OBJECTIVE:To investigate the efficacy of Gouqizi(Fructus Lycii)seed oil(FLSO)on D-gal induced inflammation in testis of rats in vitro and in vivo.METHODS:In aging Sertoli cells(TM4 cells)induced by D-galactose(D-gal),the expression of upregulated agingrelated proteins.The number of cells counted by cell counting kit(CCK)-8 assay showed a high number of cells disposed with FLSO at 50,100 and 150μg/mL compared to that for the aging model.In vivo,male Sprague–Dawley rats(n=50,8-week-old,230-255 g)were randomly categorized into control,aging model,and FLSO(low-,medium-,and high-dose)groups.The expression of nuclear factor-κB(NF-κB)and its upstream factors[Janus kinase 1(JAK1)and signal transducerand activator of transcription 1(STAT1)]was detected by Western blot and immunofluorescence,related inflammatory factors quantified by enzyme-linked immunosorbent assay.Evaluation of testicular tissue by Johnsen score,the spermatogenic function was explored.RESULTS:The expression of interleukin-1β(IL-1β)(P<0.05),IL-6(P<0.001),and tumor necrosis factorα(TNF-α)(P<0.05)was decreased significantly,while that of heme oxygenase-1(HO-1)(P<0.001)and IL-10(P<0.05)was increased in cells disposed with FLSO 100μg/mL.FLSO inhibited the expression of NF-κB and declined pp65/p65(P<0.01),as detected by Western blotting.In vivo,the levels in serum of IL-1β(P<0.001),IL-6(P<0.05),and TNF-α(P<0.01)declined while IL-10(P<0.05)was upregulated post-FLSO treatment.In addition,the expression of JAK-1 and STAT1 increased significantly in testicular tissue of rats treated with FLSO as compared to the aging model of rats(P<0.001),while the expression of NF-κB(P<0.001)declined in the testis in the FLSO group,as assessed by immunofluorescence.The levels of inhibor B and testosterone in serum both increased(P<0.05).CONCLUSIONS:In conclusion,this study determined the protective effects of FLSO to tolerate inflammatory injury in the testis,indicating that FLSO alleviates inflammation via JAK-1/STAT1/NF-κB pathway. 展开更多

Depression poses a considerable threat to global health,underscoring the critical need for safe treatments.Because of its homology of medicine and food,Gardeniae Fructus is widely used in clinical and everyday life contexts.Gardeniae Fructus oil(OGF)is a waste byproduct of the food industry with promising recycling potential.While previous research has demonstrated the antidepressant effects of OGF,the precise mechanism remains unclear.To fill this gap in the literature,this paper elucidates the composition and potential antidepressant mechanism of OGF.According to the results,OGF is mainly composed of fatty acids and unsaponifiable matter.OGF could effectively attenuate depression-like behaviors in mice with chronic unpredictable mild stress by modulating the microbiota-gut-brain axis.Mechanisms include improving gut dysbiosis,regulating short-chain fatty acids,protecting the intestinal barrier function,reducing hippocampal damage,and alleviating neuroinflammation by inhibiting the activation of the TLR4/NF-κB/NLRP3 pathway.Fecal microbiota transplantation experiments confirmed the involvement of the gut microbiota in the modulation of depression-like behavior by OGF.This study establishes the antidepressant effects of OGF as well as the underlying mechanism,demonstrating its potential as a novel dietary intervention for depression treatment. 展开更多

As one of the most common metabolic diseases,type 2 diabetes(T2D)has become a public health concern with rising global prevalence.Corni fructus(CF),a traditional Chinese herb utilized for centuries as homologous medicinal and food resources,has been widely used to treat glucose and lipid metabolism disorders.However,as a core active ingredient of CF,whether and how morroniside(MOR)improves T2D is still unclear.This study aimed to explore the pathways by which MOR ameliorates T2D in mice induced by high-fat diet(HFD)and low-dose streptozotocin through 16S rRNA gene sequencing.We found that MOR treatment significantly ameliorated body weight loss,hyperglycemia,hyperlipidemia and insulin resistance in T2D mice.In addition,MOR remarkably improved inflammation-and oxidative stress-driven hepatic and pancreatic injuries in the model mice.Mechanistically,MOR rehabilitated the dysregulated diversity and constitution of the gut microbiota in T2D mice,with significant changes in relative abundance in genus such as Lactobacillus,Lachnospiraceae_NK4A136_group,and Lachnospiraceae_UCG-006 which are believed to be highly correlated with serum parameters and insulin resistance in mice with T2D.Therefore,we infer that MOR improves T2D at least partially by maintaining the host microbiota homeostasis,and MOR may be a promising candidate for T2D treatment. 展开更多

Diaphragma juglandis(D.juglandis)Fructus is a brownish lamellar structure located between walnut kernels.As a traditional Chinese herbal medicine,it exhibits therapeutic effects including spleen-strengthening,kidney consolidation,astringency,diuresis,and heat-clearing.In this study,the polyphenolic constituents of D.juglandis and its functional instant tea were investigated in this study.Three polyphenolic compounds were isolated from 70%ethanol extract of D.juglandis Fructus as 2-methoxy juglone,regiolone and quercetin.The HPLC method was established for determination of the content of quercetin in the instant tea samples under the premise that the methodology was verifi ed to meet the standard requirements.The preparation process of D.juglandis Fructus instant tea was optimized through single factor experiments and Box-Behnken design-response surface methodology and the optimal conditions were determined as follows:feed rate of 10 mL/min,inlet temperature of 151℃,andβ-cyclodextrin addition of 9%.Then the antioxidant activity of each sample was evaluated using DPPH and ABTS radical scavenging assays.The results showed that the quercetin had significant antioxidant activity.The instant tea group demonstrated superior antioxidant effects compared to the extracts group at 50,200,and 800μg/mL(P<0.001),likely due to the optimized spray-drying process,which might have enhanced the solubility,stability,and bioavailability of the active compounds.The results provided critical foundational data for the deep processing and comprehensive utilization of D.juglandis Fructus. 展开更多

Fructus Ligustri Lucidi(FLL)refers to the dried mature fruit of Ligustrum lucidum Ait.,a species from the Oleaceae family,widely distributed across East Asia and India.This study aimed to optimize the extraction process for Fructus Ligustri Lucidi essential oil(FLLO)to develop an efficient and practical extraction method.Additionally,the chemical composition of FLLO was analyzed,and its antioxidant,antimicrobial,and cytotoxic activities were evaluated.FLLO was extracted using supercritical CO_(2) extraction,and response surface methodology was applied to optimize the extraction parameters:pressure of 16 MPa,temperature of 40℃,and extraction time of 40 min.The main components of the essential oil were identified through GC-MS analysis.Antioxidant activity was assessed using DPPH and ABTS assays,demonstrating that FLLO exhibited strong antioxidant properties,with a DPPH radical scavenging rate exceeding 80%.In antimicrobial tests,FLLO exhibited significant inhibitory effects on both Gram-positive and Gram-negative bacteria at concentrations greater than 25 mg/mL.Additionally,cytotoxicity assays revealed that FLLO enhanced the proliferation of LO2 cells.In conclusion,FLLO,extracted using supercritical CO_(2),demonstrates excellent antioxidant and antimicrobial properties,as well as favorable cell safety,supporting its potential for further development and application of Ligustrum lucidum. 展开更多

Objective:To investigate the effects of Alpiniae oxyphyllae Fructus(AOF)on renal lipid deposition in diabetic kidney disease(DKD)and elucidate its molecular mechanisms.Methods:The mechanism of AOF in treating DKD was explored by network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation.The effects of AOF on renal function and lipid deposition were assessed in a mouse model of DKD and high glucose-stressed HK-2 cells.Cell viability and lipid accumulation were detected by CCK8 and oil red O staining.The expressions of PPARαand fatty acid oxidation-related genes(ACOX1 and CPT1A)were detected by quantitative RT-PCR,Western blot,and immunofluorescence.Furthermore,PPARαknockdown was performed to examine the molecular mechanism of AOF in treating DKD.Results:Network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation showed that the active compounds in AOF targeted PPARαand thus transcriptionally regulated ACOX1 and CPT1A.AOF lowered blood glucose,improved dyslipidemia,and attenuated renal injury in DKD mice.AOF-containing serum accentuated high glucose-induced decrease in cell viability and ameliorated lipid accumulation.Additionally,it significantly upregulated the expression of PPARα,ACOX1,and CPT1A in both in vivo and in vitro experiments,which was reversed by PPARαknockdown.Conclusions:AOF may promote fatty acid oxidation via PPARαto ameliorate renal lipid deposition in DKD. 展开更多

目的 建立不同基原枳实的加味温胆汤高效液相色谱法(HPLC)指纹图谱,并采用化学模式识别分析方法综合评价不同批次加味温胆汤的质量。方法 采用Kromasil 100-5 C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%甲酸溶液为流动相,梯度洗脱... 目的 建立不同基原枳实的加味温胆汤高效液相色谱法(HPLC)指纹图谱,并采用化学模式识别分析方法综合评价不同批次加味温胆汤的质量。方法 采用Kromasil 100-5 C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%甲酸溶液为流动相,梯度洗脱;检测波长318 nm;流速1 mL·min^(-1);柱温30℃;进样量10μL,建立不同基原枳实的加味温胆汤HPLC指纹图谱,并进行相似度分析。采用聚类分析(HCA)、主成分分析(PCA)与正交偏最小二乘法判别分析(OPLS-DA)评价不同批次不同基原枳实的加味温胆汤的质量差异,并确定质量差异化合物。结果 加味温胆汤以不同基原的枳实组方可分成典型的两类HPLC指纹图谱,确定了32~33个共有峰;色谱峰主要来自枳实(酸橙枳实、甜橙枳实)、化橘红、毛冬青、竹茹、五指毛桃。通过与对照品比对鉴定了14个成分,分别为绿原酸(峰4)、新西兰牡荆苷Ⅱ(峰9)、对香豆酸(峰10)、异绿原酸B(峰15)、柚皮芸香苷(峰16)、异绿原酸A(峰17)、柚皮苷(峰18)、橙皮苷(峰19)、异绿原酸C(峰20)、新橙皮苷(峰21)、柚皮素(峰25)、补骨脂素(峰26)、川陈皮素(峰32)、橘皮素(峰33)。以酸橙枳实为枳实基原组成的10批加味温胆汤HPLC指纹图谱相似度为0.950~0.997;以甜橙枳实为枳实基原组成的3批加味温胆汤HPLC指纹图谱相似度为0.952~0.982;2种不同基原枳实组成的加味温胆汤HPLC对照指纹图谱的相似度为0.805。化学模式识别结果显示,13批样品可分为3类,分别为S1~S8、S11~S13、S9~S10,筛选出20个差异色谱峰,其中影响不同基原枳实的加味温胆汤质量差异标志物为橙皮苷、新橙皮苷、柚皮苷、川陈皮素。结论 该研究所建立的不同基原枳实的加味温胆汤HPLC指纹图谱结合化学模式识别方法可系统、全面地评价加味温胆汤的质量差异,可为其新制剂研发的工艺与质量标准研究提供实验基础。 展开更多

目的基于“异类相制”理论,探究淫羊藿Epimedii Folium和女贞子Ligustri Lucidi Fructus对补骨脂Psoraleae Fructus诱导肾阴虚模型大鼠肝损伤的影响及作用机制。方法以补骨脂诱导的肾阴虚模型大鼠肝损伤为参照,分别给予淫羊藿和女贞子... 目的基于“异类相制”理论,探究淫羊藿Epimedii Folium和女贞子Ligustri Lucidi Fructus对补骨脂Psoraleae Fructus诱导肾阴虚模型大鼠肝损伤的影响及作用机制。方法以补骨脂诱导的肾阴虚模型大鼠肝损伤为参照,分别给予淫羊藿和女贞子进行干预,通过体质量、肛温、粪便含水量及血清环磷酸腺苷(cyclic adenosine monophosphate,c AMP)/环磷酸鸟苷(cyclic guanosine monophosphate,c GMP)比值评估肾阴虚指征变化情况;通过血清中丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天冬氨酸氨基转移酶(aspartate aminotransferase,AST)活性及肝组织苏木素-伊红(hematoxylineosin,HE)染色评估肝脏功能变化;结合代谢组学、关联分析、ROC分析及京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析解析潜在机制。结果淫羊藿可显著降低模型大鼠体质量和粪便含水量(P<0.05、0.01),显著升高肛温、cAMP/cGMP比值、ALT及AST活性(P<0.05、0.001),并加重肝脏炎性细胞浸润,加剧肝损伤;女贞子则可显著逆转上述变化。代谢组学鉴定出17个淫羊藿和女贞子差异调控的共同代谢物,关联分析及ROC分析进一步发现神经酰胺、牛磺胆酸等6个诊断效能良好的差异代谢物,主要涉及亚油酸代谢、鞘脂代谢等8条通路。此外,淫羊藿可显著升高模型大鼠血清中炎性因子白细胞介素-1β(interleukin-1β,IL-1β)、IL-6、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平(P<0.05),并降低抗炎因子IL-10水平;女贞子则逆转上述变化。结论淫羊藿和女贞子可能通过差异调控肝脏亚油酸代谢等影响炎性细胞因子水平,从而导致补骨脂诱导的肾阴虚模型大鼠肝脏出现毒/效作用差异。为中医“异类相制”理论提供实验依据,并为补骨脂减毒增效研究提供借鉴。 展开更多

目的比较生草乌及诃子汤制草乌的急性毒性、抗炎、镇痛作用。方法急性毒性试验分为空白对照组[A组,0.5%羧甲基纤维素钠(CMC-Na)],生草乌1.67 g/kg剂量组(B_(1)组)、2.22 g/kg剂量组(B_(2)组)、2.96 g/kg剂量组(B_(3)组)、3.95 g/kg剂量... 目的比较生草乌及诃子汤制草乌的急性毒性、抗炎、镇痛作用。方法急性毒性试验分为空白对照组[A组,0.5%羧甲基纤维素钠(CMC-Na)],生草乌1.67 g/kg剂量组(B_(1)组)、2.22 g/kg剂量组(B_(2)组)、2.96 g/kg剂量组(B_(3)组)、3.95 g/kg剂量组(B_(4)组)、5.26 g/kg剂量组(B_(5)组)、7.01 g/kg剂量组(B6组),诃子汤制草乌组(C组,8.40 g/kg),各10只。各组小鼠按每10 g体质量单次灌胃相应药物0.4 mL,给药2次,间隔12 h,之后常规饲养14 d。考察小鼠的死亡情况、半数致死量(LD50)、体质量及血清乳酸脱氢酶(LDH)、肌酸激酶(CK)、碱性磷酸酶(ALP)、总胆汁酸(TBA)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平。抗炎、镇痛药效试验分为模型对照组(D组,等量0.5%CMC-Na),西药对照组(E组,对乙酰氨基酚0.250 g/kg),生草乌低、中、高剂量组(F_(1)组、F_(2)组、F_(3)组,0.114,0.171,0.342 g/kg),诃子汤制草乌低、中、高剂量组(G_(1)组、G_(2)组、G_(3)组,0.114,0.171,0.342 g/kg),各10只。各组小鼠均按每10 g体质量单次灌胃相应药物0.4 mL,分别进行醋酸扭体试验、热板试验、二甲苯致耳肿胀试验,考察小鼠发生扭体反应的潜伏期及发生频次、疼痛阈值、耳肿胀度及耳组织病理学变化。结果急性毒性试验结果显示,C组、B_(1)组小鼠均未死亡,B6组小鼠均死亡,生草乌的LD50[95%置信区间(95%CI)]为3.4214(2.8523,4.1039)g/kg;与C组比较,B_(3)组小鼠给药后第5,10,14天及B_(4)组小鼠给药后第10,14天的体质量均显著升高(P<0.05),B_(4)组、B_(5)组小鼠给药后第5天的体质量均显著降低(P<0.05);B_(1)组、B_(3)组、B_(4)组、B_(5)组小鼠的ALT和AST水平均有显著差异(P<0.01);B_(1)组、B_(2)组、B_(4)组、B_(5)组小鼠的TBA水平均有显著差异(P<0.01);B_(1)组、B_(2)组、B_(3)组、B_(4)组、B_(5)组小鼠的ALP,CK,LDH水平均有显著差异(P<0.01)。抗炎、镇痛药效试验结果显示,与D组比较,F_(1)组、F_(2)组、F_(3)组、G_(1)组、G_(2)组、G_(3)组小鼠第1次发生扭体反应的潜伏期均显著延长(P<0.05),F_(3)组、G_(1)组、G_(2)组、G_(3)组小鼠的扭体反应发生频次均显著降低(P<0.01);F_(1)组、F_(2)组、F_(3)组、G_(1)组、G_(2)组、G_(3)组小鼠给药后30,60,90,120 min的疼痛阈值均显著升高(P<0.01);F_(1)组、F_(2)组、F_(3)组、G_(1)组、G_(2)组、G_(3)组小鼠的耳肿胀度均显著降低(P<0.01)。生草乌与诃子汤制草乌相同剂量组间比较,F_(1)组、F_(2)组小鼠第1次发生扭体反应的潜伏期分别较G_(1)组、G_(2)组显著缩短(P<0.05),F_(1)组、F_(2)组、F_(3)组小鼠的扭体反应发生频次分别较G_(1)组、G_(2)组、G_(3)组显著升高(P<0.05);F_(1)组、F_(2)组、F_(3)组小鼠给药60,90 min后的疼痛阈值分别较G_(1)组、G_(2)组、G_(3)组显著降低(P<0.05),F_(2)组、F_(3)组小鼠给药120 min后的疼痛阈值分别较G_(2)组、G_(3)组显著降低(P<0.05);各给药组间小鼠耳肿胀度无显著差异(P>0.05)。结论生草乌及诃子汤制草乌均有一定抗炎、镇痛作用,生草乌经诃子汤炮制后具有减毒增效作用。 展开更多

为测定我国肾综合征出血热病毒 Gou3 株的 M 片段全基因序列,了解该株分子基础,并分析其与其他病毒株的差异,将提取的感染了病毒的 Vero - E6 细胞总 R N A 进行逆转录 P C R扩增,产物直接或纯化后克隆于 T 载... 为测定我国肾综合征出血热病毒 Gou3 株的 M 片段全基因序列,了解该株分子基础,并分析其与其他病毒株的差异,将提取的感染了病毒的 Vero - E6 细胞总 R N A 进行逆转录 P C R扩增,产物直接或纯化后克隆于 T 载体并测定序列,结果测得 Gou3 株 M 片段的全基因序列共3 651 个核苷酸,最大读码框架从47 到3 448 ,共编码1 134 个氨基酸。四种核苷酸的比例分别为 A3069 % , T 3001 % , G2155 % , C1775 % , G C 含量为3930 % , A T 含量为6070 % 。应用不同的载体系统、不同的转化用菌种、从不同的传代株中扩增,确证了测序结果的正确性。序列同源比较分析表明: Gou3 株与 H T N 型同源性为705 % ~715 % ,与 S E O 型为839 % ~845 % ;其氨基酸序列与 H T N 型同源性为765 % ~773 % ,与 S E O 型为932 % ~973 % 。核苷酸序列及其系统发生树分析表明: S E O 型的毒株间 M 片段全基因序列同源性较高为947 % ~995 % ,而 Gou3 株与 S E O 型毒株间差异高达154 % ~161 % , G2 展开更多

For the first time, we have utilized high-performance liquid chromatography (HPLC) to simultaneously quantify the eugenol and bancroffione in Caryophylli Fructus. The optimized parameters included: Inertsil ODS-4 column (150 mm×4.6 mm, 5 μm); column temperature: 35 ℃; mobile phase: methanol water (65:35, v/v); flow rate: 1.0 mL/min; detection wavelength: 280 nm. Eugenol and bancroftione showed good linear relationships with peak areas within the range of (0.0998 0.8982) mg/mL (r = 0.9999) and (0.1474-1.3266) mg/mL (r = 0.9999), respectively. The average recoveries were 102.52% and 100.96% for eugenol and bancroftione, respectively. Our results showed that the established method is simple, rapid, and accurate with good reproducibility to evaluate the quality of Caryophylli Fructus. 展开更多

初步探讨枸杞水溶性多糖(Lycii Fructus water-soluble polysaccharide,LFWP)的组成含量及其主链单糖组成特征,并进一步评估其对狭小空间环境应激引发小鼠肠道炎症的干预作用。采用部分酸水解、1-苯基-3甲基-5-吡唑酮(1-phenyl-3-methyl... 初步探讨枸杞水溶性多糖(Lycii Fructus water-soluble polysaccharide,LFWP)的组成含量及其主链单糖组成特征,并进一步评估其对狭小空间环境应激引发小鼠肠道炎症的干预作用。采用部分酸水解、1-苯基-3甲基-5-吡唑酮(1-phenyl-3-methyl-5-pyrazolone,PMP)柱前衍生化、高效凝胶渗透色谱法、傅里叶红外光谱和扫描电子显微镜等方法分析LFWP的组成和结构特征;HE染色和ELISA法评价LFWP对狭小空间应激引发小鼠肠道炎症的干预效果。组成结构研究表明,LFWP糖含量为90.5 mg/g,糖醛酸含量为10.7 mg/g,蛋白含量为0.4 mg/g,存在一个分布均一的主链部分,以半乳糖为主要单糖。药理活性研究表明,狭小空间环境造成小鼠结肠组织受损和炎性细胞浸润;表现为小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、IL-6含量增加(P<0.001),IL-10含量降低(P<0.001);结肠组织中脂多糖(lipopolysaccharide,LPS)、TNF-α、IL-1β、IL-6含量增加(P<0.001,P<0.05,P<0.01),钙卫蛋白(calprotectin,CALP)水平和髓过氧化物酶(myeloperoxidase,MPO)活性增加(P<0.05,P<0,01),IL-10含量降低(P<0.001),说明长期狭小空间环境应激引发小鼠肠道炎症。LFWP高剂量组干预明显降低小鼠血清TNF-α、IL-1β和IL-6水平(P<0.01,P<0.001),升高IL-10水平(P<0.001);同时明显降低小鼠结肠组织中LPS、TNF-α、IL-1β和IL-6水平(P<0.001),升高IL-10(P<0.05)水平,使结肠中CALP(P<0.001)水平和MPO(P<0.01)活性趋于正常。LFPW干预可显著改善狭小空间环境对小鼠肠道的应激损伤。 展开更多

目的:通过网络药理学与分子对接技术探究高良姜等三味山姜属中药治疗胃溃疡寒证的作用机制,并进行实验验证。方法:通过网络药理学常用技术手段,使用TCMSP、CNKI等数据库筛选三味山姜属中药中的活性成分及治疗胃溃疡疾病的相关靶点,并绘... 目的:通过网络药理学与分子对接技术探究高良姜等三味山姜属中药治疗胃溃疡寒证的作用机制,并进行实验验证。方法:通过网络药理学常用技术手段,使用TCMSP、CNKI等数据库筛选三味山姜属中药中的活性成分及治疗胃溃疡疾病的相关靶点,并绘制Venn图、蛋白质网络互作图。对关键靶点进行通路富集分析,将所获得的“药物-活性成分-靶点-通路-疾病”等数据导入Cytoscape 3.10.2进行可视化处理,并选择Degree排名靠前的化合物通过AutoDock软件进行分子对接。72只SPF级SD大鼠分为9组,包括空白对照组、模型组、阳性对照组(干姜10.8 g/kg)、高良姜高、低(10.8和5.4 g/kg)剂量组,大高良姜高、低(9.0和4.5 g/kg)剂量组,红豆蔻高、低(10.8和5.4 g/kg)剂量组,采用冰乙酸和冰知母法复制胃溃疡寒证模型,按不同剂量水煎液灌胃,1 mL/100 g体质量,每天2次,连续4 d。通过计算大鼠溃疡指数及溃疡抑制率,HE染色法观察大鼠胃组织病理学变化,ELISA法检测胃组织中AKT1、MAP2K1、m TOR蛋白的表达,从而与网络药理学的相关结果相互印证。结果:将三味山姜属中药所有成分去重后共筛选出关键成分45个,与疾病交集靶点共124个,蛋白质网络互作图共包含124个节点,923条边,京都基因与基因组百科全书(KEGG)富集共得到138条信号通路。分子对接分数均<-7.0 kcal/mol,即核心成分有效作用于核心靶点。实验验证结果显示:与模型组比较,三味山姜属中药不同剂量组对冰知母-冰乙酸法复制造成的胃溃疡寒证与胃黏膜水肿、充血等症状显著改善,溃疡面及出血点明显减少。与模型组比较,除红豆蔻低剂量组外,其他给药组AKT1蛋白表达水平均显著降低(P<0.05或P<0.01);除大高良姜低剂量组外,其余给药组MAP2K1蛋白表达水平均显著降低(P<0.05或P<0.01);除大高良姜和红豆蔻低剂量组外,各给药组mTOR蛋白表达水平均显著降低(P<0.05或P<0.01)。结论:三味山姜属中药可能通过多成分、多靶点、多通路的网络调控发挥抗炎、调控细胞增殖等作用来治疗胃溃疡。 展开更多
