Background:Cisplatin(DDP)has been used in the treatment of various human cancers.However,DDP alone lacks efficacy in treating triple-negative breast cancer(TNBC),and its clinical application is often hampered by side ...Background:Cisplatin(DDP)has been used in the treatment of various human cancers.However,DDP alone lacks efficacy in treating triple-negative breast cancer(TNBC),and its clinical application is often hampered by side effects.Astragalus polysaccharide(APS)is one of the active components extracted from Astragalus membranaceus and has gained attention for its various biological properties.This research is aimed to evaluate the effectiveness of a combination of APS and DDP on TNBC and explore the potential mechanisms.Methods:The efficacy and mechanisms of single or combined treatment were evaluated using Cell Counting Kit-8(CCK8)assay,Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)staining,wound healing assay,trans-well invasion/migration assay,hematoxylin-eosin(HE)staining,immunohistochemical(IHC)staining,Western Blot(WB)analysis,and fluorescence-activated cell sorting(FACS).An orthotopic model of TNBC was used to assess the in vivo treatment efficacy of single or combination treatment.Results:APS significantly enhanced the anti-proliferative,anti-migratory,and anti-invasive effects of DDP on TNBC cells.The combination of APS and DDP downregulated anti-apoptotic genes(Bcl2 and Bcl-xL)while upregulating pro-apoptotic genes(Puma,Cle-Caspase3,Cle-PARP),leading to enhanced apoptosis.This combination treatment increased E-cadherin levels,decreased Vimentin,Snail,Slug,and Twist levels,and effectively suppressed epithelial-mesenchymal transition(EMT)-associated cell invasion.In the orthotopic model of TNBC,a synergistic reduction in tumor growth was observed in mice treated with APS and DDP.Additionally,the combination of APS and DDP induced the infiltration of CD8+T lymphocytes into the tumor immune microenvironment.Conclusion:The combination of APS and DDP exhibits more potent tumor inhibition and anti-tumor immunity than either agent alone,representing a novel approach to enhance therapeutic efficacy without increasing the side effects of DDP.展开更多
AP(Ammonium Perchlorate)and HMX(Octogen)are the two oxidizers most often used in Nitrate-Ester-Plasticized Polyether(NEPE)rocket propellants.How the AP–HMX ratio influences the agglomeration of NEPE propellants remai...AP(Ammonium Perchlorate)and HMX(Octogen)are the two oxidizers most often used in Nitrate-Ester-Plasticized Polyether(NEPE)rocket propellants.How the AP–HMX ratio influences the agglomeration of NEPE propellants remains unclear.We experimentally investigated the effect of the AP–HMX ratio on the combustion and agglomeration of NEPE propellants using burning rate test,quenched surface analysis,microscopic observations,and the collection of condensed combustion products.It was found that with the decrease in AP content from 40wt%to 10wt%,the burning rate decreased from 14.2 mm/s to 9.2 mm/s because the adiabatic flame temperature of NEPE propellants decreased from 3828 K to 3736 K.Pockets bounded by AP particles appeared on the surface when AP content was 40wt%;however,the accumulations grew and covered the burning surface eventually as the AP–HMX ratio decreased.The time required for the accumulation to coalesce into agglomerates increased with decreasing AP content.Even with similar agglomerate sizes,the coalescence time increased by 83%when the AP content decreased from 40wt%to 30wt%.The agglomerate size in the Condensed Combustion Products(CCPs)increased from 100μm to 200μm,and the fraction of large agglomerations increased from 6.4%to 24.7%when the AP content decreased from 40wt%to 10wt%.Overall,the high flame temperature of the AP particles enhanced the decomposition of the surrounding binder,resulting in the rapid ejection of the aluminum particles into the gas,which had a separating effect on the accumulation,thus weakening the agglomeration.展开更多

Magnolia officinalis is a perennial deciduous tree that has medicinal properties.The AP2/ERF gene family has a number of roles in long-term growth and metabolism.The expression of this function varies with the growth ...Magnolia officinalis is a perennial deciduous tree that has medicinal properties.The AP2/ERF gene family has a number of roles in long-term growth and metabolism.The expression of this function varies with the growth period.In this work,based on the transcriptome data of Magnolia officinalis,the complete coding gene of Magnolia officinalis was obtained,and the corresponding protein sequence was retrieved from NCBI and compared with the model plant Arabidopsis thaliana.After screening,75 protein sequences from the AP2/ERF gene family were identified and called MoAP2/ERF1–MoAP2/ERF75,followed by bioinformatics analysis.75 AP2/ERF gene families were found and classified into four subfamilies.Their protein architectures had one or more conserved AP2 domains,which were typically unstable and hydrophilic.Subcellular research revealed that it was primarily located in the nucleus.Among them,the DREB subfamily showed stronger activity in the early growth period of Magnolia officinalis,suggesting that Magnolia officinalis had stronger resistance to adversity during this period.The 15 members of the MoAP2/ERF gene family showed significant differences during different growth periods,and they regulated the gene expression of Magnolia officinalis by binding to DNA.The 15 MoAP2/ERF gene families have a wide range of physiological activities in biological processes,cellular components,and molecular functions.Including MoAP2/ERF55 can catalyze imidazole glycerol phosphate synthase activity;MoAP2/ERF39 acts as a transcriptional activator of Pti6.展开更多
Acer paxii belongs to the evergreen species of Acer,but it exhibits a unique feature of reddish leaves in fall in subtropical regions.Although the association of AP2/ERF transcription factors with color change has bee...Acer paxii belongs to the evergreen species of Acer,but it exhibits a unique feature of reddish leaves in fall in subtropical regions.Although the association of AP2/ERF transcription factors with color change has been well-documented in prior research,molecular investigations focusing on AP2/ERF remain notably lacking in Acer paxii.This research focuses on performing an extensive genome-wide investigation to identify and characterize the AP2/ERF gene family in Acer paxii.As a result,123 ApAP2/ERFs were obtained.Phylogenetic analyses categorized the ApAP2/ERF family members into 15 subfamilies.The evolutionary traits of the ApAP2/ERFs were investigated by analyzing their chromosomal locations,conserved proteinmotifs,and gene duplication events.Moreover,investigating gene promoters revealed their potential involvement in developmental regulation,physiological processes,and stress adaptationmechanisms.Measurements of anthocyanin content revealed a notable increase in red leaves during autumn.Utilizing transcriptome data,transcriptomic profiling revealed that the majority of AP2/ERF genes in Acer paxii displayed significant differential expression between red and green leaves during the color-changing period.Furthermore,through qRT-PCR analysis,it was found that the gene expression levels of ApERF006,ApERF014,ApERF048,ApERF097,and ApERF107 were significantly elevated in red leaves.This indicates their potential participation in leaf pigmentation processes.These findings offer significant insights into the biological significance of ApAP2/ERF transcription factors and lay the groundwork for subsequent investigations into their regulatorymechanisms underlying leaf pigmentation in Acer paxii.展开更多

Mosquito-borne flaviviruses,such as Zika virus(ZIKV)and dengue virus(DENV),cause diverse severe clinical manifestations including fever,rash,hepatitis,arthralgia,and congenital anomalies.Here,we identified a host fact...Mosquito-borne flaviviruses,such as Zika virus(ZIKV)and dengue virus(DENV),cause diverse severe clinical manifestations including fever,rash,hepatitis,arthralgia,and congenital anomalies.Here,we identified a host factor,the adaptor protein complex 1 gamma 1 subunit(AP1G1),which plays an important role in both ZIKV and dengue virus 2(DENV2)infections.We explored the role of AP1G1 in ZIKV and DENV2 infections using CRISPR/Cas9 gene editing technology and RNA interference(RNAi)techniques.Knockout or silencing of AP1G1 decreases the replication of ZIKV and DENV2 in multiple human cell lines.Intriguingly,depletion of AP1G1 results in a significant reduction in ZIKV at an early stage,but decreases DENV2 replication levels during the late stage,suggesting that AP1G1 plays distinct roles in the infection by ZIKV and DENV2.Furthermore,we determined that AP1G1 mediates ZIKV-endosomal membrane fusion through inhibitor experiments and fluorescence labeling assays.Mechanistically,we found that AP1G1 exerts its pro-viral effect through binding to the ZIKV envelope glycoprotein(E protein).This interaction promotes the fusion of viral and endosomal membranes,during which the ZIKV genomic RNAs are released from the endosome into the cytoplasm,a process that facilitates viral replication.However,for DENV2 infection,AP1G1 primarily affects its viral RNA replication stage,rather than the fusion of virus-endosomal membrane.Taken together,our work demonstrates that AP1G1 plays a pro-viral role in both ZIKV and DENV2 infections via distinct mechanisms,highlighting its potential as a therapeutic target for antiviral strategies.展开更多
