Objective:To evaluate the antioxidant potential of methanolic leaf extract of Indigofera cassioides(MEIC) using various in vilro antioxidant assay systems.Methods:Antioxidant and free radical scavenging activity of MEIC was assayed by using different in vitro models like ABTS, DPPH,nitric oxide,superoxide,hydrogen peroxide and hydroxyl radical.Reductive ability of the extract was tested by the complex formation with potassium ferricyanide.Further total phenol and flavonoid contents of the crude extract were also determined.Rutin and ascorbic acid were used as standards.Results:MEIC exhibited potent and concentration dependent free radical scavenging activity in all the tested models.Reductive ability was also found to increase with increase in MEIC concentration.Total phenol and flavonoid content determination showed that the extract is rich in phenols and flavonoids.Conclusions:All the results of the in vitro antioxidant assays reveal potent antioxidant and free radical scavenging activity of the leaves of Indigofera cassioides,equivalent to that of standard ascorbic acid and rutin.This potent antioxidant activity may be attributed to its high phenolic and flavonoid contents. 展开更多

Objective:To evaluate the antitumor,cytotoxic and antioxidant activities of methanolic leaf extract of Indigofera cassioides(MEIC) against transplantable tumors and human cancer cell lines.Methods:MEIC was investigated for its short term cytotoxicity on EAC and DLA cells by trypan blue dye exclusion method and in vitro cytotoxicity on HeLa,HEp-2,HEpG-2,MCF-7, HT-29,Vero and NIH 3T3 cells by MTT assay.In vivo antitumor activity was studied on EAC and DLA tumor bearing mice.Activity was assessed by monitoring the mean survival time,effect on hematological parameters,antioxidant enzyme levels and solid tumor volume.Results:MEIC exhibit potent in vitro cytotoxicity against all the tested cancer cell lines,but it was found to be safe on normal cells.The extract significantly(P【0.001) increase the mean survival time and also have a protective effect on the hemopoietic system at the tested dose levels(200 and 400 mg/kg). The extract prevented lipid peroxidation and restored the antioxidant enzymes catalase, superoxide dismutase,glutathione peroxidase and glutathione-s-transferase in the liver of tumor control animals.It also significandy(P【0.01) reduce the solid tumor volume.Conclusions:The results strongly support that MEIC shows potent antitumor and cytotoxic effects against EAC,DLA and human cancer cell lines.The extract prevents lipid peroxidation and promotes the enzymatic antioxidant defense system in tumor bearing animals which might be due to activities like scavenging of free radicals by the phytochemicals in MEIC. 展开更多

The objective of this study was to evaluate the free radical scavenging potential and high performance thin layer chromatography (HPTLC) fingerprinting of Indigofem tinctoria (I. tinctoria), Phytochemical analysis was carried out using standard methods, and free radical scavenging activity of the plant was determined using 2,2-diphenyl-l-picrylhydrazy (DPPH), nitric oxide (NO) and superoxide anion (O2-) radical scavenging capacities. HPTLC plate was kept in CAMAG TLC Scanner 3 and the Rf values at fin- gerprint data were recorded by WlNCATS software, Aqueous extract of I. tinctoria reliably showed the total phenolics (267.2 ± 2.42 mg/g), flavonoids (75.43 ± 3.36 mg/g) and antioxidants (349.11 ±8.04 mg/g). The extract was found to have DPPH (52.08%), NO (23.12%) and 02 (26.79%) scavenging activities at the concentration of 250 pg/mL and the results were statistically significant compared with ascorbic acid standard (p 〈 0.05). HPTLC results confirmed that the extract contained several potential active com- ponents such as phenols, flavonoids, saponins and terpenoids as the slides revealed multi-colored bands of varying intensities. This study confirmed that the plant had multipotential antioxidant and free ra- dicals scavenging activities. 展开更多

Indigofera heterantha commonly called indigo bush is a member of leguminoseae family found in the Himalayan region of Kashmir. A lectin has been isolated from the seeds of Indigofera heterantha by the purification procedure involving anion exchange chromatography on DEAE-cellulose followed by gel filtration chromatography on Sephadex G 100. Molecular characterization of the lectin was done by gel filtration and SDS-PAGE. Activity of the lectin was checked by hemagglutination assay and the sugar specificity by sugar inhibition tests. The antimicrobial activity of the purified lectin was carried out by Agar disc diffusion using appropriate standards. On the ion exchange column, the bound protein when eluted with 0-0.5 M NaCl gradient emerged as three peaks—peak I, peak II and peak III out of which only peak II showed the hemagglutinating activity. The lectin further resolved into two peaks G1 and G2 on gel filtration, with the lectin activity residing in G1, corresponding to a molecular weight of 70 KDa. The purified lectin named as Indigofera heterantha Lectin (IHL) produced a single band on SDS PAGE (18 KDa), revealing the tetrameric nature of the lectin. It agglutinated human erythrocytes (A, B, AB, and O). Hemagglutination was inhibited by D-galactose, Dmannose and D-arabinose. The lectin is reasonably thermostable showing full activity within a temperature range of 30&degC to 90&degC. pH stability of the lectin falls in the range of 2-9. IHL demonstrated a remarkable antibacterial activity against the pathogenic bacteria Klebsiella pneumoniae, Staphylococcus aureus, Escherichia coli, and Bacillus subtilis. IHL also inhibited the growth of phytopathogenic fungi Aspergillus niger, Aspergillus oryzae and Fusarium oxysporum. 展开更多

OBJECTIVE:To compare and elucidate the antioxidant effi cacy of ethanolic and hydroethanolic extracts of Indigofera tinctoria Linn.(Fabaceae family).METHODS:Various in-vitro antioxidant assays and free radical-scavenging assays were done.Quantitative measurements of various phytoconstituents,reductive abilities and chelating potential were carried out along with standard compounds.Half inhibitory concentration(IC50)values for ethanol and hydroethanol extracts were analyzed and compared with respective standards.RESULTS:Hydroethanolic extracts showed considerably more potent antioxidant activity in comparison to ethanol extracts.Hydroethanolic extracts had lower IC50values than ethanol extracts in the case of DPPH,metal chelation and hydroxyl radical-scavenging capacity(829,659 and 26.7μg/m L)but had slightly higher values than ethanol in case of SO2-and NO2-scavenging activity(P〈0.001 vs standard).Quantitative measurements also showed that the abundance of phenolic and fl avonoid bioactive phytoconstituents were signifi cantly(P〈0.001)greater in hydroethanol extracts(212.920 and 149.770 mg GAE and rutin/g of plant extract respectively)than in ethanol extracts(211.691 and 132.603 mg GAE and rutin/g of plant extract respectively).K arl Pearson’s co rrelation analysis(r2)between various antioxidant parameters and bioactive components also associated the antioxidant potential of I.tinctoria with various phytoconstituents,especially phenolics,fl avonoids,saponins and tannins.CONCLUSION:This study may be helpful to draw the attention of researchers towards the hydroethanol extracts of I.tinctoria,which has a high yield,and great prospects in herbal industries to produce inexpensive and powerful herbal products. 展开更多

This study investigated the possible neuroprotective role oflndigofera oblongifolia leaf methanolic extract (IOLME) against lead-induced neurotoxicity. Rats were intraperitoneally injected with lead acetate, with or without IOLME (intragastric administration for 5 days), and the neuroprotective effect of IOLME was assessed by measuring the lead concentration, redox status (lipid peroxidation, nitric oxide and glutathione), enzymatic antioxidant activities (superoxide dismutase, catalase, glutathione peroxidase and reductase), PCR assays of apoptosis markers (Bax and Bcl-2) and histopathology of the brain. The increases in the lipid peroxidation, nitric oxide, and apoptosis, the decreases in the glutathione level and the activity of antioxidant enzymes, and the altered histology of the brain induced by lead acetate were mitigated in the brain of rats pre-treated with IOLME. These findings indicate that IOLME has beneficial effects and it mitigates lead acetate-induced neurotoxicity via its antiox-idant and anti-apoptotic activities. 展开更多

(Fabaceae), used in traditional medicine in Burkina Faso to treat malaria, helminthiasis, and aqueous extracts of the leaves from Indigofera berhautiana Gillet (Fabaceae) used to treat hepatitis and typhoid fever. <strong>Methods:</strong> Aqueous decoction of leaves from <em>Sesbania pachycarpa</em> DC. or <em>Indigofera berhautiana</em> Gillett which are the form of use recommended by traditional health practitioners, were used for tests. Aqueous extracts used to evaluate the acute toxicity, were studied on mice of strain NMRI. For the toxicity study, the doses of the aqueous extracts were respectively 5 mg/kg, 50 mg/kg, 300 mg/kg and 2000 mg/kg of body weight. Phytochemical screening was done to identify the phytochemicals contained in extracts. <strong>Results:</strong> Aqueous extracts of two species: <em>Sesbania pachycarpa</em> DC., <em>Indigofera berhautiana</em> Gillet were not toxic at the maximum dose of 2000 mg/kg of body weight. Phytochemical screening, showed coumarins, flavonoids, tannins, polyphenols compounds, steroids and/or triterpenes and saponosides in the aqueous extracts of the two species. 针对干热河谷区柑橘园气候干旱燥热、水热矛盾突出、植被覆盖率低、水土流失严重等问题,开展生草栽培对柑橘园土壤理化性质、酶活性及土壤微生物多样性的影响研究,旨在为干热河谷区柑橘园绿色可持续生产-生草栽培模式的应用提供参考。... 针对干热河谷区柑橘园气候干旱燥热、水热矛盾突出、植被覆盖率低、水土流失严重等问题,开展生草栽培对柑橘园土壤理化性质、酶活性及土壤微生物多样性的影响研究,旨在为干热河谷区柑橘园绿色可持续生产-生草栽培模式的应用提供参考。结果表明:与清耕相比,紫花苜蓿和铺地木蓝两种生草模式使0~20 cm土层土壤温度分别显著降低了3.97℃和5.80℃,均显著改善了土壤持水能力(土壤质量含水量分别提高至13.34%和15.07%),并降低了土壤容重(分别降低17.26%和8.93%),从而改善了土壤物理性质;在土壤化学性质方面,两种生草模式均提高了土壤pH值、有机质含量和有效磷含量,但对全氮、全磷、全钾含量有降低作用;土壤综合肥力评价显示,铺地木蓝处理的土壤综合肥力最高,其次为紫花苜蓿处理,清耕处理最低;两种生草模式均增加了土壤细菌多样性,提高了土壤脲酶、蔗糖酶、纤维素酶活性,但对过氧化氢酶活性的影响因草种而异。两种生草模式通过协同调控土壤水热平衡、养分循环及微生物代谢,均显著提升了干热河谷区柑橘园土壤综合质量。因此,紫花苜蓿、铺地木蓝均适合在干热河谷区柑橘园间种。 展开更多

干旱及复水对河北木蓝生物量分配与渗透调节特征的影响 被引量:1

植物在遭受干旱胁迫后复水,会经历应激适应、伤害、修复过程,且不同植物对水分亏缺的敏感性不同,复水后的恢复效应也存在差异。因此,研究植物耐旱性与复水后的恢复效应对植物抗旱节水育种具有重要意义。本研究采用盆栽控水模拟干旱胁迫... 植物在遭受干旱胁迫后复水,会经历应激适应、伤害、修复过程,且不同植物对水分亏缺的敏感性不同,复水后的恢复效应也存在差异。因此,研究植物耐旱性与复水后的恢复效应对植物抗旱节水育种具有重要意义。本研究采用盆栽控水模拟干旱胁迫和复水处理,以野生型和栽培型河北木蓝为对象,在持续干旱胁迫3、6、10和13 d以及复水2和7 d后进行生长及生理生化指标测定,研究干旱胁迫和复水条件下2种木蓝的生物量分配策略和响应干旱的渗透调节生理机制及其种内差异。结果表明:1)随干旱持续时间的延长,2种木蓝器官生物量、根系分布特征(总根长、根表面积、根体积、根分枝数)、淀粉含量以及根-中性转化酶(neutral invertase,NIV)活性逐渐降低;而根冠比、脯氨酸(Pro)含量、丙二醛(MDA)含量、蔗糖含量、蔗糖合酶(sucrose synthase,SUS)活性、α-淀粉酶(α-amylase,α-AL)活性、叶-NIV活性均逐渐增加。2)复水处理7 d后,除了根系分布特征和MDA含量外,其他指标均恢复至对照水平,均具有补偿生长能力。3)基于各指标的主成分分析结果,相比之下,栽培型木蓝各指标变化幅度均显著大于野生型,对干旱胁迫具有更强的耐受性。由此可见,2种木蓝在干旱胁迫下积累渗透调节物质,以此维持细胞膨压,缓解细胞膜受损,并调控蔗糖和淀粉代谢酶活性,进而使生物量分配倾向于根系,为促进根系对水分吸收而提供能量,复水后二者均具有显著的补偿效应。 展开更多

不同比例‘热研4号’王草与穗序木蓝混合青贮效果

为生产优质的青贮饲料,探讨不同比例‘热研4号’王草(Pennisetum purpureum×P. glaucum‘Reyan No.4’)和穗序木蓝(Indigofera hendecaphylla)混合青贮后其营养成分和发酵品质,以‘热研4号’王草和穗序木蓝按质量比8:2(A1组)、6:4... 为生产优质的青贮饲料,探讨不同比例‘热研4号’王草(Pennisetum purpureum×P. glaucum‘Reyan No.4’)和穗序木蓝(Indigofera hendecaphylla)混合青贮后其营养成分和发酵品质,以‘热研4号’王草和穗序木蓝按质量比8:2(A1组)、6:4(A2组)、4:6(A3组)、2:8(A4组)进行混合青贮,对照组(CK)为‘热研4号’王草单独青贮,发酵60 d后取样分析。结果表明,随着穗序木蓝比例的增加,处理组的干物质、粗蛋白质、Ca含量及pH、相对饲喂价值呈增加趋势,脂肪、粗灰分、中性洗涤纤维、氨含量及相对饲草品质呈降低趋势;各处理组的干物质、粗蛋白、脂肪、粗灰分、中性洗涤纤维、Ca含量及pH、相对饲喂价值、相对饲草品质呈现不同程度的显著性差异(P<0.05)。各处理组的干物质、粗蛋白、Ca、乳酸含量及相对饲喂价值均显著高于CK组(P<0.05),粗灰分、中性洗涤纤维、可溶性碳水化合物含量均显著低于CK组(P<0.05),脂肪、酸性洗涤纤维、氨、丁酸、总可消化养分含量及相对饲草品质在处理组和CK组间均呈现差异不显著(P > 0.05)。通过发酵品质评价,各处理组的费氏评分均显著高于CK组(P<0.05),A4组得分最高;各处理组的费氏青贮得分均低于CK组,A4组得分最低。通过混合青贮效果的综合评价可知,各处理组和CK组的排序为A2组> A4组> A3组> A1组> CK组。因此,在‘热研4号’王草中添加穗序木蓝进行混合青贮,青贮效果得到改善,当‘热研4号’王草与穗序木蓝的比例为6:4时最佳。 展开更多

为明确苗药血人参的化学成分及抗炎活性物质,本研究通过建立血人参UPLC指纹图谱方法,利用化学计量法对其成分进行比较分析,结合网络药理学-分子对接预测血人参抗炎的活性有效靶点,并采取体外细胞实验对其抗炎药效进行验证。血人参指纹... 为明确苗药血人参的化学成分及抗炎活性物质,本研究通过建立血人参UPLC指纹图谱方法,利用化学计量法对其成分进行比较分析,结合网络药理学-分子对接预测血人参抗炎的活性有效靶点,并采取体外细胞实验对其抗炎药效进行验证。血人参指纹图谱结合化学计量法分析结果显示,不同产地血人参共得到28个共有峰,其中指认了10个共有峰,并首次指认出血人参所含成分牡荆素、木犀草苷、金丝桃苷和汉黄芩素。聚类分析可将不同产地血人参聚为3类,主成分分析与聚类分析结果一致,可相互佐证。体外细胞实验表明,不同产地的血人参对炎症细胞有不同程度的修复作用。网络药理学结果表明,表皮生长因子受体(epidermal growth factor receptor,EGFR)和过氧化物酶体增殖物激活受体(peroxisome proliferator-activated receptor gamma,PPARG)是抗炎潜在的靶点,EGFR酪氨酸激酶抑制剂耐药性被认为是抗炎的主要途径之一。本研究建立了血人参的谱-效关系,并结合网络药理学阐明血人参抗炎的关键靶点及作用通路,为血人参的质量控制和品质评价提供参考依据。 展开更多

为探索多花木蓝灌草利用方式对贵州黑山羊生长性能及瘤胃发酵参数的影响,选取5月龄体重相近的贵州黑山羊36只,随机分为3组,分别饲喂象草(CK)、多花木蓝灌草鲜饲(T1)和多花木蓝灌草混贮料(T2)等不同粗饲料,试验期57 d。结果表明:多花木... 为探索多花木蓝灌草利用方式对贵州黑山羊生长性能及瘤胃发酵参数的影响,选取5月龄体重相近的贵州黑山羊36只,随机分为3组,分别饲喂象草(CK)、多花木蓝灌草鲜饲(T1)和多花木蓝灌草混贮料(T2)等不同粗饲料,试验期57 d。结果表明:多花木蓝灌草利用方式对贵州黑山羊的平均日增重(ADG)、干物质采食量(ADFI)、料重比(F/G)、瘤胃液NH_(3)-N及乙酸(AA)、丙酸(PA)、丁酸(BA)含量均有显著影响(P<0.05)。T1、T2处理试验羊的ADG、ADFI均显著高于CK处理(P<0.05),增幅分别在28.35%和9.39%以上,而F/G则显著低于CK处理,降幅分别为14.99%和12.98%,但两处理间无显著差异。各处理间试验羊瘤胃液pH和AA/PA无显著差异,但T2处理瘤胃液的NH_(3)-N显著高于T1和CK处理,而AA、PA、BA含量显著低于T1和CK处理(P<0.05)。综上所述,多花木蓝混合鲜饲或以灌草混合青贮方式饲喂贵州黑山羊均可提高其生长性能,维持瘤胃功能稳定,且以灌草混贮饲喂有利于提高能量利用率。 展开更多

载血人参石油醚部位胶束-白及多糖水凝胶对大鼠慢性创面愈合的促进作用

目的观察载血人参石油醚部位胶束-白及多糖水凝胶(ISs-M-Gel)对大鼠慢性创面愈合的促进作用。方法80只雄性SPF级SD大鼠随机分为A、B、C、D、E组,每组16只。在各组大鼠背部制作2.5 cm×2.5 cm大小的创口,肌内注射青霉素钾(4000单位/... 目的观察载血人参石油醚部位胶束-白及多糖水凝胶(ISs-M-Gel)对大鼠慢性创面愈合的促进作用。方法80只雄性SPF级SD大鼠随机分为A、B、C、D、E组,每组16只。在各组大鼠背部制作2.5 cm×2.5 cm大小的创口,肌内注射青霉素钾(4000单位/次,1次/天,连续4 d)和醋酸氢化可的松(80 mg/kg,1次/天,连续7 d),以制备慢性创面模型。制模成功后,A组大鼠创面涂抹纯水,B组大鼠创面涂抹牛碱性成纤维细胞生长因子外用溶液,C组大鼠创面涂抹血人参石油醚部位(ISs),D组大鼠创面涂抹空白胶束-白及多糖水凝胶(Blank-M-Gel),E组大鼠创面涂抹ISs-M-Gel,每日涂抹1次,连续21 d。拍摄干预第3、7、14、21天的创面,计算创面愈合率;在上述时间点选取各组大鼠,取血制备血清,采用酶联免疫吸附试验检测血清白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α),并取创面再生皮肤组织进行HE、Masson染色,另采用总超氧化物歧化酶(SOD)和丙二醛(MDA)试剂盒检测创面再生皮肤组织SOD和MDA含量,使用免疫组化法检测创面再生皮肤组织CD31,以评估血管生成情况。结果干预第14、21天,与A组比较,其余各组创面愈合率均提高(P均<0.05);与A组比较,干预第7、14天B组和E组大鼠血清IL-6与TNF-α水平降低,干预第14、21天B组和E组创面再生皮肤组织SOD升高而MDA降低(P均<0.05)。与其他给药组比较,B组与E组再生皮肤纤维组织沉积更密集。干预第14、21天,与A组比较,B、C、E组大鼠创面再生皮肤组织CD31阳性表达增多,血管生成情况最好。结论ISs-M-Gel可促进大鼠慢性创面愈合,且效果好于ISs、Blank-M-Gel,与牛碱性成纤维细胞生长因子外用溶液效果相当。 展开更多

【目的】分析不同程度的干旱胁迫处理对马棘生理指标的影响,探索其抗旱生理响应特征,为马棘抗旱种质筛选和遗传改良提供参考依据。【方法】以重庆地区广泛收集的36份野生马棘种质资源为试验材料,采用盆栽培养的方式待马棘幼苗生长至第9... 【目的】分析不同程度的干旱胁迫处理对马棘生理指标的影响,探索其抗旱生理响应特征,为马棘抗旱种质筛选和遗传改良提供参考依据。【方法】以重庆地区广泛收集的36份野生马棘种质资源为试验材料,采用盆栽培养的方式待马棘幼苗生长至第90天时开始干旱处理,分别在胁迫0 d(对照组CK,土壤相对含水量为田间持水量的80%~85%)、胁迫7 d(中度干旱,土壤含水量为田间持水量的40%~50%)和胁迫14 d(重度干旱,土壤相对含水量为田间持水量的20%~25%)时测定各处理下马棘叶片的渗透调节物质含量、相对含水量和抗氧化酶活性,利用抗旱系数结合隶属函数和聚类分析法对马棘的抗旱性进行综合评价。【结果】在干旱胁迫下,36份马棘种质的生理响应变异丰富,不同种质的抗旱能力差异显著。通过对同一指标的均值比较分析发现,与对照相比,渗透调节物质丙二醛(MDA)、可溶性蛋白(SP)和脯氨酸(Pro)含量随着干旱程度增加而上升;超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性随着干旱程度增加而呈“凸”字形变化;相对含水量(RWC)随着干旱程度增加而降低。7个检测指标中,POD活性的变异系数最大,而RWC的变异系数最小。通过隶属函数、综合D值和聚类分析可将36份马棘种质的抗旱能力分为抗旱型(8份种质)和敏感型(28份种质)两类。通过对两种类型种质的生理指标分析发现,抗旱型种质的抗氧化酶活性和渗透调节物质Pro含量高于敏感型种质。【结论】干旱胁迫下36份马棘种质资源的生理响应具有丰富的多样性和变异,通过综合评价分析初步筛选出8份抗旱型马棘种质。 展开更多

铺地木蓝对肇庆地区柑橘园土壤养分及细菌群落的影响 被引量:3

【目的】探究铺地木蓝在柑橘果园种植1年后对杂草、0~20 cm土层土壤养分含量和土壤细菌多样性的影响。【方法】设置自然生草(CK1)、土壤覆盖遮草布(PD1)、土壤覆盖银黑地膜+种植铺地木蓝(PD2)、种植铺地木蓝(PD3)4个处理,1年后调查杂草... 【目的】探究铺地木蓝在柑橘果园种植1年后对杂草、0~20 cm土层土壤养分含量和土壤细菌多样性的影响。【方法】设置自然生草(CK1)、土壤覆盖遮草布(PD1)、土壤覆盖银黑地膜+种植铺地木蓝(PD2)、种植铺地木蓝(PD3)4个处理,1年后调查杂草种类、测定土壤理化性质(pH、有机质、全氮、碱解氮、全磷、速效磷、全钾和速效钾)和土壤细菌群落情况。【结果】PD2处理土壤全氮(0.86 mg/g)、全磷(0.78 g/kg)、速效磷(23.84 mg/kg)、全钾(11.91 g/kg)、有机质(34.82 g/kg)含量最高;PD2处理土壤全氮和碱解氮含量比PD1处理的分别增加了0.37、27.43 mg/kg,PD1处理土壤全磷含量最低(0.41 g/kg),PD3处理速效磷(10.67 mg/kg)和有机质(25.99 g/kg)含量最低;PD1~PD3处理的土壤全钾含量均大于CK1处理,速效钾含量则相反;杂草调查结果表明PD1、PD2处理均可很好地抑制杂草的生长。与CK1相比,PD1~PD3处理均可降低土壤细菌群落多样性,但不影响细菌群落分布均匀性。PD2和PD3处理的土壤细菌群落结构和丰度较相似,表现为变形菌门(Proteobacteria)相对丰度降低,绿弯菌门(Chloroflexi)、酸杆菌门(Acidobacteria)、浮霉菌门(Planctomycetes)、罗库菌门(Rokubacteria)、芽单胞菌门(Gemmatimonadetes)等有益细菌的相对丰度增加;其中PD2处理的优势菌属为MND1(3.47%)、Sh765B-TzT-35(4.37%),PD3处理的优势菌属为MND1(4.29%)。从PD1处理土壤中检测到视为土壤环境恶化的标志类群异常球菌-栖热菌门(5.10%)。【结论】柑橘果园行间道覆盖地膜后再种植铺地木蓝的综合表现最好,可以增加土壤养分和土壤中有益细菌群落的相对丰度。 展开更多

花青素还原酶(ANR)及无色花青素还原酶(LAR)是原花青素单体生物合成过程中两个关键限速酶,对原花青素的生物合成及积累具有重要作用。本研究以黔产血人参为材料,根据其转录组测序结果,采用生物信息学方法从数据库中鉴定IsANR和IsLAR基因... 花青素还原酶(ANR)及无色花青素还原酶(LAR)是原花青素单体生物合成过程中两个关键限速酶,对原花青素的生物合成及积累具有重要作用。本研究以黔产血人参为材料,根据其转录组测序结果,采用生物信息学方法从数据库中鉴定IsANR和IsLAR基因,并分析其蛋白结构及表达模式,揭示其与血人参原花青素积累的关系。本研究从血人参中鉴定出了2个IsANRs、2个IsLARs基因,其中IsLAR1蛋白的氮端具有叶绿体转运体。IsANR2基因的编码区中存在终止子,导致蛋白翻译提前终止。二级结构分析揭示IsANR1比IsANR2有较高比例的α-螺旋,较低的无规卷曲,IsLAR1比IsLAR2有较高的β-转角。IsANR2主要在叶中表达,IsANR1、IsLAR1在血人参根部具有较高的表达量,且IsLAR1和IsLAR2在根中的表达量随年限的增加而上升。因此,血人参根部原花青素的积累可能与LAR基因(尤其是IsLAR1)的上调表达有密切的关系。本研究结果不仅有助于揭示根部原花青素生物的合成机制,同时也为血人参药材的质量控制提供了理论参考。 展开更多

目的制备一种载苗药血人参Indigofera stachyodes提取物的聚乙烯醇(PVA)/木质素(Lig)/壳聚糖(CS)水凝胶并探究其促创面愈合机制。方法在单因素考察的基础上,正交试验优选处方工艺并对其进行质量评价和体外生物评价,通过建立大鼠全层皮... 目的制备一种载苗药血人参Indigofera stachyodes提取物的聚乙烯醇(PVA)/木质素(Lig)/壳聚糖(CS)水凝胶并探究其促创面愈合机制。方法在单因素考察的基础上,正交试验优选处方工艺并对其进行质量评价和体外生物评价,通过建立大鼠全层皮肤损伤模型,评价其促创愈合效果。结果携载提取物的PVA/Lig/CS水凝胶的最优处方和制备工艺为PVA用量7.0 g、CS用量4.0 g、Lig用量0.5 g、冻融循环次数4次,每次冻结6 h。该凝胶具有多孔网状结构、良好的溶胀性、保湿性及一定的机械强度,同时具有良好的生物相容性、抑菌性以及有效抑制巨噬细胞细胞迁移,且抑制作用可能与时间呈正比。该水凝胶的体内药效学研究显示,其促创面愈合作用可能与减少创面组织炎性细胞浸润,促进巨噬细胞向M2型极化、增加毛血细管生成和纤维组织形成有关。结论制备的载血人参提取物的PVA/Lig/CS水凝胶剂制备工艺简单、稳定可行、具有良好的缓释性能,且对全层皮肤损伤具较好的治疗效果。 展开更多
