Saikosaponins are the major pharmacologically active components in Bupleurum genus and exhibit significant application potential in multiple fields such as immune regulation and anti-tumor activity.To elucidate the biosynthetic pathway of saikosaponins,we identified two cytochrome P450 monooxygenases,CYP716A41 and CYP716Y4,in Bupleurum chinense.These enzymes catalyze the C-28 oxidation and C-16 hydroxylation of oleanane-type triterpene skeletons,respectively.The catalytic efficiency of CYP716A41 from a southern B.chinense variety was significantly higher than that from a northern variety.Molecular docking and mutagenesis experiments revealed that amino acid residues at sites 9 and 35 may contribute to this difference in catalytic efficiency.Additionally,under cold stress,the expression levels of both CYP450 genes and the saikosaponin contents in the leaves of southern varieties were significantly higher compared to those in northern varieties.The variation in the catalytic efficiency of CYP716A41 and the differential expression of the two CYP450 genes under cold stress during winter are associated with the differences in saikosaponin biosynthesis in the leaves of southern and northern B.chinense varieties.This is consistent with the distinct medicinal usage practices observed between southern and northern China. 展开更多

The present study aimed to investigate the effects of polysaccharides extracted from Bupleurum chinense DC(BCPs) on macrophage functions. In the in vivo experiment, 1 m L of 5% sodium thioglycollate was injected into the abdomen of the mice on Day 0 and macrophages were harvested on Day 4. The macrophages were cultured in plates and treated with different concentrations of BCPs and stimulus. Effects of BCPs on macrophage functions were assessed by chemotaxis assay, phagocytosis assay and Enzyme-Linked Immunosorbent Assay(ELISA). Our results showed the enhanced chemotaxis, phagocytosis and secretion of nitric oxide(NO) and inflammatory cytokines by macrophages when treated with BCPs. However, when chemotaxis and phagocytosis were up-regulated by complement components or opsonized particles, BCPs inhibited these effects. Also, the NO production induced by lipopolysaccharides(LPS) was suppressed by BCPs mildly. Moreover, BCPs had an inhibitory effect on the [Ca^(2+)]i elevation of macrophages. These results suggested that BCPs exerted modulatory effects on macrophage functions, which may contribute to developing novel approaches to treating inflammatory diseases. 展开更多

A quantitative evaluation method for the quality consistency of Bupleurum chinense DC. was developed by combining multiwavelength fusion fingerprint with chemometric methods. The fingerprint information of Bupleurum at five wavelengths (210, 254, 265, 280 and 320 nm) collected by HPLC-DAD instrument was fused to overall control its quality. The fusion fingerprints and four components (saikosaponin a, saikosaponin d, rutin, quercitrin) were studied by systematic quantitative fingerprint method and principal component analysis, respectively. Besides, fingerprint-efficacy relationship was constructed in vitro using partial least-squares model. In conclusion, all work can serve as a valid reference for the quality control of Bupleurum . 展开更多

In the present study, the 70% EtO H extract of the roots of Bupleurum chinense was... 5-Hydroxytryptamine 2C(5-HT2C) receptor is one of the major targets of anti-obesity agents, due to its role in regulation of appetite. In the present study, the 70% EtO H extract of the roots of Bupleurum chinense was revealed to have agonistic activity on 5-HT2 C receptor, and the subsequent bioassay-guided isolation led to identification of several saikosaponins as the active constituents with 5-HT2 C receptor agonistic activity in vitro and anti-obesity activity in vivo. The new compound, 22-oxosaikosaponin d(1), was determined by extensive spectroscopic analyses(HR-ESI-MS, IR, and 1D and 2D NMR). The primary structure-activity relationship study suggested that the intramolecular ether bond between C-13 and C-28 and the number of sugars at C-3 position were closely related to the 5-HT2 C receptor agonistic activity. Saikosaponin a(3), the main saponin in B. chinense, showed obviously agonistic activity on 5-HT2 C receptor with an EC50 value of 21.08 ± 0.33 μmol×L^(–1) in vitro and could reduce food intake by 39.1% and 69.2%, and weight gain by 13.6% and 16.4%, respectively, at 3.0 and 6.0 mg×kg^(–1) in vivo. This investigation provided valuable information for the potential use of B. chinense as anti-obesity agent. 展开更多

Bupleurum chinense is a genuine medicinal material in Shanxi Province,and planting B.chinense is one of the main sources for local farmers to increase their income.In B.chinense cultivation,wheat-B.chinense,maize-B.chinense and soybean-B.chinense are commonly used as multiple cropping patterns.In the present study,we studied the effects of three different multiple cropping patterns on N,P and K levels of soil,photosynthetic parameters,agronomic characteristics,saikosaponin A,C and D contents,yield and economic benefits of B.chinense.The results showed that the soybean-B.chinense multiple cropping pattern was beneficial to the enrichment of N in the soil,thus promoting photosynthetic parameters,growth,saikosaponin accumulation,yield and economic benefits of B.chinense.It is suggested that soybean-B.chinense multiple cropping was an appropriate multiple cropping pattern for local cultivation of B.chinense. 展开更多

Effects of Different Treatments on Seed Germination of Bupleurum chinensis

In order to solve the problem of low emergence rate of Bupleurum chinense seeds, and screen out the best germination conditions, the seeds of Bupleurum chinense L. were treated with different soaking time, germination temperature and exogenous regulating substances to improve the germination rate. The results showed that the germination rate of fresh water at 12 h was significantly higher than that at 8 h and 36 h; The germination rate of seeds treated with constant temperature 15℃ and variable temperature 25 ℃/15 ℃ (day/night) was significantly higher than that of other temperature treatments. Seed soaking time and culture temperature also influenced seed germination start day and peak day. Different concentrations of regulatory substances could promote seeds germination. The germination rate of seeds induced by 0.50 mg/L 6-BA was the highest, the start day and peak day were 4 days and 5 days earlier than control check (CK) respectively. 展开更多

基于网络药理学与分子对接探究北柴胡叶提取物抗APAP肝损伤作用

为探究北柴胡叶提取物(BCLE)抗对乙酰氨基酚(APAP)肝损伤的作用,采用液相-质谱法(LC-MS)、网络药理学、分子对接,并建立小鼠模型研究BCLE治疗效果及抗APAP肝损伤作用机制。网络药理学结果表明,BCLE活性成分有19种,BCLE与DILI公共靶点244... 为探究北柴胡叶提取物(BCLE)抗对乙酰氨基酚(APAP)肝损伤的作用,采用液相-质谱法(LC-MS)、网络药理学、分子对接,并建立小鼠模型研究BCLE治疗效果及抗APAP肝损伤作用机制。网络药理学结果表明,BCLE活性成分有19种,BCLE与DILI公共靶点244个,核心靶点44个,且活性成分与脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)信号通路相关性较高,分子对接显示活性成分与通路之间亦有良好的亲和力。动物试验结果表明,BCLE对APAP肝损伤有良好的治疗效果,可显著降低APAP模型组小鼠血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)活力,缓解肝细胞损伤,减轻N-乙酰基-P-苯醌亚胺(NAPQI)的聚积,升高还原型谷胱甘肽(GSH)含量和超氧化物歧化酶(SOD)活性,降低丙二醛(MDA)和过氧化氢(H_(2)O_(2))含量,并显著上调APAP肝损伤小鼠磷酸化脂酰肌醇3-激酶(p-PI3K)、磷酸化蛋白激酶B(p-AKT)蛋白表达。综上,网络药理学与分子对接预测BCLE通过PI3K-AKT通路作用于APAP肝损伤,体内试验表明,BCLE可通过保护肝细胞,缓解氧化应激,上调PI3K-AKT通路抗APAP肝损伤。研究结果可为进一步提升北柴胡的药用价值,研发抗APAP肝损伤新型药物提供理论思路和试验依据。 展开更多

As a traditional Chinese herbal medicine exhibiting analgesic,fever-reducing and anti-inflammatory effects,Radix Bupleuri(Chai-Hu) is commonly used for the treatment of influenza,which is derived from the dried roots of Bupleurum chinense DC.and Bupleurum scorzonerifolium Willd.Among of diverse chemical components,saikosaponins are the key active components of the herb medicine.In the present study,we established a method of high performance liquid chromatography(HPLC) coupled with evaporative light scattering detection(ELSD) for simultaneous determination of saikosaponin a,c and d in root,stem,leaf and flower of Bupleurum chinense(B chinense) collected from different areas of Shanxi Province,China.The results from 16 samples of root,stem,leaf and flower of B chinense demonstrated that the total contents of the three saikosaponins in the root of B chinense collected from Dongshan Taiyuan,Xishan,Tianlongshan and Pangquangou were 4.26 mg/g,3.22 mg/g,4.23 mg/g and 3.05 mg/g,respectively.However,there was scarcely any saikosaponins in the stem,leaf and flower of B chinense collected from above-mentioned areas.The method of HPLC coupled with ELSD was suitable for quality control of Radix Bupleuri.The result also confirmed that the root of B chinense was the best medicinal part. 展开更多

目的:探索南、北柴胡油对LO2肝细胞安全性差异以及潜在的作用机制。方法:采用气相-四极杆-飞行时间质谱(GC-Q-TOF-MS/MS)技术和化学计量学,对南、北柴胡挥发油的化学成分进行系统分析鉴定。通过体外培养LO2肝细胞,采用细胞形态学和CCK-... 目的:探索南、北柴胡油对LO2肝细胞安全性差异以及潜在的作用机制。方法:采用气相-四极杆-飞行时间质谱(GC-Q-TOF-MS/MS)技术和化学计量学,对南、北柴胡挥发油的化学成分进行系统分析鉴定。通过体外培养LO2肝细胞,采用细胞形态学和CCK-8检测,分别观察和检测南、北柴胡油对LO2肝细胞形态、数量、增殖和凋亡的影响。采用Western Blot测定LO2肝细胞凋亡通路相关蛋白表达。结果:南、北柴胡油共检测化学成分116种,其中北柴胡油83种,南柴胡油76种,共有组分43种。经OPLS-DA判别分析获得14个主要特征标志物(VIP>1.5)。细胞实验显示,南、北柴胡油对LO2肝细胞作用的凋亡率呈浓度依赖性增加(P<0.05),但二者安全剂量差异明显,北柴胡油IC_(50)值为(331.42±12.43)μg/mL,南柴胡油IC_(50)为(809.17±32.64)μg/mL。二者皆可通过影响抗凋亡蛋白Bcl-2和促凋亡蛋白Bax的表达抑制LO2肝细胞的增殖(P<0.01)。结论:南柴胡油的安全性高于北柴胡油。南、北柴胡挥发油成分中烯类、酚类特征化合物质可能是二者安全性差异的潜在物质基础。高剂量下,两种柴胡油影响Bcl-2/Bax蛋白比例的平衡,并通过内源性途径促进LO2肝细胞的凋亡。 展开更多

目的发掘植物激素信号通路中与北柴胡Bupleurum chinense侧根发育紧密关联的基因。方法以自组拼接的北柴胡基因组为模板,对北柴胡推广品种川北柴1号苗期根系进行有参考基因组的转录组分析,筛选参与植物激素信号转导的关键基因,利用实时... 目的发掘植物激素信号通路中与北柴胡Bupleurum chinense侧根发育紧密关联的基因。方法以自组拼接的北柴胡基因组为模板,对北柴胡推广品种川北柴1号苗期根系进行有参考基因组的转录组分析,筛选参与植物激素信号转导的关键基因,利用实时荧光定量聚合酶链式反应(real-time fluorescent quantitative polymerase chain reaction,qRT-PCR)技术验证关键基因对外源生长素处理的表达应答。结果有参转录组分析得到32259个基因,681个基因在有侧根成熟区(RO)样本中特异表达。加权基因共表达网络分析将基因分为56个模块,2个模块与主根长度和侧根数目显著相关。与侧根形成高度相关的生长素信号转导与运输通路上共发掘到88个差异基因,其中26个基因表达模式高度相关。转录组差异基因表达相关性与外源生长素处理后的基因表达结果联合分析表明,BcIAA13、BcARF19、BcSAUR55和BcSAUR72基因可能是侧根生长发育的关键基因。结论进一步揭示了在RO样本中表达量高的基因可能与侧根发育密切相关,并筛选到转录因子BcARF19与BcIAA13密切相关,其可能调控下游基因的表达,进而影响侧根的生长发育。 展开更多

目的研究南北柴胡气味物质差异。方法应用电子鼻技术和气相色谱-离子迁移谱联用(GC-IMS)技术检测南北柴胡挥发性物质差异,采用NIST、IMS数据库对挥发性物质进行定性和定量分析;使用仪器配套的插件对南北柴胡样品进行动态主成分分析(Dyna... 目的研究南北柴胡气味物质差异。方法应用电子鼻技术和气相色谱-离子迁移谱联用(GC-IMS)技术检测南北柴胡挥发性物质差异,采用NIST、IMS数据库对挥发性物质进行定性和定量分析;使用仪器配套的插件对南北柴胡样品进行动态主成分分析(Dynamic PCA),利用SIMCA-P 14.0软件对其进行正交偏最小二乘法-判别分析(OPLS-DA),以微生信平台对其进行聚类热图分析;对比南北柴胡指纹图谱,分析南北柴胡特征物质,筛选Student's t-test P<0.05且正交偏最小二乘法-判别分析模型中变量投影重要性(VIP)>1的挥发性物质为气味差异标志物。结果南北柴胡挥发性成分具有显著差异,南柴胡辨识出4个特征物质,分别是癸醛(单体)、氧化芳樟醇、2,3-二乙基-5-甲基吡嗪、异戊烯醛(二聚体);北柴胡辨识出2个特征物质,分别是3-甲基丁醇(二聚体)、2-甲基丙酸。共筛选出24个气味差异标志物,这些成分可以作为区分南北柴胡的依据。结论应用GC-IMS或电子鼻气味辨识技术能够快速鉴别南北柴胡,南柴胡败油气可能与其特征物质异戊烯醛(二聚体)及气味差异标志物癸醛、反-4-己烯醛、正辛醛、庚醛、正己醛、丙烯醛、异丁醛等醛类物质有关。 展开更多

采用薄层色谱、硅胶柱色谱、制备型高效液相色谱等多种色谱分离技术,对北柴胡茎叶中的化学成分进行分离,根据理化性质及波谱数据对结构进行鉴定。采用Ellman法测定化合物的乙酰胆碱酯酶抑制活性,并利用MTT法检测其对神经细胞的保护活性... 采用薄层色谱、硅胶柱色谱、制备型高效液相色谱等多种色谱分离技术,对北柴胡茎叶中的化学成分进行分离,根据理化性质及波谱数据对结构进行鉴定。采用Ellman法测定化合物的乙酰胆碱酯酶抑制活性,并利用MTT法检测其对神经细胞的保护活性。从北柴胡茎叶中分离得到15个化合物,Erigeroflavanone(1),槲皮素(2),槲皮素-3-鼠李糖苷(3),山柰酚3-O-α-L-鼠李糖甙(4),木犀草素(5),Morin-3-O-α-rhamnopyranoside(6),咖啡酸甲酯(7),焦谷氨酸甲酯(8),3,4-二羟基苯甲酸甲酯(9),香草酸(10),3,4-二羟基苯甲酸(11),对苯二酚(12),1,2,4-苯三酚(13),9(Z)-octadecenamide(14)(Z)-21-methyl-8-pentatriacontene(15)。化合物1,4,6,8,12,14,15为首次从柴胡属植物中分离得到。化合物5~9,11,13~15具有乙酰胆碱酯酶抑制活性,IC50值分别为78.92μM、9.60μM、20.15μM、30.99μM、0.76μM、0.17μM、60.25μM、12.70μM、9.53μM。化合物1,3,5,9~12显示出神经细胞保护作用,最高细胞存活率分别为58.45%、56.88%、60.98%、84.4%、62.50%、60.43%、59.58%,其中化合物9神经保护作用最强。 展开更多

目的明确炒柴胡Bupleurum chinense炮制的关键影响因素,同时探究炒柴胡外观颜色与品质的内在关系,为炒柴胡的规模化生产提供参考依据。方法以炒制温度和炒制时间为考察因素,外观性状、柴胡皂苷a(saikosaponin a,SSa)、柴胡皂苷b1(saikos... 目的明确炒柴胡Bupleurum chinense炮制的关键影响因素,同时探究炒柴胡外观颜色与品质的内在关系,为炒柴胡的规模化生产提供参考依据。方法以炒制温度和炒制时间为考察因素,外观性状、柴胡皂苷a(saikosaponin a,SSa)、柴胡皂苷b1(saikosaponin b1,SSb1)、柴胡皂苷b2(saikosaponin b2,SSb2)、柴胡皂苷c(saikosaponin c,SSc)、柴胡皂苷d(saikosaponin d,SSd)、醇溶性浸出物为评价指标,通过AHP-CRITIC综合赋权法确定各指标权重系数并计算综合评分(OD值);基于单因素实验结果,应用中心复合设计-响应面法(central composite design-response surface methodology,CCD-RSM)优选炒柴胡的最佳炮制工艺,联合IRIS电子眼定量测定柴胡炒制前后颜色变化,运用SPSS AU软件进行颜色与指标成分相关分析,建立回归模型。结果AHP-CRITIC法确定外观性状、SSa、SSb1、SSb2、SSc、SSd、醇溶性浸出物的权重系数分别为0.5436、0.1024、0.0908、0.0969、0.0362、0.0990、0.0310,优选的炒柴胡饮片炮制工艺为炒制温度160℃、炒制时间10 min,综合得分(OD值)为81.55,5次验证结果与预测值之间的RSD为0.91%,说明模型具有良好的预测性。优化的炒柴胡饮片炮制工艺稳定性较好,炒制之后柴胡表面呈现深黄色,微具焦斑,具有炒香气。由相关性分析可知,L^(*)、b^(*)与SSa、SSc、SSd含量呈正相关(P<0.05);L^(*)与SSb1、SSb2含量呈负相关(P<0.05);而a^(*)则与SSb1、SSb2含量呈正相关(P<0.05)。提示色度值可在一定程度上预测SSa、SSb1、SSb2、SSc、SSd含量。结论优选出的炒柴胡炮制工艺稳定、合理、可行,可为炒柴胡饮片的质量标准建立及未来工业化、数字化生产提供科学依据。 展开更多
