摘要
应用响应面法优化白囊耙齿菌中腺苷的超声法提取工艺。在单因素试验的基础上,采用3因素3水平的Box-Behnken中心组合设计和响应面分析法,考察超声提取时间、超声功率和液料比对腺苷得率的影响,并优化腺苷超声法提取条件。白囊耙齿菌中腺苷的最佳超声提取条件为:提取时间378 s、功率414 W、液料比37:1(v/w),理论提取率为0.88%,实际提取率为0.89%,相对误差为1.14%。
A response surface analysis method was applied to optimize the ultrasonic conditions for extraction of adenosine in Irpex lacteuss mycelia.Based on the single factor experiments,a three-factor-three-level experiment was designed by Box-Behnken central composite design method.The influences of extraction time,ultrasonic power and solvent/solid ratio on the yield of adenosine were studied.The response surface method was employed to analyze the results of experiments.The results indicated that the optimum ultrasonic extraction conditions of adenosine were as follows:extraction time 378 s,ultrasonic power 414 W and solvent/solid ratio 37:1(v/w).The predictive maximum yield of adenosine was 0.88%.The average yield of adenosine in three validation experiments under the optimum process conditions was 0.89%and the relative error was 1.14%.
