Study on the Mechanism of Mahuang Xixin Fuzi Decoction in Treating AR and Prediction of Its Q-Markers Based on Network Pharmacology and Molecular Docking

Objective This study aimed to investigate the potentialmechanism of Mahuang Xixin Fuzi Decoction in treating allergic rhinitis(AR)and predict its quality markers(Q-markers)using network pharmacology and molecular docking techniques.Methods The chemical components of the herbal constituents in Mahuang Xixin Fuzi Decoction were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Active component-related targets were screened using the SwissTargetPrediction database,while AR-related targets were obtained from the GeneCards database.The intersection targets(potential therapeutic targets of the Mahuang Xixin Fuzi Decoction for AR)were identified via the Venn 2.1.0 platform,and a Venn diagram was constructed.A“herb–active component–potential target”network was established using Cytoscape 3.10.0,and core components were screened via topological analysis.Protein–protein interaction(PPI)network of the intersection targets was built using the String database,followed by topological analysis to identify core targets.Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed on the core targets using the DAVID database.Molecular docking of core components and targets was conducted using AutoDock Tools 1.5.7.Results Twenty-seven active components were identified from TCMSP,with 506 corresponding targets predicted by SwissTargetPrediction.A total of 2,447 AR-related targets were retrieved from GeneCards,yielding 165 intersection targets.Network analysis revealed naringenin,genkwanin,deoxyandrographolide,karakoline,and karanjin as core components.PPI network analysis identified 32 core targets.GO enrichment analysis screened 834 functional items,including 618 biological processes,72 cellular components,and 144 molecular functions.KEGG analysis identified 165 signaling pathways.Molecular docking confirmed stable binding between core components and key targets.Conclusion Multiple chemical components in Mahuang Xixin Fuzi Decoction may ameliorate AR by regulating diverse targets and biological processes.Naringenin,genkwanin,and deoxyandrographolide are proposed as potential Q-markers for this decoction in AR treatment. 展开更多

Objective The objective of this study was to investigate the preparation process of Mahuang Xixin Fuzi decoction granules based on the clinical pharmacological effects of the classic formulas and to control the quality of the product.Methods An orthogonal experimental design was used,with extraction yield and transfer rate of key ingredients as evaluation indicators,to optimize the extraction process ofMahuang Xixin Fuzi decoction.The granulation process was optimized based on indicators such as granule formation rate,dissolution rate,and moisture absorption rate.Results The optimal extraction process for Mahuang Xixin Fuzi decoction was determined as the following:using 10 times the amount of water to soak for 30 minutes,extracting three times,each for 60minutes,with a concentrated extract relative density of 1.3 g/mL.A mixture of dextrin and starch at a 1:2 ratio was used as a binder for wet granulation.The granule formation rate,dissolution time,and dilution rate of three repeated batches showed relative standard deviation(RSD)values of 1.48,2.83,and 1.55%.Conclusion This method is stable and feasible,providing valuable data for the industrial production of Mahuang Xixin Fuzi decoction granules and further research on this formulation. 展开更多

Objective: To reveal the mechanism of Mahuang Lianqiao Chixiaodou decoction and its disassembled formula for improving the skin barrier function in a mouse model of atopic dermatitis(AD).Methods: Sixty specific-pathogen free male BALB/c mice were randomly divided into the control group,model group, whole formula group(WF), exterior-releasing formula group(ERF), interior-clearing formula group(ICF), and positive control group(PC). A mouse model of AD was established using the semiantigen 2,4-dinitrofluorobenzene induction method. The lesion scores, transepidermal water loss and p H, and skin histopathology of mice in each group were observed. The expressions of filaggrin, loricrin,and involucrin were detected by the streptavidin peroxidase immunohistochemical method and western blotting, and their mRNA expressions were detected by quantitative polymerase chain reaction.Results: Mice in the WF, ERF, ICF, and PC groups showed reduced skin lesion performance, improved histopathology, decreased skin lesion score, transepidermal water loss and pH, and upregulated expressions of proteins including filaggrin, loricrin, and involucrin, and their mRNAs. The most obvious regulatory effect was observed in the WF group, followed by the ICF, ERF, and PC groups, accordingly.Conclusions: Mahuang Lianqiao Chixiaodou decoction and its disassembled formula can improve the skin barrier function in a mouse model of AD by upregulating filaggrin, loricrin, and involucrin, and their mRNA expressions, and the most optimal effect was noted in the WF group, followed by the ICF and ERF groups, which suggests that the effect of clearing heat and resolving dampness in improving the skin barrier function of AD is more obvious and is one of the key treatments for AD. 展开更多

Objective:To explore the potential mechanism of intervention on the immune imbalance of atopic dermatitis(AD) by studying the effects of Mahuang Lianqiao Chixiaodou decoction(MLCD) on skin damage and inflammation factors in an AD-like mouse model.Methods:Ninety-six male BALB/c mice were divided into normal,model,positive control(mometasone furoate),and traditional Chinese medicine treatment(MLCD) groups by a random number table.2,4-dinitrofluorobenzene was used to induce AD-like mice in all groups except the normal group.The treatment or intervention was administered for seven consecutive days on days 4,18,32,and 39.The mRNA relative expressions of interleukin-4(IL-4),IL-10,interferon-γ(IFN-γ),thymic stromal lymphopoietin(TSLP),and the TSLP receptor(TSLPR) were measured using quantitative real-time polymerase chain reaction,and the serum immunoglobulin E,IL-4,IL-10,and IFN-γ levels were detected using enzyme-linked immunosorbent assay.Results:Compared with the normal group,the hematoxylin-eosin staining of the skin lesions of the mice in the model group was significantly thickened on days 11,25,and 39.Compared with the model group,the epidermal thickness of the positive control group was significantly alleviated on day 39(P <.001),and that of the MLCD group was significantly improved on days 25 and 39(P <.001).Compared with the four observation time points,MLCD had the best treatment effect on day 39 of the experiment and significantly improved the skin damage performance and relieved pathological lesions.On day 39,compared with the model group,MLCD downregulated the skin mRNA relative expressions of IL-4(P=.009),TSLP(P=.030),and TSLPR(P <.001),and reduced the mouse serum levels of IL-4(P=.003).For other serum indicators,no significant difference was observed between the model and MLCD groups.Conclusion:MLCD improved AD-like mice skin damage by regulating the Th1/Th2 immune imbalance. 展开更多

Methods: Randomized controlled trials of all the languages of MahuangFuzi Xixin Decoction on sick sinus syndrome were collected by computer search and manual retrieval. Theretrieval time was from January 2000 to January 2017. According to the inclusion and exclusion criteria, 2reviewers independently selected and extracted data, then evaluated the quality, cross-checked the information andevaluated the quality of menthodology. Through discussion or third reviewer to help solve the divergence, RevMan5.3 software was used to perform meta analysis. Results: A total of 7 documents (n = 612) were finally enrolled,with 358 in Mahuang Fuzi Xixin Decoction group (treatment group) and 254 in control group. Meta analysisshowed that the treatment (86.9%) was more effective than the control (70.1%), the difference was statisticallysignificant (RR = 1.25, 95% CI:(1.15-1.37), P 〈 0.001); the treatment (17.0%) was safer than the control (49.8%),the difference was statistically significant (RR=0.23,95% CI:(0.06-0.93), P =0.04). Conclusion: The existingclinical studies suggest that Mahuang Fuzi Xixin Decoction on sick sinus syndrome is effective and safe; due to thelimited quality of the enrolled documents, the above conclusions need more high-quality randomized controlledtrials to be verified. 展开更多

Objective:As a traditional herbal formula,Mahuang Xixin Fuzi decoction(MXFD)has been used to treat allergic rhinitis(AR).It regulates the transcription factor GATA3 in T cells,blocks Th2 skewing and rebalances the Th1/Th2 immunity.Type 2 innate lymphoid cells(ILC2s)are closely associated with GATA3.However,it remains unknown whether ILC2s could be one mechanism through which MXFD acts.We sought to elucidate the efficacy and mechanism of action of this decoction in AR treatment.Methods:Forty C57BL/6J female mice were equally divided into control,model,loratadine-and MXFDtreated groups in random.AR was induced by ovalbumin(OVA),and then treatment with loratadine or MXFD was administered.AR scores were evaluated and compared before and after treatment.Pathological changes in the nasal mucosa and lung were observed using hematoxylin-eosin staining of tissue samples.Activation of ILC2 in nasal mucosa was assessed by immunofluorescence and quantitative polymerase chain reaction analysis.ILC2 cell count in lungs was measured by flow cytometry and levels of interleukin-(IL)4,IL-5 and IL-13 in serum were detected by ELISA.Results:The decoction alleviated the symptoms of AR in mice,improved vascular congestion and expansion,glandular hyperplasia and inflammatory cell infiltration in mouse nasal mucosa and slowly improved the interstitial pneumonia and lesions.Meanwhile,MXFD reduced the number and percentage of ILC2s in the nasal mucosa and lungs,downregulated the expression of GATA3 mRNA and RORa mRNA,and reduced the related inflammatory cytokine levels,including IL-4,IL-5 and IL-13.Conclusion:These data suggest that inhibition of ILC2s by MXFD may be an important means by which to treat AR. 展开更多

Methods: the children with acute exacerbation of bronchial asthma who were treated in our hospital from January 2021 to January 2022 were selected as the research subjects, and the effects of different treatments were compared, including shegan mahuang decoction and acupoint application + shegan mahuang decoction. Results: after the effective treatment plan was implemented, the condition improved, especially the children treated with acupoint application + shegan mahuang decoction had a higher total effective rate, a lower incidence of adverse reactions, a good trend in FVC and other levels, and symptoms and signs. The scores were lower, and the children who received acupoint application treatment were relatively poor, and the difference was significant (P<0.05). Conclusion: in the treatment of acute exacerbation of bronchial asthma in children, shegan mahuang decoction combined with acupoint application has obvious application value and high safety, and plays a positive role in further shortening the course of the disease in children. 展开更多

目的 探讨麻黄连翘赤小豆汤治疗湿疹风寒外束、湿热蕴肤型患者临床疗效。方法 选择湿疹风寒外束、湿热蕴肤型患者96例,按照简单随机数字表法分为对照组与观察组,各48例。对照组使用曲安奈德益康唑乳膏联合依巴斯汀治疗,观察组在对照组... 目的 探讨麻黄连翘赤小豆汤治疗湿疹风寒外束、湿热蕴肤型患者临床疗效。方法 选择湿疹风寒外束、湿热蕴肤型患者96例,按照简单随机数字表法分为对照组与观察组,各48例。对照组使用曲安奈德益康唑乳膏联合依巴斯汀治疗,观察组在对照组基础上加用麻黄连翘赤小豆汤治疗。评估2组临床疗效和症状积分,比较2组外周血干扰素γ(IFN-γ)、白细胞介素-4(IL-4)水平及特异性转录因子T-bet、GATA结合蛋白3(GATA-3)表达变化情况。结果 观察组临床疗效总有效率(95.83%,46/48)高于对照组(83.33%,40/48)(P<0.05)。治疗后,观察组单项症状评分、外周血IL-4水平、T-bet/GATA-3低于对照组(P<0.05),外周血IFN-γ水平、GATA-3高于对照组(P<0.05)。结论 麻黄连翘赤小豆汤可明显提高湿疹风寒外束、湿热蕴肤型患者临床疗效,缓解临床症状,改善辅助性T细胞1(Th1)/辅助性T细胞2(Th2)失衡。 展开更多

探究麻黄连翘赤小豆汤(MHLQ)及其主要效应成分治疗IgA肾病(IgAN)的可能作用机制。采用牛血清白蛋白(BSA)灌胃、皮下注射四氯化碳(CCl4)、尾静脉注射脂多糖(LPS)等综合措施建立IgAN大鼠模型。成模后,将IgAN大鼠随机分为模型组,MHLQ低、... 探究麻黄连翘赤小豆汤(MHLQ)及其主要效应成分治疗IgA肾病(IgAN)的可能作用机制。采用牛血清白蛋白(BSA)灌胃、皮下注射四氯化碳(CCl4)、尾静脉注射脂多糖(LPS)等综合措施建立IgAN大鼠模型。成模后,将IgAN大鼠随机分为模型组,MHLQ低、中、高剂量组,连翘苷(PHI)组,盐酸伪麻黄碱(PSE)组及氯沙坦钾(LP)组,并以Wistar大鼠为对照。MHLQ低、中、高剂量组分别给予MHLQ颗粒剂1.773、3.545、7.090 g·kg^(-1)灌胃,PHI组给予PHI 0.020 g·kg^(-1)灌胃,PSE组给予PSE 0.020 g·kg^(-1)灌胃,LP组给予LP 9.003 mg·kg^(-1)灌胃,对照组和模型组每日灌胃等体积生理盐水,各组均连续干预4周。给药结束后收集各组大鼠尿液、血清、肝组织和肾组织。检测24 h尿蛋白定量(24 h UTP)和肾功能;病理染色观察肾组织病理改变;免疫荧光(IF)检测肾脏IgA和补体C3/C3b/C3c表达;电镜观察肾组织超微结构;酶联免疫吸附测定(ELISA)法检测血清和肾组织补体C3、可溶性端补体复合物(sC5b-9)蛋白表达;蛋白免疫印迹(Western blot)法检测肝、肾补体C3/C3b/C3c、C5/C5a、C5b-9及B因子(CFB)蛋白表达;免疫组化(IHC)进一步检测肾组织补体C3蛋白表达。结果显示,与对照组比较,模型组大鼠尿素氮(BUN)、血肌酐(SCr)含量升高,肾小球代偿性扩张,球内系膜细胞增殖,系膜基质扩张,肾小球可见IgA免疫复合物或电子致密物沉积;血清补体C3含量增加;肾组织CFB,补体C3/C3b/C3c、C5/C5a、C5b-9蛋白表达升高;肝组织中补体C3/C3b/C3c和C5b-9蛋白表达升高。而经过MHLQ及其效应成分干预后,上述指标皆逆转。综上所述,MHLQ及其效应成分干预IgAN大鼠疗效肯定,能够改善肾功能,减轻肾脏免疫复合物沉积和病理损伤,其机制可能与抑制IgAN大鼠替代途径(AP)补体激活有关。 展开更多

目的基于“诸气膹郁,皆属于肺”理论,通过孟德尔随机化(Mendelian randomization,MR)分析探讨肺腺癌(lung adenocarcinoma,LUAD)与重度抑郁症(major depressive disorder,MDD)的遗传因果关系,并预测干预LUAD相关性抑郁(lung adenocarcin... 目的基于“诸气膹郁,皆属于肺”理论,通过孟德尔随机化(Mendelian randomization,MR)分析探讨肺腺癌(lung adenocarcinoma,LUAD)与重度抑郁症(major depressive disorder,MDD)的遗传因果关系,并预测干预LUAD相关性抑郁(lung adenocarcinoma-related depression,LAD)的潜在中药。方法从IEU OpenGWAS数据库获取LUAD与MDD的全基因组关联研究(genome-wide association studies,GWAS)数据集,主要通过R软件的TwoSampleMR包进行MR分析和结果的稳健性评估。通过Ensembl数据库预测工具变量的邻近基因(instrumental variable nearby genes,IVNGs),基于IVNGs进行功能富集分析并分别通过CTD数据库和ITCM数据库预测相关的化学成分和潜在的干预中药。应用古今医案云平台(V2.3.9)对中药进行统计分析,以探讨其治疗疾病的理法方药。结果MR分析的逆方差加权(inverse variance weighted,IVW)方法结果(β=0.00205,P=0.01899)提示LUAD与MDD发生存在弱正向因果效应,稳健性分析未发现显著的异质性和水平多效性,结果可靠。富集分析结果显示主要与胆碱能信号通路有关。聚类分析结果显示频次在前30的中药可大致分为5大类。所有潜在干预中药的四气以温为主,平、寒次之;五味以甘为主,苦、辛次之;归经以肝为主,肺、脾、胃、肾和心次之;功效主要以清热解毒为主,凉血止血、解毒、止血及清热利湿次之。对频次前15的中药进行经典名方识别,结果显示主要是通过麻黄、栀子、薏苡仁、蒲公英和肉苁蓉5味中药识别,相似的经典名方有麻黄类方、槐花散、黄连解毒汤、苇茎汤、五味消毒饮、济川煎、化肝煎和桑杏汤等。结论基于“诸气膹郁,皆属于肺”理论,发现LUAD与MDD存在弱正向遗传因果效应,LUAD可能通过影响神经信号传递及神经调控机制,尤其是胆碱能信号通路促进MDD的发生。结合中医“态靶”医学理论和中药预测结果探析了中医治疗LAD的理法方药,旨在为LAD患者的中医药治疗提供参考,以改善LUAD患者的整体治疗效果及心理健康。 展开更多

经典名方是中医药古籍中的精髓,凝聚着中医药文化的核心价值,是中华民族瑰宝。对经典名方的关键信息进行考证总结,有利于全面挖掘其发展方向和发展现状,为后续的研究奠定基础。厚朴麻黄汤作为国家公布的百首经典名方之一,最早来源于东... 经典名方是中医药古籍中的精髓,凝聚着中医药文化的核心价值,是中华民族瑰宝。对经典名方的关键信息进行考证总结,有利于全面挖掘其发展方向和发展现状,为后续的研究奠定基础。厚朴麻黄汤作为国家公布的百首经典名方之一,最早来源于东汉张仲景的《金匮要略》,由厚朴、麻黄、杏仁、石膏、五味子、半夏、干姜、细辛和小麦九味药物组成,功效为散饮除热、止咳平喘,主要用于治疗寒饮内停、挟热迫肺证。作者系统整理历代中医古籍中有关厚朴麻黄汤的文献记载,对其来源、药物组成、主治病证、药物用量、药物基原和炮制、服法和制法等关键信息进行考证与分析。共收集相关中医古籍55部(包含日本古籍6部),其中26部中医古籍详细记载了厚朴麻黄汤组成、主治病证、炮制、剂量等内容。研究表明,厚朴麻黄汤在明清时期应用最为广泛。随着后世流传,其主治病证、组成、用药剂量、炮制方法、煎服方法等均略有变化,但大体与《金匮要略》一致。通过对记载厚朴麻黄汤的古籍挖掘整理,以期对厚朴麻黄汤的临床应用及成方制剂提供理论参考。 展开更多

【目的】探究麻黄鱼腥散治疗牛支原体肺炎的作用机制。【方法】通过中药系统药理学数据库与分析平台(TCSMP)筛选麻黄鱼腥散中有效成分的作用靶点。采用GeneCards、OMIM、DrugBank、CTD和PharmGkb数据库搜索与牛支原体肺炎相关的靶点。... 【目的】探究麻黄鱼腥散治疗牛支原体肺炎的作用机制。【方法】通过中药系统药理学数据库与分析平台(TCSMP)筛选麻黄鱼腥散中有效成分的作用靶点。采用GeneCards、OMIM、DrugBank、CTD和PharmGkb数据库搜索与牛支原体肺炎相关的靶点。取化合物靶点和疾病靶点的交集,获取麻黄鱼腥散治疗牛支原体肺炎的潜在作用靶点。采用Cytoscape软件构建“MHYXS-牛支原体肺炎-靶点”网络图。将交集靶点导入STRING数据库构建PPI蛋白互作网络并结合拓扑学分析筛选关键靶点。随后通过基因本体(GO)功能富集、京都基因与基因组百科全书(KEGG)通路富集分析对交集靶点进一步细化研究,最后使用Autodock Vina软件对关键靶点与作用的活性化合物进行分子对接。【结果】从麻黄鱼腥散5味药材中筛选有效成分106个,潜在作用靶点125个;筛选牛支原体肺炎靶点9090个;预测得到麻黄鱼腥散治疗牛支原体的潜在作用靶点共119个;蛋白互作网络分析发现,CDK1和CCNA2可能是麻黄鱼腥散治疗牛支原体肺炎的核心靶点;经GO富集分析得到890个细胞生物学过程,主要涉及刺激反应的正向调节、凋亡过程和程序性细胞死亡等生物学过程;KEGG通路富集分析获得166条相关信号通路,主要包括PI3K、TNF、IL17和MAPK等信号通路;分子对接结果显示,CDK1与黄芩素有较高亲和力,CCNA2与脱氧喜树碱具有较高亲和力。【结论】麻黄鱼腥散可能通过黄芩素、脱氧喜树碱等活性成分作用于CDK1和CCNA2等靶点,通过PI3K、TNF和IL-17信号通路等发挥治疗牛支原体肺炎的作用。 展开更多

麻黄升麻汤是《伤寒论》中颇具争议的一方,历代医家对其方证、条文病机、组方思路、临床应用等众说纷纭。该文以麻黄升麻汤的药物剂量为线索明确药物主次,以药证为基础剖析组方原则,归纳其方证总结治则治法,进而揭示条文背后的潜在病机... 麻黄升麻汤是《伤寒论》中颇具争议的一方,历代医家对其方证、条文病机、组方思路、临床应用等众说纷纭。该文以麻黄升麻汤的药物剂量为线索明确药物主次,以药证为基础剖析组方原则,归纳其方证总结治则治法,进而揭示条文背后的潜在病机为热毒郁闭,邪气阻滞气机于内,阳气不达四末则见手足厥逆;邪无出路则充斥上下,上攻咽喉则见咳唾脓血;下行大肠,耗伤阴液则见下利不止。在此病机基础上对本方的适应证及适用人群做了进一步的宏观刻画,增加了原条文未记载的方证应用范畴,以此为临床用方提供一定参考。同时,该文也对几个争议问题做了初步探讨,认为条文中“下部脉”应为下肢动脉搏动,条文所述的症状和体征与感染性休克类似,麻黄升麻汤应属治温病方,并非仲景原方。本文应用从药物剂量、配伍原则、药证相应等方面归纳经方方证、内在病机、临床应用的方法,从新的角度解读了麻黄升麻汤的内涵,为理解其临床应用提供了更为全面的视角和新的思路。 展开更多

射干麻黄汤“鼻肺同治”调控T淋巴细胞改善小鼠气道高反应的免疫机制

目的观察射干麻黄汤对气道高反应(AHR)小鼠模型上、下气道T淋巴细胞亚群的影响,分析其“鼻肺同治”控制AHR的生物学基础。方法采用T细胞β受体基因敲除小鼠提纯γδT细胞;采用卵清蛋白气道致敏的AHR模型小鼠,平均随机分为正常对照组(Ctr... 目的观察射干麻黄汤对气道高反应(AHR)小鼠模型上、下气道T淋巴细胞亚群的影响,分析其“鼻肺同治”控制AHR的生物学基础。方法采用T细胞β受体基因敲除小鼠提纯γδT细胞;采用卵清蛋白气道致敏的AHR模型小鼠,平均随机分为正常对照组(Ctrl组)、气道高反应模型组(AHR组)、过继免疫γδT细胞组(AIT组)、射干麻黄汤干预组(SGMHD组)、射干麻黄汤+过继免疫组(SMA组),AIT组予以每日γδT细胞尾静脉注射,SGMHD组予以每日射干麻黄汤灌胃,SMA组予以每日γδT细胞尾静脉注射联合射干麻黄汤灌胃,干预7 d。观察各组小鼠鼻黏膜和肺组织中T淋巴细胞亚群标志物干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、视黄酸相关孤儿受体γt(RORγt)、叉头状转录因子P3(FoxP3)及γδT细胞受体(γδTCR)分布情况,外周血Th1、Th2、Th17、Treg、γδT细胞及其细胞因子的表达水平。结果与Ctrl组相比,AHR组鼻黏膜和肺组织中IFN-γ、IL-4、FoxP3、RORγt、γδTCR表达较高,外周血Th2、Th17、γδT细胞比例较高,IFN-γ、IL-2、IL-4、IL-5及IL-17浓度均较高,IL-10、TGF-β则表达较低;与AHR组相比,AIT组、SGMHD组及SMA组鼻黏膜IFN-γ、IL-4、RORγt、γδTCR表达较低,肺组织IL-4、γδTCR较低,外周血Th1比例较高,Th17、γδT较低,SGMHD组和SMA组IFN-γ、IL-2、IL-4、IL-5及IL-17浓度较低,IL-10、TGF-β较高。结论明确了射干麻黄汤“鼻肺同治”改善AHR模型小鼠的作用,其机制与全面调控T淋巴细胞亚群有关。研究为揭示射干麻黄汤治疗AHR作用机制、促进临床应用奠定基础。 展开更多
