the biosynthesis and secretion of 1,9-D and pinpoint genetic loci for crop breeding to improve NUE by enhancing 1,9-D secretion,with the potential to reduce NO_(3)^(‒)leaching and N2O emissions from agricultural soils. 展开更多

The use of nitrification inhibitors has been suggested as a strategy to decrease cadmium(Cd)accumulation in crops.However,the most efficient nitrification inhibitor for mitigating crop Cd accumulation remains to be elucidated,and whether and how changes in soil microbial structure are involved in this process also remains unclear.To address these questions,this study applied three commercial nitrification inhibitors,namely,dicyandiamide(DCD),3,4-dimethylpyrazole phosphate(DMPP),and nitrapyrin(NP),to pakchoi.The results showed that both DCD and DMPP(but not NP)could efficiently decrease Cd concentrations in pakchoi in urea-and ammonium-fertilized soils.In addition,among the three tested nitrification inhibitors,DMPP was the most efficient in decreasing the Cd concentration in pakchoi.The nitrification inhibitors decreased pakchoi Cd concentrations by suppressing acidification-induced Cd availability and reshaping the soil microbial structure;the most effective nitrification inhibitor was DMPP.Ammonia oxidation generates the most protons during nitrification and is inhibited by nitrification inhibitors.Changes in environmental factors and predatory bacterial abundance caused by the nitrification inhibitors changed the soil microbial structure and increased the potential participants in plant Cd accumulation.In summary,our study identified DMPP as the most efficient nitrification inhibitor for mitigating crop Cd contamination and observed that the soil microbial structural changes caused by the nitrification inhibitors contributed to decreasing Cd concentration in pakchoi. 展开更多

Nitrification and Denitrification Activities and N_2O Emission of Orchard Soils Cultivated for Different Periods of Time

[Method] Incubation experiment was conducted to determine the ni- trification and denitrification activities and N20 emission of three types of orchard soil samples that had been cultivated for 5, 12 and 20 years, respectively, by using the virgin soil sample as control. [Result] After 26 d of incubation, the nitrification rates of nitrogen fertilizer in the virgin soil sample and the orchard soil samples cultivated for 5, 12 and 20 years were 6.85%, 10.26%, 13.29% and 12.90%, respectively, which were positively correlated with content of soil organic matter, ammonium nitro- gen and total nitrogen (P〈0.05), and negatively correlated with soil carbon-nitrogen ratio and pH value (P〈0.05). The denitrification activities of these soil samples in- creased with the increase of cultivation years. The amount of nitrogen loss by deni- trification accounted for 0.01%-3.11% of the amount of fertilizer nitrogen, and had a positive correlation with the content of soil organic matter (P〈0.05). The N20 emis- sions of orchard soil samples were higher than that of the virgin soil samples (P〈 0.05). [Conclusion] In South China, the nitrification activity of orchard soil is relatively low, but it has a tendency to increase as the cultivation years increases; the denitri- fication activity is relatively high, and increases significantly with the increase of culti- vation years. 展开更多

Nitrification of the MBBR was inhibited almost completely during start-up period. Sudden increase of influent total NH 3 concentration was the main factor inducing nitrification inhibition. Increasing DO concentration in the bulk liquid (from 2 to 3 mg/L) had little effect on nitrification recovery. Nitrification of the MBBR recovered partially by the addition of nitrifying sludge into the reactor and almost ceased within 5 days. Nitrification ratio of the MBBR achieved 65% within 12 days by increasing dilute ratio of the influent wastewater with tap water. The ratio of nitrification decreased to 25% when influent COD concentration increased from 650 to 1000 mg/L after nitrification recovery and recovered 70% for another 4 days. 展开更多

For urban wastewater treatment,we conducted a novel four-stage step-feed wastewater treatment system combined with a fluidized bed laboratory experiment to investigate chemical oxygen demand(COD),NH4+-N,and total nitrogen(TN) removal performance.The removal rates of COD,NH4+-N and TN were 88.2%,95.7%,and 86.4% with e?uent concentrations of COD,NH4+-N and TN less than 50,8,and 10 mg/L,respectively.Biomass and bacterial activities were also measured,with results showing more nitrobacteria in the activated sludge than in the biofilm;however,bacterial activity of the biofilm biomass and the activated sludge were similar.Nitrogen concentrations during the process were also detected,with simultaneous nitrification and denitrification found to be obvious. 展开更多

The effects of chlorothalonil and carbendazim on nitrification and denitrification in six soils in upland and rice paddy environments were investigated. Laboratory aerobic (60% water holding capacity) and anaerobic (flooded) conditions were studied at 25℃ and fungicide addition rates of 5.5 mg/kg A. I. (field rate, FR), 20 times (20FR) and 40 times (40FR) field rate, respectively. The results indicated that chlorothalonil at the field rate had a slight inhibitory effect on one soil only, and that soil did not nitrify much in the first place. But chlorothalonil at higher rates inhibited nitrification significantly in all soils. For soils JXP and JXU with a pH of less than 5.0, chlorothalonil almost completely stopped their nitrification at 20FR and 40FR during the whole 14 d incubation period. For soils HNP and HNU with a pH of greater than 8.0, chlorothalonil also significantly inhibit nitrification at 20FR and 40FR (p 〈 0.05). However, NH4+ that was added to the soil was also almost completely nitrified by the end of the incubation period in these two soils. The effects of chlorothalonil at 20FR and 40FR on the nitrification of JSP and JSU soils, with a pH of 5.4 and 7.2, respectively, were intermediate between the other soil types. Chlorothalonil had no effect on denitrification at the field rate and had little effect at the higher rates of application in some soils. Carbendazim had essentially no effect on nitrification and denitrification in soils assessed. 展开更多

A facility of BaPS (Barometric Process Separation) was used to determine soil respiration, gross nitrification and denitrification in a winter wheat field with depths of 0-7, 7--14 and 14-21 cm. N2O production was determined by a gas chromatograph. Crop root mass and relevant soil parameters were measured. Results showed that soil respiration and gross nitrification decreased with the increase of soil depth, while denitrification did not change significantly. In comparison with no-plowing plot, soil respiration increased significantly in plowing plot, especially in the surface soil of 0-7 cm, while gross nitrification and denitrification rates were not affected by plowing. Cropping practice in previous season was found to affect soil gross nitrification in the following wheat-growing season. Higher gross nitrification rate occurred in the filed plot with preceding crop of rice compared with that of maize for all the three depths of 0-7, 7-14 and 14-21 cm. A further investigation indicated that the nitrification for all the cases accounted for about 76% of the total nitrogen transformation processes of nitrification and denitrification and the N2O production correlated with nitrification significantly, suggesting that nitrification is a key process of soil N2O production in the wheat field. In addition, the variations of soil respiration and gross nitrification were exponentially dependent on root mass (p〈0.00l). 展开更多

The distillery wastewater of Guangdong Jiujiang Distillery, which is characteristic of containing high organic matters and rich total nitrogen, was treated by a combination of methane fermentation and denitrification/nitrification processes. 80% of COD in the raw wastewater was removed by methane fermentation at the COD volumetric loading rate of 20 kg COD/(m^3·d) using the expanded granule sludge bed (EGSB) process. However, almost all the organic nitrogen in the raw wastewater was converted into ammonia by ammonification there. Ammonia and volatile fatty acids (VFA) remaining in the anaerobically treated wastewater were simultaneously removed utilizing VFA as an electron donor by denitrification occurring in the other EGSB reactor and nitrification using PEG-immobilized nitrifying bacteria with recirculation process. An aerobic biological contact oxidization reactor was designed between denitrification/nitrification reactor for further COD removal. With the above treatment system, 18000-28000 mg/L of COD in raw wastewater was reduced to less than 100 mg/L. Also, ammonia in the effluent of the system was not detected and the system had a high removal rate for 900-1200 mg/L of TN in the raw wastewater, only leaving 400 mg/L of nitrate nitrogen. 展开更多

The ammonification, nitrification and denitrification in maize rhizosphere of alluvial soils were compared with those in the bulk soil after exposure to different kinds of heavy metals. The addition of cadmium at low levels (2 mg kg-1 and 5 mg kg-1) could stimulate the ammonification and nitrification in the soils, while inhibition influences were found at high levels of Cd addition (10 mg kg-1 and 20 mg kg-1). The relationship between microbial activity and cadmium concentration varied with the kind of microorganisms. The nitrifying bacteria were more sensitive to cadmium pollution than the ammonifying bacteria. When Cd(II), Cu(II) and Cr(VI) were compared at the same addition concentration of 20 mg kg-1 soil, Cd(II) was the most effective inhibitor of ammonification and denitrification among the three investigated heavy metals, and Cr(VI) had the most strong inhibitory influence on the nitrifying bacteria. The microbial activities in rhizosphere were higher than those in the bulk soil for most of the treatments. Cr(VI) proved to be the most effective in enhancing the microbial activities in rhizosphere, and this could be caused by the positive reduction of Cr(VI) to Cr(III) in rhizosphere, and the relatively sufficient existence of organic matter which intensified the adsorption of the metal. It seemed that the rhizosphere had some mitigation effect on heavy metal toxicity. 展开更多

In order to improve nitrogen removal in anoxic/oxic(A/O) process effectively for treating domestic wastewaters, the influence factors, DO(dissolved oxygen), nitrate recirculation, sludge recycle, SRT(solids residence time), influent COD/TN and HRT(hydraulic retention time) were studied. Results indicated that it was possible to increase nitrogen removal by using corresponding control strategies, such as, adjusting the DO set point according to effluent ammonia concentration; manipulating nitrate recirculation flow according to nitrate concentration at the end of anoxic zone. Based on the experiments results, a knowledge-based approach for supervision of the nitrogen removal problems was considered, and decision trees for diagnosing nitrification and denitrification problems were built and successfully applied to A/O process. 展开更多
