Plant-parasitic nematodes(PPNs)(Meloidogyne sp.,Globodera sp.,and Pratylenchus sp.)and fungi are two of the most economically important groups of organisms affecting agricultural productivity worldwide.The interaction...Plant-parasitic nematodes(PPNs)(Meloidogyne sp.,Globodera sp.,and Pratylenchus sp.)and fungi are two of the most economically important groups of organisms affecting agricultural productivity worldwide.The interactions among PPNs,biocontrol fungi,and soil ecosystem can significantly impact plant health,disease management,and ecosystem functioning.We aimed to provide a comprehensive overview of the complex relationships between PPNs and biocontrol fungi,including pathogenic and biocontrol interactions.We summarized the molecular and ecological mechanisms underlying these interactions,highlighting the key players,signaling pathways,and environmental factors that influence the interactions.We also reviewed current knowledge on fungus-based biocontrol strategies against PPNs,including the development of novel management approaches.Furthermore,we explored the prospects of nematode-fungus interactions in agriculture,including the potential applications and technologies,precision agriculture,and integrated pest management approaches.This review highlights the need for further research on nematode-fungus interactions and their impact on plant infection and productivity,with an emphasis on the development of sustainable and effective strategies for managing PPNs and enhancing plant health in agricultural ecosystems.展开更多

Cucumber is one of the most important vegetables and economic crops in the world.The occurrence of fungal diseases in cucumbers seriously threatens the safety of cucumber production,with powdery mildew being one of th...Cucumber is one of the most important vegetables and economic crops in the world.The occurrence of fungal diseases in cucumbers seriously threatens the safety of cucumber production,with powdery mildew being one of the most common fungal diseases.With the rapid development of computer technology,more and more deep learning algorithms are being applied to identify powdery mildew fungus.However,existing algorithms suffer from low accuracy in recognizing small and occluded targets,as well as insufficient localization precision.To address this issue,the parallelized patch-aware attention(PPA)module was firstly introduced into the backbone network of YOLO v8s.By employing a parallel multi-branch structure and attention mechanism,it effectively captured multi-scale features of small targets,preserved critical information during multiple downsampling processes,and enhanced the performance of small target detection.Additionally,the global-to-local spatial aggregation(GLSA)module was introduced into the neck,which combined global contextual information with local detail features,significantly improving the model’s feature representation capability.This module enhanced the detection performance for small targets and complex scenes by better capturing multi-scale features.Experimental results showed that PG-YOLO v8s significantly improved powdery mildew fungus detection performance compared with YOLO v8s.The network achieved high precision in detecting powdery mildew fungus,with notable improvements in the detection accuracy of small and occluded targets.The research result can provide a high-throughput method for detecting powdery mildew fungus,enabling precise early detection and guiding early intelligent decision-making in cucumber production.This approach can help to improve disease control efficiency,ensure cucumber yield and quality,and it was of great significance for the sustainable development of agricultural production.展开更多

This study identified six novel azaphilones,isochromophilones G-L(1-6),and three novel biosynthetically related congeners(7-9)from Diaporthe sp.SCSIO 41011.The structures and absolute configurations were elucidated th...This study identified six novel azaphilones,isochromophilones G-L(1-6),and three novel biosynthetically related congeners(7-9)from Diaporthe sp.SCSIO 41011.The structures and absolute configurations were elucidated through comprehensive spectroscopic analyses combined with experimental and calculated electronic circular dichroism(ECD)spectra.Significantly,three highly oxygenated azaphilones contain an acetyl group at the terminal chain(4)or linear conjugated polyenoid moieties(5 and 6),which occur infrequently in the azaphilone family.Additionally,several compounds demonstrated inhibition of lipopolysaccharide(LPS)-induced nuclear factor kappa-B(NF-κB)activation in RAW 264.7 macrophages at 20μmol·L^(-1).The novel compound(1)effectively inhibited receptor activator of NF-κB ligand(RANKL)-induced osteoclast differentiation without exhibiting cytotoxicity in bone marrow and RAW264.7 macrophages,indicating its potential as a promising lead compound for osteolytic disease treatment.This research presents the first documented evidence of azaphilone derivatives as inhibitors of RANKL-induced osteoclastogenesis.展开更多
The Elongator complex is conserved in a wide range of species and plays crucial roles in diverse cellular processes.We have previously shown that the Elongator protein PoElp3 was involved in the asexual development,pa...The Elongator complex is conserved in a wide range of species and plays crucial roles in diverse cellular processes.We have previously shown that the Elongator protein PoElp3 was involved in the asexual development,pathogenicity,and autophagy of the rice blast fungus.In this study,we further revealed that PoElp3 functions via tRNA-mediated protein integrity.Phenotypic analyses revealed that overexpression of two of the tRNAs,tK(UUU)and tQ(UUG)could rescue the defects inΔPoelp3 strain.TMT-based proteomic and transcriptional analyses demonstrated that 386 proteins were down-regulated inΔPoelp3 strain compared with wild type strain Guy11,in a transcription-independent manner.Codon usage assays revealed an enrichment of Glutamine CAA-biased mRNA in the 386 proteins compared with the 70-15 genome.In addition to those reported previously,we also found that PoErp9,a sphingolipid C9-methyltransferase,was down-regulated in theΔPoelp3strain.Through an ILV2-specific integration of PoERP9-GFP into the wild type andΔPoelp3 strain,we were able to show that PoErp9 was positively regulated by PoElp3 translationally but not transcriptionally.Functional analyses revealed that PoErp9 was involved in the fungal growth,conidial development,pathogenicity,and TORrelated autophagy homeostasis in Pyricularia oryzae.Taken together,our results suggested that PoElp3 acts through the tRNA-mediated translational efficiency to regulate asexual development,pathogenicity,sphingolipid metabolism,and autophagy in the rice blast fungus.展开更多

Among them, 1 was a harziane diterpene, 2 was a sesquiterpene alcohol, and 3 was a polyketide with a completely symmetric configuration. Experimental results showed that the pure culture of G. umbellata was unable to form sclerotia, whereas sclerotia produced easily in flasks or on trunks when the companion fungus was inoculated together with strain of G. umbellata. The companion fungus is critical for sclerotium formation from hyphae of G. umbellata. Morphological differences were found in cultures of the two fungi. The companion fungus possesses thin-walled narrow hyphae, while G. umbellata is of wider hyphae which are either thick- or thin-walled.展开更多
To investigate the growth conditions of white-rot fungus and simulate its metabolism kinetic models, the rules how the factors such as biomass, culture fluid, pH value, glucose consumption and exopolysaccharides gener...To investigate the growth conditions of white-rot fungus and simulate its metabolism kinetic models, the rules how the factors such as biomass, culture fluid, pH value, glucose consumption and exopolysaccharides generation, etc., changed during the batch culture process of white-rot fungi by using an air-lift fermenter, as well as metabolic kinetics of white-rot fungi were studied. Based on Logistic equation, Luedeking-Piret equation and experimental data, the correlation model parameters of mycelia biomass, glucose consumption and exopolysaccharide generation were obtained and found to be change with time in metabolism process. Detailedly, μm=0.071 8 h-1,α= 0.831 8 g/(g·h), β= 0.002 g/(g·h), b1=0.016 3 g/(g·h) and b2=3.023 3 g/(g·h). Hence the mycelial growth kinetic model, exopolysaccharide generation kinetic model and substrate consumption kinetic model which describe fermentation process of white-rot fungi were established. Meanwhile, the experimental data were verified by this model, and a good fitting result with an average relative error less than 10% between the data obtained from experiments and the model was yielded. The results show that these models can predict the growth and metabolic rules of white-rot fungus, the fermentation process of exopolysaccharides and the kinetic mechanism of white-rot fungus accurately.展开更多

[Method] The pot experiment was used to assess the effects of different inoculated proportion of arbuscular mycorrhizal fungi (AMF) and Phosphate-sotubilizing fungus. Apophysomyces spartina, on growth, chlorophyll contents, P-uptake of castor bean (Ricinus communis L.) and rhizosphere soil pH values, available P concentrations, enzyme activities. [Result] The mixed inoculation of AMF and A. spartina significantly reduced soil pH value, increased soil available phosphorous contents, improved the activities of soil invertase, urease, neutral phosphatase, and alkaline phosphatase. Chlorophyll contents, P-uptake, and plant dry weight of castor bean were also in- creased. The optimal proportion of the number of AMF spores to A. spartina colonies was 28.56:11.5×10^5, which had positive effects on saline soil and could stimulate plant growth under greenhouse condition. [Conclusion] Appropriate propor- tion of AMF and A. spartina had the potential to enhance coastal saline soil prop- erty and promote castor bean growth.展开更多

[Method] Black fungi were cultivated in forest with middle-high altitude of 500-1 600 m. The germination time of fungus stick, germination days of buds, growth days of fruiting body, commodity exterior, marketing effect and biological efficiency of black fungi were investigated. [Result] Compared with traditional cultivation, black fungus products cultivated under forest had the taste and flavor of wild black fungus. Fungus chaff wastage was directly decomposed by the roots in forest land, improving the soil structure of forest land. The need of moisture retention and the frequent access of administrative staff reduced the probability of forest fire occurrence. [Conclusion] Cultivation of black fungus under forest saves more than 70% fixed-asset investment on fungus shed, and the biological efficiency reached 12.2%.展开更多

Shenxianggen No.4) were resulted from inoculation with two different virulent races of rice blast fungus ( Magnaporthe grisea (Hebert) Barr), and thus an effective infecting system was established between rice plants and the rice blast pathogen. Two cDNA clones that showed induced and temporal patterns in expression in the very early stage in response to infection of the fungus were obtained from the plants by use of differential display. Of the two cDNA clones, Fastresp_a was induced to express in both compatible and incompatible interactions although it was expressed earlier in the former reaction. The second one, Fastresp_b , was only expressed in incompatible interaction. Southern blot analysis of the rice genomic DNA indicated that both of the two clones were from genome of the plant. No significant homology to the two genes was found from the rice gene database. This suggested that they were novel genes in rice and may play important roles in rice resistant response to infection of rice blast fungus.展开更多

文摘This study identified six novel azaphilones,isochromophilones G-L(1-6),and three novel biosynthetically related congeners(7-9)from Diaporthe sp.SCSIO 41011.The structures and absolute configurations were elucidated through comprehensive spectroscopic analyses combined with experimental and calculated electronic circular dichroism(ECD)spectra.Significantly,three highly oxygenated azaphilones contain an acetyl group at the terminal chain(4)or linear conjugated polyenoid moieties(5 and 6),which occur infrequently in the azaphilone family.Additionally,several compounds demonstrated inhibition of lipopolysaccharide(LPS)-induced nuclear factor kappa-B(NF-κB)activation in RAW 264.7 macrophages at 20μmol·L^(-1).The novel compound(1)effectively inhibited receptor activator of NF-κB ligand(RANKL)-induced osteoclast differentiation without exhibiting cytotoxicity in bone marrow and RAW264.7 macrophages,indicating its potential as a promising lead compound for osteolytic disease treatment.This research presents the first documented evidence of azaphilone derivatives as inhibitors of RANKL-induced osteoclastogenesis.

文摘The Elongator complex is conserved in a wide range of species and plays crucial roles in diverse cellular processes.We have previously shown that the Elongator protein PoElp3 was involved in the asexual development,pathogenicity,and autophagy of the rice blast fungus.In this study,we further revealed that PoElp3 functions via tRNA-mediated protein integrity.Phenotypic analyses revealed that overexpression of two of the tRNAs,tK(UUU)and tQ(UUG)could rescue the defects inΔPoelp3 strain.TMT-based proteomic and transcriptional analyses demonstrated that 386 proteins were down-regulated inΔPoelp3 strain compared with wild type strain Guy11,in a transcription-independent manner.Codon usage assays revealed an enrichment of Glutamine CAA-biased mRNA in the 386 proteins compared with the 70-15 genome.In addition to those reported previously,we also found that PoErp9,a sphingolipid C9-methyltransferase,was down-regulated in theΔPoelp3strain.Through an ILV2-specific integration of PoERP9-GFP into the wild type andΔPoelp3 strain,we were able to show that PoErp9 was positively regulated by PoElp3 translationally but not transcriptionally.Functional analyses revealed that PoErp9 was involved in the fungal growth,conidial development,pathogenicity,and TORrelated autophagy homeostasis in Pyricularia oryzae.Taken together,our results suggested that PoElp3 acts through the tRNA-mediated translational efficiency to regulate asexual development,pathogenicity,sphingolipid metabolism,and autophagy in the rice blast fungus.

文摘Three known compounds were isolated from the endophytic fungus Trichoderma sp Xy24 from a mangrove plant Xylocarpus granatum by using various column chromatography techniques. Their structures were identified as harzianone (1), trichoacorenol (2), and trichodimerol (3) by extensive spectroscopic analysis. Among them, 1 was a harziane diterpene, 2 was a sesquiterpene alcohol, and 3 was a polyketide with a completely symmetric configuration. Compound 3 exhibited medium inhibitory activity with an IC50 value of 74.6 μM using a NA (H7N9)/MUNANA model.
文摘The companion fungus ( Grifola sp.) related to sclerotial formation from hyphae of Grifola umbellata (Pers.) Pilat was isolated from the cavity associated with sclerotia of G. umbellata in natural condition. Experimental results showed that the pure culture of G. umbellata was unable to form sclerotia, whereas sclerotia produced easily in flasks or on trunks when the companion fungus was inoculated together with strain of G. umbellata. The companion fungus is critical for sclerotium formation from hyphae of G. umbellata. Morphological differences were found in cultures of the two fungi. The companion fungus possesses thin-walled narrow hyphae, while G. umbellata is of wider hyphae which are either thick- or thin-walled.

文摘To investigate the growth conditions of white-rot fungus and simulate its metabolism kinetic models, the rules how the factors such as biomass, culture fluid, pH value, glucose consumption and exopolysaccharides generation, etc., changed during the batch culture process of white-rot fungi by using an air-lift fermenter, as well as metabolic kinetics of white-rot fungi were studied. Based on Logistic equation, Luedeking-Piret equation and experimental data, the correlation model parameters of mycelia biomass, glucose consumption and exopolysaccharide generation were obtained and found to be change with time in metabolism process. Detailedly, μm=0.071 8 h-1,α= 0.831 8 g/(g·h), β= 0.002 g/(g·h), b1=0.016 3 g/(g·h) and b2=3.023 3 g/(g·h). Hence the mycelial growth kinetic model, exopolysaccharide generation kinetic model and substrate consumption kinetic model which describe fermentation process of white-rot fungi were established. Meanwhile, the experimental data were verified by this model, and a good fitting result with an average relative error less than 10% between the data obtained from experiments and the model was yielded. The results show that these models can predict the growth and metabolic rules of white-rot fungus, the fermentation process of exopolysaccharides and the kinetic mechanism of white-rot fungus accurately.

文摘Chemical examination of an unidentified sponge associated marine fungus Penicillium terrestre resulted in the isolation of eight compounds. Their structures are identified as cyclo (glycyl-D-proline) (1), cyclo (L-prolyl-L-alanine) (2), cyclo (L-prolyl- D-alanine) (3), cyclo (L-leucyl-trans-4-hydroxy-L-proline) (4), cyclo (L-leucyl-cis-4-hydroxy-D-proline) (5), cyclo (L-trans-(4- hydroxyprolinyl)-L-phenylalanine) (6), hexylitaconic acid (7), and hexylitaconic methylate (8), based on spectroscopic analysis (1D, 2D NMR). Compounds 7, 8 were obtained from the genus Penicillium for the first time, while compounds 1-6 were isolated from the fungus Penicillium terrestre for the first time.

文摘[Objective] In order to explore the mechanism of combined inoculation mi- croorganisms in improving coastal saline soil property and plant growth. [Method] The pot experiment was used to assess the effects of different inoculated proportion of arbuscular mycorrhizal fungi (AMF) and Phosphate-sotubilizing fungus. Apophysomyces spartina, on growth, chlorophyll contents, P-uptake of castor bean (Ricinus communis L.) and rhizosphere soil pH values, available P concentrations, enzyme activities. [Result] The mixed inoculation of AMF and A. spartina significantly reduced soil pH value, increased soil available phosphorous contents, improved the activities of soil invertase, urease, neutral phosphatase, and alkaline phosphatase. Chlorophyll contents, P-uptake, and plant dry weight of castor bean were also in- creased. The optimal proportion of the number of AMF spores to A. spartina colonies was 28.56:11.5×10^5, which had positive effects on saline soil and could stimulate plant growth under greenhouse condition. [Conclusion] Appropriate propor- tion of AMF and A. spartina had the potential to enhance coastal saline soil prop- erty and promote castor bean growth.

文摘[Objective] The study was to solve the contradiction between fungus and forest during the production of black fungi, and to obtain relatively high economic benefits and ecological benefits. [Method] Black fungi were cultivated in forest with middle-high altitude of 500-1 600 m. The germination time of fungus stick, germination days of buds, growth days of fruiting body, commodity exterior, marketing effect and biological efficiency of black fungi were investigated. [Result] Compared with traditional cultivation, black fungus products cultivated under forest had the taste and flavor of wild black fungus. Fungus chaff wastage was directly decomposed by the roots in forest land, improving the soil structure of forest land. The need of moisture retention and the frequent access of administrative staff reduced the probability of forest fire occurrence. [Conclusion] Cultivation of black fungus under forest saves more than 70% fixed-asset investment on fungus shed, and the biological efficiency reached 12.2%.
文摘Two metabolites (A and B) were isolated from the mycelium of mangrove endophytic fungus Stysanus like sp. (#2492) from the South China Sea. Their structures were identified by spectral data as N-(2-hydroxytetracosyl)-2-amino-1,3,4-trihydroxyoctadecane (A) and γ -stearolactone (B). It is the first report that γ -stearolactone (B) is isolated from marine fungus as natural product.

文摘The sponge-associated fungus Nectria sp.HLS206,which belongs to the order of Hypocreales,was isolated from the marine sponge Gelliodes carnosa collected from the South China Sea.The secondary metabolites of the fungus were isolated.From the solid cultures,five compounds were purified,which are chalmicrin(1),hypocrealesate(2),fucosterol(3),stigmasterol(4) and β-sitosterol(5).The structures of the 5 compounds were elucidated by ESI-MS,1D and 2D NMR spectra(COSY,HMQC and HMBC).To the best of our knowledge,it is the first time compound 1 was isolated from the order of Hypocreales,and the spectral data of compound 2 are also firstly reported.
文摘Compatible and incompatible reactions in rice plants ( Oryza sativa L. cv. Shenxianggen No.4) were resulted from inoculation with two different virulent races of rice blast fungus ( Magnaporthe grisea (Hebert) Barr), and thus an effective infecting system was established between rice plants and the rice blast pathogen. Two cDNA clones that showed induced and temporal patterns in expression in the very early stage in response to infection of the fungus were obtained from the plants by use of differential display. Of the two cDNA clones, Fastresp_a was induced to express in both compatible and incompatible interactions although it was expressed earlier in the former reaction. The second one, Fastresp_b , was only expressed in incompatible interaction. Southern blot analysis of the rice genomic DNA indicated that both of the two clones were from genome of the plant. No significant homology to the two genes was found from the rice gene database. This suggested that they were novel genes in rice and may play important roles in rice resistant response to infection of rice blast fungus.

文摘A new abietane diterpene 12-hydroxy-7-oxo-5, 8, 11, 13-tetraene-18, 6-abietanolide, together with a new natural sterol stigmasta-7, 22-diene-3β, 5α 6α-triol have been isolated from the fruiting body of the fungus Phellinus igniarius. Their structures were elucidated by spectroscopic methods including 2D NMR techniques.
