Bioactive peptides and proteins(BAPPs)are promising therapeutic agents for tissue repair with considerable advantages,including multifunctionality,specificity,biocompatibility,and biodegradability.However,the high com...Bioactive peptides and proteins(BAPPs)are promising therapeutic agents for tissue repair with considerable advantages,including multifunctionality,specificity,biocompatibility,and biodegradability.However,the high complexity of tissue microenvironments and their inherent deficiencies such as short half-live and susceptibility to enzymatic degradation,adversely affect their therapeutic efficacy and clinical applications.Investigating the fundamental mechanisms by which BAPPs modulate the microenvironment and developing rational delivery strategies are essential for optimizing their administration in distinct tissue repairs and facilitating clinical translation.This review initially focuses on the mechanisms through which BAPPs influence the microenvironment for tissue repair via reactive oxygen species,blood and lymphatic vessels,immune cells,and repair cells.Then,a variety of delivery platforms,including scaffolds and hydrogels,electrospun fibers,surface coatings,assisted particles,nanotubes,two-dimensional nanomaterials,and nanoparticles engineered cells,are summarized to incorporate BAPPs for effective tissue repair,modification strategies aimed at enhancing loading efficiencies and release kinetics are also reviewed.Additionally,the delivery of BAPPs can be precisely regulated by endogenous stimuli(glucose,reactive oxygen species,enzymes,pH)or exogenous stimuli(ultrasound,heat,light,magnetic field,and electric field)to achieve on-demand release tailored for specific tissue repair needs.Furthermore,this review focuses on the clinical potential of BAPPs in facilitating tissue repair across various types,including bone,cartilage,intervertebral discs,muscle,tendons,periodontal tissues,skin,myocardium,nervous system(encompassing brain,spinal cord,and peripheral nerve),endometrium,as well as ear and ocular tissue.Finally,current challenges and prospects are discussed.展开更多

The introduction of neurotrophic factors into injured peripheral nerve sites is beneficial to peripheral nerve regeneration.However,neurotrophic facto rs are rapidly degraded in vivo and obstruct axonal regeneration w...The introduction of neurotrophic factors into injured peripheral nerve sites is beneficial to peripheral nerve regeneration.However,neurotrophic facto rs are rapidly degraded in vivo and obstruct axonal regeneration when used at a supraphysiological dose,which limits their clinical benefits.Bioactive mimetic peptides have been developed to be used in place of neurotrophic factors because they have a similar mode of action to the original growth fa ctors and can activate the equivalent receptors but have simplified sequences and structures.In this study,we created polydopamine-modified chitin conduits loaded with brain-derived neurotrophic factor mimetic peptides and vascular endothelial growth fa ctor mimetic peptides(Chi/PDA-Ps).We found that the Chi/PDA-Ps conduits were less cytotoxic in vitro than chitin conduits alone and provided sustained release of functional peptides.In this study,we evaluated the biocompatibility of the Chi/P DA-Ps conduits.Brain-derived neurotrophic factor mimetic peptide and vascular endothelial growth fa ctor mimetic peptide synergistically promoted prolife ration of Schwann cells and secretion of neurotrophic factors by Schwann cells and attachment and migration of endothelial cells in vitro.The Chi/P DA-Ps conduits were used to bridge a 2 mm gap between the nerve stumps in rat models of sciatic nerve injury.We found that the application of Chi/PDA-Ps conduits could improve the motor function of rats and reduce gastrocnemius atrophy.The electrophysiological results and the microstructure of regenerative nerves showed that the nerve conduction function and re myelination was further resto red.These findings suggest that the Chi/PDA-Ps conduits have great potential in peripheral nerve injury repair.展开更多

[ Method ] The dialysis bag of 8.0, 3.5 and 1.0 kD were used to classify the hydrolyzate derived from alcalase and flavourzyme, peanut bioactive poptides of differ-ent molecular weight were obtained and then their scavenging capacity of free radicals was measured. The molecular weight composition was stud-ied by Tricine-SDS-PAGE.[Result] The content of peptides 〈 1.0 kD were 76.21% and 83.42% in the total hydrolyzate from alcalase and fla-vourzyme respectively. All hydrolyzate with different molecular grades showed free radical scavenging capacity, which was increased with the reduc- tion of molecular mass. The peptides 〈 1.0 kD exhibited higher radical scavenging capacity of (87.41 ±0.66) % (alcalase) and (67.88 ±0.48)% ( flavourzyme), respectively. [ Conclusion] Peanut bioactive peptides had strong effect of antioxidant capacity, especially that 〈 1.0 kD, which had great prospect.展开更多
The present study here establishes a complete and effective method for isolating,purifying and identifying extracellular and intracellular peptides,and also describes the characters and bioactivities of peptides from ...The present study here establishes a complete and effective method for isolating,purifying and identifying extracellular and intracellular peptides,and also describes the characters and bioactivities of peptides from fermented casein with Lactobacillus helveticus.Intracellular peptides are much larger in quantity and more complex in composition than extracellular peptides,between which the correlation reveals proteolytic and metabolic mechanisms.In addition,totally 241 different peptide sequences were identified by Nano LC–MS/MS from casein(212)and Lactobacillus helveticus proteins(29).These casein-derived peptides mostly originated from-casein,followed byS1-casein,-casein,andS2-casein,and came from extracell(69)and intracell(143),in which common peptides have a total of 27.Forty-four of the identified peptides were previously described as bioactive,including angiotensinconverting enzyme(ACE)-inhibitory,antioxidant,immunomodulating,antimicrobial,DPP-IV inhibitory,antiamnesic and anticancer effects and so on.Thirteen peptides with the potential of some biological activities are obtained,which were described in previous studies.A total of 47 novel peptides of 5 to 26 amino acids that were not disclosed were obtained.The new sources of natural bioactive peptides may have the very high application value as potential new peptide drugs for treatment human diseases.The product peptide DELQDKIHPF found in both extracell and intracell was quantitatively analyzed using the MRM mode of UPLC-U3Q,23.1 and 9.76 ng/mL,respectively.The quantitative analysis of the potential bioactive peptide may also advance the production of peptide products in the future.展开更多

Peptides in the hydrolysate produced by the proteases from strains HN2 -3 and N13d had higher angiotensin I-converting-enzyme (ACE)-inhibitory activity. The two marine yeast strains were identified to be Aureobasidium pullulans according to the results of routine yeast identification and molecular methods. After purification of the proteases from the two marine yeast strains, it was found that the optimal pH for them was both 9.0, both of them were serine alkaline protease. However, the optimal temperature for the protease from the strain HN2 -3 was 52℃ while that from strain N13d was 48℃. ACE-inhibitory activity of the peptides in the hydrolysate of shrimp protein produced by the purified protease from the strain HN2 -3 was the highest while antioxidant activity in the hydrolysate of spirulina protein produced by the purified protease from the strain N13d was the highest.展开更多

The changes in growth performance, body composition, n...The present study was conducted to evaluate the feasibility of replacing fish meal(FM) with bioactive peptides(BPs) in diet of white shrimp(Litopenaeus vannamei). The changes in growth performance, body composition, non-specific immunity, and water quality were examined after the shrimp were fed four diets, in which 0%(control), 33.3%, 66.7% and 100% of FM was replaced by BPs, respectively. The groups were designated as Con, 1/3BPs, 2/3BPs, and 3/3BPs. A total of 720 shrimp with an initial body weight of 1.46 ± 0.78 g were fed the experimental diets for 56 days. The results revealed that: 1) the weight gain rate(WGR) in 1/3BPs, 2/3BPs, and 3/3BPs was significantly higher than that in Con(P < 0.05), while no significant difference was found on survival rate and feed conversion ratio(FCR); 2) the whole-body crude protein(CP) and crude lipids(CL) were significantly different among groups, while there was no significant difference between crude ash and phosphorus contents; 3) the levels of acid phosphatase(ACP), lysozyme(LZM), superoxide dismutase(SOD), phenol oxidase(PO) and bactericidal activity increased significantly with the inclusion of BPs; 4) in terms of water quality, no significant difference was found in p H and dissolved oxygen among diets during the whole experimental period. Moreover, even though nitrite and ammonium levels tended to increase with time, there was no significant difference among groups. The results indicated that BPs is an applicable alternative of protein source, which can substitute FM in the diets of L. vannamei; it is able to effectively promote growth performance and improve immunity. Moreover, BPs in the diets had no negative impact on water quality.展开更多

Particularly, it is closest to human breast milk and has high levels of nutrients and bioactive components. The aim of this study was to explore the antioxidant peptides derived from protein fractions of camel milk. Camel milk proteins(CMP) were fractionated into camel casein protein(CCP) and camel whey protein(CWP), which were hydrolyzed with pepsin to produce peptic digests P-CCP and P-CWP, respectively. RP-HPLC was used for fractionation of the peptides from the P-CCP and P-CWP. The antioxidant activities were evaluated using superoxide anion generating system of xanthine oxidase(XOD)and 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging assay. Active peptides were analyzed using matrixassisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) whereas a number of antioxidant peptides, with masses ranging from 913 to 2,951 Da, derived mainly from alpha-casein, lactophorin and lactoferrin, were identified. When yeast cells are used as a system for modeling mitochondrial disease, the peptides in caseins and whey fractions significantly enhanced the tolerance of yeast cells against peroxide-induced oxidative stress. The results show that both caseins and whey proteins of camel milk possess bioactive peptides with significant radical-scavenging activities and thus herald a fascinating opportunity for their potential as nutraceuticals or therapeutic peptides for prevention and treatment of oxidative stress-associated diseases.展开更多
The objective of this study was to evaluate the changes on flavor and quality characteristics of goat yogurt during storage with the addition of dairy bioactive peptides and a mixture of lotus seed and lily bulb powde...The objective of this study was to evaluate the changes on flavor and quality characteristics of goat yogurt during storage with the addition of dairy bioactive peptides and a mixture of lotus seed and lily bulb powder.Both the addition of dairy bioactive peptides and lotus seeds/lily bulb powder resulted in a fermentation time reduction,water holding capacity(WHC)improvement,and inhibition of post-acidification of goat yogurt during storage.A total of 50 volatile flavor components were detected in the goat milk yogurt samples,and the addition of the above two supplements were shown to be effective in improving the flavor and the textural properties of the goat milk yogurt during storage.The group added with lotus seeds/lily bulb powder showed the best WHC and textural properties,followed by the group added with dairy bioactive peptides alone and the group with both added.The addition of both supplements was found to be useful in making up for the two defects of goat yogurt,prolonged fermentation and soft curds.Therefore,dairy bioactive peptides and lotus seeds/lily bulb powder are recommended to be added to goat yogurt as nutritional supplements.展开更多

Bioactive peptides are unique,low molecular weight peptide sequences generally consisting of 2–20 amino acid residues.These peptide sequences are inactive within the parent protein but they become physiologically act...Bioactive peptides are unique,low molecular weight peptide sequences generally consisting of 2–20 amino acid residues.These peptide sequences are inactive within the parent protein but they become physiologically active once released from the native polypeptide sequence via hydrolysis.There are multiple methods for producing bioactive peptides from precursor protein molecules,with microbial fermentation of various dietary matrices indubitably being a novel method to produce peptides with specialized bioactivity.Fermented foods especially fermented dairy products,legumes,cereals,meat and marine life as a source of bioactive peptides have been well documented.These peptides have gained scientific attention owing to their biofunctional attributes.The food-derived bioactive peptides have the potential to serve as valuable ingredients in functional foods and nutraceutical products to promote health.Bioactive peptides are known to possess various health-promoting properties including anti-carcinogenic,anti-hypertensive,anti-microbial,antioxidant,anti-diabetic,and immunomodulatory effects.The COVID-19 pandemic has put the world’s health,economy,and social stability in jeopardy.The SARS-CoV-2 infection contributes to severe conditions and higher mortality in COVID-19 patients with comorbidities.The viral infection not only causes severe respiratory infection but also causes malfunctioning of the Renin-Angiotensin system(RAS),resulting in the downregulation of Angiotensin-converting enzyme Ⅱ(ACE-Ⅱ)and subsequent accumulation of Angiotensin Ⅱ.Several synthetic ACE inhibitory medications are being used to minimize the severity of Angiotensin Ⅱ adverse effects such as hypertension.The growing concern about the side effects associated with these pharmaceuticals has prompted researchers to look for alternatives in the form of foods and nutraceuticals with health-promoting features.Biologically active peptides have the potential to be used as a new-generation pharmaceutical product for various diseases including COVID-19.The multi-functional food-derived peptides could be a promising approach against COVID-19 infection in patients with chronic complications through their therapeutic actions.However,more in vitro and in vivo studies are required to validate their efficacy in enhancing the survivability and viability of COVID-19 patients.Although many peptides have demonstrated their positive effects via biochemical assays,cell culture,and animal models,the translation of these findings into practical application is limited.This might be related to the bioavailability issues,which influence the correlation of in vitro results with in vivo functions of peptides.To exert a health-promoting impact,these peptides need to withstand severe gastrointestinal conditions and the action of digestive enzymes to reach the target site in an active state.Therefore it is critical to thoroughly investigate the gastrointestinal stability and transport of these biopeptides and devise strategies to improve their absorption and bioavailability.展开更多

文摘Bioactive peptides and proteins(BAPPs)are promising therapeutic agents for tissue repair with considerable advantages,including multifunctionality,specificity,biocompatibility,and biodegradability.However,the high complexity of tissue microenvironments and their inherent deficiencies such as short half-live and susceptibility to enzymatic degradation,adversely affect their therapeutic efficacy and clinical applications.Investigating the fundamental mechanisms by which BAPPs modulate the microenvironment and developing rational delivery strategies are essential for optimizing their administration in distinct tissue repairs and facilitating clinical translation.This review initially focuses on the mechanisms through which BAPPs influence the microenvironment for tissue repair via reactive oxygen species,blood and lymphatic vessels,immune cells,and repair cells.Then,a variety of delivery platforms,including scaffolds and hydrogels,electrospun fibers,surface coatings,assisted particles,nanotubes,two-dimensional nanomaterials,and nanoparticles engineered cells,are summarized to incorporate BAPPs for effective tissue repair,modification strategies aimed at enhancing loading efficiencies and release kinetics are also reviewed.Additionally,the delivery of BAPPs can be precisely regulated by endogenous stimuli(glucose,reactive oxygen species,enzymes,pH)or exogenous stimuli(ultrasound,heat,light,magnetic field,and electric field)to achieve on-demand release tailored for specific tissue repair needs.Furthermore,this review focuses on the clinical potential of BAPPs in facilitating tissue repair across various types,including bone,cartilage,intervertebral discs,muscle,tendons,periodontal tissues,skin,myocardium,nervous system(encompassing brain,spinal cord,and peripheral nerve),endometrium,as well as ear and ocular tissue.Finally,current challenges and prospects are discussed.

文摘Oxidative stress is caused by an imbalance of the antioxidant defense system and excessive production of free radicals,which can damage biological macromolecules such as proteins,lipids and nucleic acids,and can cause aging,ischemia-reperfusion injury,inflammatory injury,liver and kidney injury and other diseases.The nuclear factor erytheroid-2-related factor 2(Nrf2)is a major regulator of redox balance.The Nrf2 pathway also exerts a central function in cell apoptosis,oxidative stress damage and metabolic diseases.Bioactive peptides are small molecular peptides composed of amino acid residues.They have many biological activities and play important physiological roles in human body.Antioxidant peptide is a kind of peptide which can reduce oxidative stress damage.They are safe,non-toxic and easily absorbed.In this review,we summarized the mechanism of bioactive peptides in regulating oxidative stress,especially antioxidant peptides,through regulating the Nrf2 signaling pathway and the expressions of oxidative stress-related genes,to alleviate oxidative stress-induced damage.The paper will provide valuable reference to investigators in the antioxidant peptide field and also promote the applications of antioxidant peptides in the oxidative stress-associated diseases.
文摘The cosmetic sector is a multibillion-dollar industry that requires constant attention being paid to innovative product development and engagement.Notably,its market value is projected to exceed 750 billion U.S.dollars by 2025,and it is expanding as novel,climate-friendly,green,and sustainable components from natural sources are incorporated.This review is written based on the numerous reports on the potential applications of food-derived peptides while focusing on their possible uses in the formulation of cosmeceutical and skincare products.First,the production methods of bioactive peptides linked to cosmeceutical uses are described.Then,we discuss the obtainment and characterization of different anti-inflammatory,antimicrobial,antioxidant,anti-aging,and other pleiotropic peptides with their specific mechanisms,from various food sources.The review concludes with salient considerations of the cost of production and pilot scale operation,stability,compatibility,user safety,site-specificity,and delivery methods,when designing or developing biopeptide-based cosmeceutical products.

文摘Peptides are functional active fragments of proteins which can provide nutrients needed for human growth and development,and they also have unique physiological activity characteristics relative to proteins.Bioactive peptides contain a great deal of development potential.More specifically,food-derived bioactive peptides have the advantages of a wide variety of sources,unique structures,high efficiency and safety,so they have broad development prospects.This review provides an overview of the current advances regarding the preparation,functional characteristics,and structure–activity relationships of food-derived bioactive peptides.Moreover,the prospects for the future development and application of food-derived bioactive peptides are discussed.This review may provide a better understanding of foodderived bioactive peptides,and some constructive inspirations for further research and applications in the food industry.

文摘The introduction of neurotrophic factors into injured peripheral nerve sites is beneficial to peripheral nerve regeneration.However,neurotrophic facto rs are rapidly degraded in vivo and obstruct axonal regeneration when used at a supraphysiological dose,which limits their clinical benefits.Bioactive mimetic peptides have been developed to be used in place of neurotrophic factors because they have a similar mode of action to the original growth fa ctors and can activate the equivalent receptors but have simplified sequences and structures.In this study,we created polydopamine-modified chitin conduits loaded with brain-derived neurotrophic factor mimetic peptides and vascular endothelial growth fa ctor mimetic peptides(Chi/PDA-Ps).We found that the Chi/PDA-Ps conduits were less cytotoxic in vitro than chitin conduits alone and provided sustained release of functional peptides.In this study,we evaluated the biocompatibility of the Chi/P DA-Ps conduits.Brain-derived neurotrophic factor mimetic peptide and vascular endothelial growth fa ctor mimetic peptide synergistically promoted prolife ration of Schwann cells and secretion of neurotrophic factors by Schwann cells and attachment and migration of endothelial cells in vitro.The Chi/P DA-Ps conduits were used to bridge a 2 mm gap between the nerve stumps in rat models of sciatic nerve injury.We found that the application of Chi/PDA-Ps conduits could improve the motor function of rats and reduce gastrocnemius atrophy.The electrophysiological results and the microstructure of regenerative nerves showed that the nerve conduction function and re myelination was further resto red.These findings suggest that the Chi/PDA-Ps conduits have great potential in peripheral nerve injury repair.

文摘[ Objective] To prepare peanut bioactive peptides and analyze their molecular weight composition and antioxidant activity. [ Method ] The dialysis bag of 8.0, 3.5 and 1.0 kD were used to classify the hydrolyzate derived from alcalase and flavourzyme, peanut bioactive poptides of differ-ent molecular weight were obtained and then their scavenging capacity of free radicals was measured. The molecular weight composition was stud-ied by Tricine-SDS-PAGE.[Result] The content of peptides 〈 1.0 kD were 76.21% and 83.42% in the total hydrolyzate from alcalase and fla-vourzyme respectively. All hydrolyzate with different molecular grades showed free radical scavenging capacity, which was increased with the reduc- tion of molecular mass. The peptides 〈 1.0 kD exhibited higher radical scavenging capacity of (87.41 ±0.66) % (alcalase) and (67.88 ±0.48)% ( flavourzyme), respectively. [ Conclusion] Peanut bioactive peptides had strong effect of antioxidant capacity, especially that 〈 1.0 kD, which had great prospect.
文摘The present study here establishes a complete and effective method for isolating,purifying and identifying extracellular and intracellular peptides,and also describes the characters and bioactivities of peptides from fermented casein with Lactobacillus helveticus.Intracellular peptides are much larger in quantity and more complex in composition than extracellular peptides,between which the correlation reveals proteolytic and metabolic mechanisms.In addition,totally 241 different peptide sequences were identified by Nano LC–MS/MS from casein(212)and Lactobacillus helveticus proteins(29).These casein-derived peptides mostly originated from-casein,followed byS1-casein,-casein,andS2-casein,and came from extracell(69)and intracell(143),in which common peptides have a total of 27.Forty-four of the identified peptides were previously described as bioactive,including angiotensinconverting enzyme(ACE)-inhibitory,antioxidant,immunomodulating,antimicrobial,DPP-IV inhibitory,antiamnesic and anticancer effects and so on.Thirteen peptides with the potential of some biological activities are obtained,which were described in previous studies.A total of 47 novel peptides of 5 to 26 amino acids that were not disclosed were obtained.The new sources of natural bioactive peptides may have the very high application value as potential new peptide drugs for treatment human diseases.The product peptide DELQDKIHPF found in both extracell and intracell was quantitatively analyzed using the MRM mode of UPLC-U3Q,23.1 and 9.76 ng/mL,respectively.The quantitative analysis of the potential bioactive peptide may also advance the production of peptide products in the future.

文摘Over 400 yeast strains from seawater and sediments were obtained, but only five strains named HN2 -3, N13d, N13C, Mb5 and HN3 - 2 among them could form clear zones around their colonies on the double plates with 2.0% casein. Peptides in the hydrolysate produced by the proteases from strains HN2 -3 and N13d had higher angiotensin I-converting-enzyme (ACE)-inhibitory activity. The two marine yeast strains were identified to be Aureobasidium pullulans according to the results of routine yeast identification and molecular methods. After purification of the proteases from the two marine yeast strains, it was found that the optimal pH for them was both 9.0, both of them were serine alkaline protease. However, the optimal temperature for the protease from the strain HN2 -3 was 52℃ while that from strain N13d was 48℃. ACE-inhibitory activity of the peptides in the hydrolysate of shrimp protein produced by the purified protease from the strain HN2 -3 was the highest while antioxidant activity in the hydrolysate of spirulina protein produced by the purified protease from the strain N13d was the highest.

文摘The present study was conducted to evaluate the feasibility of replacing fish meal(FM) with bioactive peptides(BPs) in diet of white shrimp(Litopenaeus vannamei). The changes in growth performance, body composition, non-specific immunity, and water quality were examined after the shrimp were fed four diets, in which 0%(control), 33.3%, 66.7% and 100% of FM was replaced by BPs, respectively. The groups were designated as Con, 1/3BPs, 2/3BPs, and 3/3BPs. A total of 720 shrimp with an initial body weight of 1.46 ± 0.78 g were fed the experimental diets for 56 days. The results revealed that: 1) the weight gain rate(WGR) in 1/3BPs, 2/3BPs, and 3/3BPs was significantly higher than that in Con(P < 0.05), while no significant difference was found on survival rate and feed conversion ratio(FCR); 2) the whole-body crude protein(CP) and crude lipids(CL) were significantly different among groups, while there was no significant difference between crude ash and phosphorus contents; 3) the levels of acid phosphatase(ACP), lysozyme(LZM), superoxide dismutase(SOD), phenol oxidase(PO) and bactericidal activity increased significantly with the inclusion of BPs; 4) in terms of water quality, no significant difference was found in p H and dissolved oxygen among diets during the whole experimental period. Moreover, even though nitrite and ammonium levels tended to increase with time, there was no significant difference among groups. The results indicated that BPs is an applicable alternative of protein source, which can substitute FM in the diets of L. vannamei; it is able to effectively promote growth performance and improve immunity. Moreover, BPs in the diets had no negative impact on water quality.

文摘Bioactive peptides(BAPs),typically comprising two to 20 amino acids,are widely acknowledged for their distinctive biological functions.They can be broadly classified as endogenous or exogenous peptides.Integrating BAPs into feed is now deemed an efficacious nutritional strategy to enhance the resilience of farmed aquatic species against stress-induced damage and pathogen infections.Moreover,dietary BAPs have exhibited significantly favorable effects on fish and shrimp's feed utilization and growth performance.The present review comprehensively explores the extensive research on the diverse applications of BAPs in aquaculture,explicitly focusing on recent advances in utilizing antioxidative,antimicrobial,antiviral,anti-inflammatory and immunomodulatory peptides.Additionally,the study incorporates several common dipeptides and tripeptides to address the challenges associated with inefficient absorption,high production costs,and the inherent instability of amino acid monomers.Furthermore,the review explores the methods for obtaining BAPs through enzymatic hydrolysis technology,highlighting key challenges that require attention in aquatic research.This review is expected to encourage further investigation in the related fields.
文摘Camel milk is traditionally considered to have medicinal characteristics that it has potential health benefits and could help to treat several illnesses. Particularly, it is closest to human breast milk and has high levels of nutrients and bioactive components. The aim of this study was to explore the antioxidant peptides derived from protein fractions of camel milk. Camel milk proteins(CMP) were fractionated into camel casein protein(CCP) and camel whey protein(CWP), which were hydrolyzed with pepsin to produce peptic digests P-CCP and P-CWP, respectively. RP-HPLC was used for fractionation of the peptides from the P-CCP and P-CWP. The antioxidant activities were evaluated using superoxide anion generating system of xanthine oxidase(XOD)and 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging assay. Active peptides were analyzed using matrixassisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) whereas a number of antioxidant peptides, with masses ranging from 913 to 2,951 Da, derived mainly from alpha-casein, lactophorin and lactoferrin, were identified. When yeast cells are used as a system for modeling mitochondrial disease, the peptides in caseins and whey fractions significantly enhanced the tolerance of yeast cells against peroxide-induced oxidative stress. The results show that both caseins and whey proteins of camel milk possess bioactive peptides with significant radical-scavenging activities and thus herald a fascinating opportunity for their potential as nutraceuticals or therapeutic peptides for prevention and treatment of oxidative stress-associated diseases.

文摘The objective of this study was to evaluate the changes on flavor and quality characteristics of goat yogurt during storage with the addition of dairy bioactive peptides and a mixture of lotus seed and lily bulb powder.Both the addition of dairy bioactive peptides and lotus seeds/lily bulb powder resulted in a fermentation time reduction,water holding capacity(WHC)improvement,and inhibition of post-acidification of goat yogurt during storage.A total of 50 volatile flavor components were detected in the goat milk yogurt samples,and the addition of the above two supplements were shown to be effective in improving the flavor and the textural properties of the goat milk yogurt during storage.The group added with lotus seeds/lily bulb powder showed the best WHC and textural properties,followed by the group added with dairy bioactive peptides alone and the group with both added.The addition of both supplements was found to be useful in making up for the two defects of goat yogurt,prolonged fermentation and soft curds.Therefore,dairy bioactive peptides and lotus seeds/lily bulb powder are recommended to be added to goat yogurt as nutritional supplements.
文摘Milk from different species has been exploited for the isolation of various functional ingredients for decades.Irrespective of the source,milk is considered as a complete food,as it provides essential nutrients required by the human body.Proteins and their fractions are valuable sources of bioactive peptides that might exert a health beneficial role in the human body such as immune-modulation,antioxidant activity,ACE-inhibitory activity,anti-neoplastic,anti-microbial,etc.In milk,bioactive peptides may either be present in their natural form or released from their parental proteins due to enzymatic action.The increasing interest in bioactive peptides among researchers has lately augmented the exploration of minor dairy species such as sheep,goat,camel,mithun,mare,and donkey.Alternative to cow,milk from minor dairy species have also been proven to be healthier from infancy to older age owing to their higher digestibility and other nutritive components.Therefore,realizing the significance of milk from such species and incentivized interest towards the derivatization of bioactive peptides,the present review highlights the significant research achievements on bioactive peptides from milk and milk products of minor dairy species.

文摘Plant proteins are good alternative to animal sources in bioactive peptide production due to their safety,low-cost,rich resources and easy for large scale production.The bioactivities of the peptides derived from plant proteins including antihypertensive,hypocholesterolemic,immunomodulatory,antioxidant,antimicrobial,anti-diabetic,opiate and hepatoprotective effect are being increasingly recognized with regards to health promoting and disease prevention.In addition to bioactivities,the plant-derived bioactive peptides possess various functional properties including emulsifying and foaming properties,and water and oil retention capacities owning to the hydrophilic/lipophilic property,conformational flexibility and chain length of peptides.Hence,the plant-derived bioactive peptides are promising ingredients for the development of health-care and functional products.This review provides the research progress on the biological activities and functional properties of plant derived peptides.The mechanism of action and structure-activity relationships of the bioactive peptide were also described,when known.Furthermore,recent developments,major challenges and prospect in the application of the plant-derive bioactive peptides in the development of functional products are discussed and summarized.The objective of this review is to improve the understanding of plant-derive bioactive peptides,and provide some constructive inspirations for further research and industrial application in the food and medicine industry.
文摘Bioactive peptides are released during the production of fermented dairy,vegetables,fruits,legumes,fish,and meat prod-ucts.The proteolytic specificity of lactic acid bacteria,Bacillus spp.,yeasts,and mold,apart from their ability to synthesize bioactive peptides,plays an important role in the generation of specific bioactive peptides in traditional fermented foods.Controlled fermentation using defined starter strains has been explored for the development of bioactive peptides enriched novel fermented foods with potential functionality.Bioactive peptides enriched foods exert diverse health benefits,such as antioxidant,antihypertensive,antidiabetic,and immunomodulatory effects.Bioactive peptides can be used as alternatives to synthetic compounds due to negligible side effects and high valuation in the nutraceutical and functional food market.However,challenges associated with the identification,quantification,organoleptic properties,and bioavailability of bioac-tive peptides need to be addressed before exploiting the potential of bioactive peptides in the functional food industry.In the present review,we have discussed the production of bioactive peptides in diverse fermented foods.Structural and sequence specificity of peptides and their effect on the expression of distinct health beneficial effects have been described.The potential of utilizing these bioactive peptides for the development of novel functional fermented foods is discussed.Recent advances in peptide identification,quantification,debittering of peptides,and increasing peptide bioavailability have been explained.

文摘Evidences suggest that the homeostasis of gut microbiota is among the most important factors for maintaining the physical and mental health of the host.Among the multiple factors affecting the homeostasis of gut microbiota,diet is one of the decisive factors.Bioactive peptides derived from protein hydrolyzed by protease or fermented by microorganism have many physiological activities that their parent proteins do not have.Currently,bioactive peptides attract more and more attention due to their bidirectional interaction with gut microbes.It has been reported that some bioactive peptides could alter the composition of gut microbiota by influencing the intestinal microenvironment.Meanwhile,quite a few bioactive peptides that are released by gut microbes or intestinal cells could resist the pathogenic bacteria to sustain the homeostasis of gut microbiota.In this review,some exogenous bioactive peptides derived from food and some endogenous bioactive peptides released from intestinal cells or microbes were discussed to summary their effects on the modulation of gut microbiota.This review is expected to provide new ideas for related research,and as well to promote the application of bioactive peptides in the fields of food and medicine.
文摘Bioactive peptides are unique,low molecular weight peptide sequences generally consisting of 2–20 amino acid residues.These peptide sequences are inactive within the parent protein but they become physiologically active once released from the native polypeptide sequence via hydrolysis.There are multiple methods for producing bioactive peptides from precursor protein molecules,with microbial fermentation of various dietary matrices indubitably being a novel method to produce peptides with specialized bioactivity.Fermented foods especially fermented dairy products,legumes,cereals,meat and marine life as a source of bioactive peptides have been well documented.These peptides have gained scientific attention owing to their biofunctional attributes.The food-derived bioactive peptides have the potential to serve as valuable ingredients in functional foods and nutraceutical products to promote health.Bioactive peptides are known to possess various health-promoting properties including anti-carcinogenic,anti-hypertensive,anti-microbial,antioxidant,anti-diabetic,and immunomodulatory effects.The COVID-19 pandemic has put the world’s health,economy,and social stability in jeopardy.The SARS-CoV-2 infection contributes to severe conditions and higher mortality in COVID-19 patients with comorbidities.The viral infection not only causes severe respiratory infection but also causes malfunctioning of the Renin-Angiotensin system(RAS),resulting in the downregulation of Angiotensin-converting enzyme Ⅱ(ACE-Ⅱ)and subsequent accumulation of Angiotensin Ⅱ.Several synthetic ACE inhibitory medications are being used to minimize the severity of Angiotensin Ⅱ adverse effects such as hypertension.The growing concern about the side effects associated with these pharmaceuticals has prompted researchers to look for alternatives in the form of foods and nutraceuticals with health-promoting features.Biologically active peptides have the potential to be used as a new-generation pharmaceutical product for various diseases including COVID-19.The multi-functional food-derived peptides could be a promising approach against COVID-19 infection in patients with chronic complications through their therapeutic actions.However,more in vitro and in vivo studies are required to validate their efficacy in enhancing the survivability and viability of COVID-19 patients.Although many peptides have demonstrated their positive effects via biochemical assays,cell culture,and animal models,the translation of these findings into practical application is limited.This might be related to the bioavailability issues,which influence the correlation of in vitro results with in vivo functions of peptides.To exert a health-promoting impact,these peptides need to withstand severe gastrointestinal conditions and the action of digestive enzymes to reach the target site in an active state.Therefore it is critical to thoroughly investigate the gastrointestinal stability and transport of these biopeptides and devise strategies to improve their absorption and bioavailability.

文摘Hyperlipidemia poses a serious threat to human health and evaluating the ability of natural active substances to regulate disorders of lipid metabolism is the focus of food functionality research in recent years. Bioactive peptides are distinguished by their broad range of sources, high nutritional content, ease of absorption and use by the body, and ease of determining their sequences. Bioactive peptides have a wide range of potential applications in the area of medicines and food. The regulation of lipid metabolism disorder caused by bioactive peptides from different sources provides a reference for the development and research of bioactive peptides for lipid reduction.

文摘Bioactive peptides have various excellent biological activities and serve as functional foods to prevent chronic diseases in the human body.This article investigated the regulatory effect of egg white peptides(EPs)on the intestine barrier of young mice with colitis.The results showed that the intake of EPs could protect the intestines from inflammation damage.Besides,the expression of tight junction proteins and mucin is upregulated.Markedly,the intake of EPs can increase the concentration of amino acids in the serum of young mice,which is crucial for nutritional supplementation during intestinal inflammation process.Proteomics analysis implied that EPs can regulate protein expression in the intestine,involving multiple inflammatory pathways including phosphoinositide 3-kinase(PI3K)-protein kinase B(AKT)and mitogen-activated protein kinase(MAPK)signaling pathway.This demonstrated the health benefits of bioactive peptides and provides a theoretical basis for the development of animal derived proteins as functional foods.
