Two unusual sesquiterpenoids,irpexlactones A(1)and B(2),were isolated from cultures of the basidiomycete Irpex lacteus.Their structures were established by means of spectroscopic methods,the single crystal X-ray diffraction,as well as electronic circular dichroism(ECD)calculations.They possess a novel carbon skeleton with a 5/6/3-fused ring system that may derive from tremulane type sesquiterpenoids with ring-rearrangement.Both compounds show significant inhibitory activities against nitric oxide production with half maximal inhibitory concentration(IC50)values of 2.2 and 1.4μmol/L,respectively.Their anti-inflammatory effects were further evaluated by enzyme-linked immunosorbent assay(ELISA)and Western blot. 展开更多

Irpex lacteus has been widely used for treating chronic glomerulonephritis as a traditional Chinese medicine.Seven water-soluble polysaccharide fractions(ILN I,ILN II,ILN III,ILA I,ILA II,ILB I and ILB II) were isolated and purified from Irpex lacteus by hot-water extraction,deproteinization,decolorization,dicthy laminoethyl(DEAE)-cellulose ion exchange and sephadex G100 chromatographies,respectively.The average molecular weights and monosaccharide composition of these polysaccharide fractions greatly differed from each other.The antitumor and antinephritis activities of the seven polysaccharide fractions were evaluated.It was found that ILN III displayed significant inhibition effects on both humar hepatocellular liver carcinoma(HepG2) and hentietta lacks(HeLa) tumor cells with IC 50 values of 60.95 and 99.95 μg/mL,respectively.ILA I exhibited significant inhibition effects on murine mesangial cells(HBZT-1) with an IC 50 value of 185.06 μg/mL.The inhibition effects of other polysaccharide fractions on these three cells were significantly different.These results suggest that the polysaccharide fractions isolated from Irpex lacteus have potential antitumor and antinephritis activities. 展开更多

Objective:To investigate the cytotoxic activity of endophytic fungi isolated from mangrove fungi.Methods:In the present study the DNA was isolated and the ITS region of 5.8s rRNA was amplified using specific primers ITS 1 and ITS4 and sequence was determined using automated sequencers.Blast search sequence similarity was found against the existing non redundant nucleotide sequence database thus,identified as Aspergilus flavus,Hyporcaea lixii,Aspergillus niger,Eutorium amstelodami,Irpex hydnoides and Neurospora crassa.Among the seven isolates, one fungi Irpex hydnoides was selected for further studies.The fungi were grown in sabouraud broth for five days and filtrate were separated and subjected to ethyl acetate for further studies. Results:Nearly half(49.25%) of the extracts showed activity(IC_(50) of 125 μ g/mL).These values were within the cutoff point of the National Cancer Institute criteria for cytotoxicity(IC_(50)<20 μ g/mL) in the screening of crude plant extracts.The GC MS analysis revealed that the active principals might be Tetradecane(6.26%) with the RT 8.606.Conclusions:It is clear from the present study that mangrove fungi with bioactive metabolites can be expected to provide high quality biological material for high throughout biochemical,anti cancer screening programmes.The results help us conclude that me potential of using metabolic engineering and post genomic approaches to isolate more novel bioactive compounds and to make their possible commercial application is not far off. 展开更多

Traditional autoclaving,slow degradation rate and preservation of biomass treated by fungi are the main factors restricting biological treatment.In our previous studies,strains with high efficiency and selective lignin degradation ability were obtained.To further solve the limiting factors of biological treatment,this paper proposed a composite treatment technology,which could replace autoclaves for fungal treatment and improve the preservation and utilization of fungal-pretreated straw.The autoclaved and expanded buckwheat straw were,respectively,degraded by Irpex lacteus for 14 days(CIL,EIL),followed by ensiling of raw materials(CK)and biodegraded straw of CIL and EIL samples with Lactobacillus plantarum for different days,respectively(CP,CIP,EIP).An expansion led to lactic acid bacteria,mold,and yeast of the samples below the detection line,and aerobic bacteria was significantly reduced,indicating a positive sterilization effect.Expansion before I.lacteus significantly enhanced lignin selective degradation by about 6%,and the absolute content of natural detergent solute was about 5%higher than that of the CIL.Moreover,EIL decreased pH by producing higher organic acids.The combination treatment created favorable conditions for ensiling.During ensiling,EIP silage produced high lactic acid about 26.83 g/kg DM and the highest acetic acid about 22.35 g/kg DM,and the pH value could be stable at 4.50.Expansion before I.lacteus optimized the microbial community for ensiling,resulting in EIP silage co-dominated by Lactobacillus,Pediococcus and Weissella,whereas only Lactobacillus was always dominant in CP and CIP silage.Clavispora gradually replaced Irpex in EIP silage,which potentially promoted lactic acid bacteria growth and acetic acid production.In vitro gas production(IVGP)in EIL was increased by 30%relative to CK and was higher than 24%in CIL.The role of expansion was more significant after ensiling,the IVGP in EIP was increased by 22%relative to CP,while that in CIP silage was only increased by 9%.Silage of fungal-treated samples reduced methane emissions by 28%to 31%.The study demonstrated that expansion provides advantages for fungal colonization and delignification,and further improves the microbial community and fermentation quality for silage,enhancing the nutrition and utilization value.This has practical application value for scaling up biological treatment and preserving the fungal-treated lignocellulose. 展开更多

本文旨在研究白耙齿菌提取物(Metabolites of Irpex lacteus,MIL)对2型糖尿病(Type 2 Diabetes Mellitus,T2DM)小鼠的降糖作用及其作用机制。通过腹腔注射链脲佐菌素(STZ)诱导T2DM小鼠模型,将小鼠分为正常组、模型组、二甲双胍组及MIL... 本文旨在研究白耙齿菌提取物(Metabolites of Irpex lacteus,MIL)对2型糖尿病(Type 2 Diabetes Mellitus,T2DM)小鼠的降糖作用及其作用机制。通过腹腔注射链脲佐菌素(STZ)诱导T2DM小鼠模型,将小鼠分为正常组、模型组、二甲双胍组及MIL高、中、低剂量组,给药5周后检测空腹血糖(FBG)、口服糖耐量(OGTT)、肝糖原(Liver Glycogen)、肌酐(SCR)、尿氮素(BUN)、胰岛素(INS)及葡萄糖转运蛋白4(GLUT4)的含量,并对肾脏进行病理学观察;通过网络药理学预测通路及靶点,采用RT-qPCR、Western blot技术对T2DM小鼠相关基因的mRNA和蛋白表达量进行检测。结果表明,MIL能极显著降低T2DM小鼠FBG、BUN、SCR、INS及OGTT-AUC水平(P<0.01),极显著升高GLUT4及Liver Glycogen水平(P<0.01);肾脏病理学观察显示MIL可以改善肾脏组织中细胞的炎症状态,有效缓解高血糖引起的肾炎病症;MIL还可以显著上调蛋白激酶B(AKT)、GLUT4、磷脂酰肌醇3激酶(PI3K)并下调INS的m RNA表达量(P<0.05),显著上调pPI3K和p-AKT的蛋白表达量(P<0.05)。上述结果表明,MIL可能通过调节PI3K/AKT信号通路来调节血糖紊乱。 展开更多

目的研究白耙齿菌(Irpex lacteus)的次级代谢产物及其降脂活性。方法运用硅胶柱色谱及制备液相色谱Pre-HPLC等多种色谱分离方法对白耙齿菌的次级代谢产物进行分离纯化,结合核磁共振波谱、高分辨质谱、红外光谱、紫外光谱等谱学技术对分... 目的研究白耙齿菌(Irpex lacteus)的次级代谢产物及其降脂活性。方法运用硅胶柱色谱及制备液相色谱Pre-HPLC等多种色谱分离方法对白耙齿菌的次级代谢产物进行分离纯化,结合核磁共振波谱、高分辨质谱、红外光谱、紫外光谱等谱学技术对分离所得的单体化合物进行结构鉴定。建立高脂HepG2细胞模型进行降脂活性测试。结果从白耙齿菌中分离并鉴定了12个化合物,分别为(2α,4β,5β,7β,10α)-2,5,11-eudesmanetriol(1)、11,12-epoxy-5,6-secotremula-1,6(13)-dien-5,12-olide(2)、8α,10α-di-O-acetyl-lactarorufin A(3)、(+)-(2S,3R,6S,7S)-tremul-1(10)-ene-2,12-diol(4)、conocenolide A(5)、irpexolactin G(6)、2,2′-oxybis(1,4-di-tert-butylbenzene)(7)、4′-hydroxy-5,7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone(8)、环(异亮-缬)二肽(9)、环(L-脯-L-亮)二肽(10)、环(亮-异亮)二肽(11)和环(亮-亮)二肽(12)。降脂活性结果显示,化合物1~4、8、9均具有降脂活性,其中化合物1、4的各剂量组及化合物2、3、8的高、中剂量组对甘油三酯(TG)含量的降低效果均优于阳性药组。结论化合物1~4,7~12为首次从该菌中分离得到。化合物1~4、8和9均具有较好的降脂效果。 展开更多

为了解白囊耙齿菌腐朽木材的分子机制,利用基因差异显示技术(Differential display reverse transcriptase,PCRDDRT-PCR)研究了白囊耙齿菌腐朽白桦木材前后基因表达的差异.共分离出5个腐朽后在菌丝体中特异表达的cDNA片段,分别命名为A2... 为了解白囊耙齿菌腐朽木材的分子机制,利用基因差异显示技术(Differential display reverse transcriptase,PCRDDRT-PCR)研究了白囊耙齿菌腐朽白桦木材前后基因表达的差异.共分离出5个腐朽后在菌丝体中特异表达的cDNA片段,分别命名为A2、A7、C6、B8和B3.序列同源性比对发现:片段A2、A7和C6分别与编码漆酶(Laccase)、纤维素酶A(Cellulase A)和麦芽糊精磷酸化酶(Maltodextrin phosphorylase)等3个基因具有高度同源性;B8与一个编码响应调节蛋白的基因具有高度同源性.研究结果显示,所筛选的差异表达基因A2、A7和C6等与木材降解有密切关系. 展开更多
