[ Methods ] Acetylcholinesterase (ACHE) was added to cowpea extract, to determine the inhibition rate of extract against enzyme. The influences of different sampiing methods and sampling parts on detection results were compared. [ Results] The positive rate of standard sampling was 18.18% higher than that of non-stand- ard sampling, and the positive rate of samples collected from cowpea tail was 16.67% higher than that collected from other parts. Objective:To investigate the application value of thrombelastogram(TEG)in the detection of platelet inhibition rate for antiplatelet therapy for acute non-cardiogenic stroke.Methods:A total of 100 patients with ischemic non-cardiogenic stroke were selected for this study from September 2020 to October 2021.Patients were randomly divided into experimental group and control group,with 50 cases for each group.Before and after 1 week of antiplatelet drug treatment,the platelet inhibition rate in the experimental group was measured with arachidonic acid(AA)and adenosine diphosphate(ADP)by TEG;no platelet inhibition rates detection was conducted for the control group.The dose and type of drugs were adjusted for the experimental group according to the platelet functions and medication based on the clinical experience conducted for the control group.The neurological deficits of the discharged patients were scored with NIHSS score,mRS score,stroke recurrence,hemorrhage,and other events were followed up at the 3rd month of discharge.Results:In the experimental group,the inhibition rates of AA and ADP were significantly higher than those before treatment(both P<0.05).After treatment,the inhibition rates of AA and ADP in dual antiplatelet patients were higher than those of monoclonal antiplatelets(both P<0.05).The NIHSS score at discharge and the mRS score at the 3rd-month-follow-up in the experimental group were lower than those in the control group(both P<0.05).The incidences of stroke recurrence and hemorrhage events in the experimental group were lower than those in the control group(P<0.05).Conclusion:The application of a thrombelastogram in the detection of platelet inhibition rate to guide antiplatelet therapy in patients with acute non-cardiogenic stroke reduces the recurrences of cerebral infarction and the risk of hemorrhage and improves patients’clinical prognosis. 展开更多

The morphological changes in osteosarcoma cells were observed by fluorescence microscopy. It was found that osteosarcoma cells exposed with 153Sm-EDTMP displayed significant nuclear fragmentation and marked pyknosis. With the prolongation of observing period, the membrane bound apoptotic bodies formation was observed. It should be noted, that with the lengthening of irradiation time by 153Sm-EDTMP, the inhibition rate of proliferation of osteosarcoma cells increased progressively. 展开更多

为高值化开发利用我国资源丰富的樟树籽、筛选樟树籽仁源α-葡萄糖苷酶抑制肽,本研究以樟树籽仁蛋白为原料,采用水解度(DH)和α-葡萄糖苷酶抑制率作为评价指标,通过单因素和正交试验优化制备得到樟树籽仁源α-葡萄糖苷酶抑制肽,并测定... 为高值化开发利用我国资源丰富的樟树籽、筛选樟树籽仁源α-葡萄糖苷酶抑制肽,本研究以樟树籽仁蛋白为原料,采用水解度(DH)和α-葡萄糖苷酶抑制率作为评价指标,通过单因素和正交试验优化制备得到樟树籽仁源α-葡萄糖苷酶抑制肽,并测定分析其氨基酸组成和体外抗氧化活性。结果表明,樟树籽仁源α-葡萄糖苷酶抑制肽的最优酶解工艺为:以木瓜蛋白酶为水解酶,底物浓度为2%(w/v)、酶解温度为55℃、酶解pH为6.0、酶解时间为2.0 h、酶添加量为16000 U/g,该条件下樟树籽仁蛋白酶解物的DH为26.81%。制得的樟树籽仁源α-葡萄糖苷酶抑制肽对α-葡萄糖苷酶、DPPH自由基和ABTS+自由基的半数抑制率(IC50值)分别为11.52、5.42和0.64 mg/mL。本研究为樟树籽仁源α-葡萄糖苷酶抑制肽的挖掘提供了理论依据。 展开更多

针对胜利油田CO_(2)驱大规模开采面临的井下高含水严重CO_(2)腐蚀问题,以缓蚀剂单体咪唑啉、季铵盐、硫脲、醇胺、脂肪醇为原料,复配优选出一种新型耐高温抗CO_(2)缓蚀剂。通过测试,优选缓蚀剂主要缓蚀组分为咪唑啉20%+季铵盐10%+硫脲4%... 针对胜利油田CO_(2)驱大规模开采面临的井下高含水严重CO_(2)腐蚀问题,以缓蚀剂单体咪唑啉、季铵盐、硫脲、醇胺、脂肪醇为原料,复配优选出一种新型耐高温抗CO_(2)缓蚀剂。通过测试,优选缓蚀剂主要缓蚀组分为咪唑啉20%+季铵盐10%+硫脲4%+醇胺10%。试验表明,缓蚀剂投加浓度200 mg/L以上时,缓蚀率均能达到85%及以上,适用于不同温度、流速和CO_(2)压力条件。缓蚀机理研究表明,吸附作用遵循Langmuir吸附模型,增大了表面电荷传递电阻,抑制了腐蚀阳极过程,极大减轻了钢表面腐蚀。现场试验加药浓度为200 mg/L,可实现井下腐蚀速率小于0.076 mm/a,满足防护需求。 展开更多

Inhibition Effect of Antagonistic Fungal Strain BOS-013 against the Pathogen of Rice Blast (Pyricularia oryzae)

[ ObjeeUve ] To search for the beneficial microorganisms and their metabolic products with control effect against rice Mast, the paper studied the inhibi- tion effect of the fermentation broth of actinomycetes BOS-013 strain against the pathogen of rice blast (Pyricularia oryzae). [ Method ] With P. oryzae as the target indicator fungus, the inhibition effect of the fermentation broth of BOS-013 strain and its extracts from different solvents against the pathogen were determined using cylinder plate method and filter paper method. [ Result] The results showed that the inhibition rate of the fermentation broth of BOS-013 strain with fungus amount of 4×10^8 cfu/ml against P. oryzae was 92.0% ±0.5% ; the fermentation broth still had inhibition effect against P. oryaae after inactivation under high temperature and high pressure, and the inhibition rate was 60.0%±0.5%. After absorbed by non-polar CAD-45 type macroperous adsorption resin and eluted by 60% etha- nol, the antifungal activity of the fermentation broth was still high, and the diameter of inhibition zone reached 33.0 mm. [ Conclusion] The fermentation broth of BOS-O13 strain had significant inhibition effect against P. oryzae. The results would provide foundation for further clarification of inhibition mechanism of BOS-013 strain fermentation broth against P. oryzae and its practical application. 展开更多

青稞麸皮β-葡聚糖提取工艺优化及体外降脂活性

以青稞麸皮为原料,通过水提法提取β-葡聚糖,以β-葡聚糖提取率为评价指标,通过单因素和响应面试验优化青稞麸皮β-葡聚糖的提取工艺;以纯化后β-葡聚糖纯度和提取率为主要评价指标,优化青稞麸皮β-葡聚糖的纯化工艺。结果表明,青稞麸皮... 以青稞麸皮为原料,通过水提法提取β-葡聚糖,以β-葡聚糖提取率为评价指标,通过单因素和响应面试验优化青稞麸皮β-葡聚糖的提取工艺;以纯化后β-葡聚糖纯度和提取率为主要评价指标,优化青稞麸皮β-葡聚糖的纯化工艺。结果表明,青稞麸皮β-葡聚糖最优提取工艺为提取液pH8、料液比1∶20(g/mL)、提取温度66℃、提取时间2.25 h。最优条件下β-葡聚糖提取率为(5.027±0.064)%,与预测值5.014%相符合。青稞麸皮β-葡聚糖最优纯化条件为在提取液中加入α-淀粉酶(50 U/L),60℃水浴1 h,后加入糖化酶(250 U/L),60℃孵浴1 h,离心(3000 r/min、10 min)后取上清液浓缩,醇沉离心,沉淀热水复溶,冻融后离心,冻干沉淀。纯化得到的青稞麸皮β-葡聚糖纯度为(77.50±0.60)%。对所得β-葡聚糖进行胰脂肪酶抑制能力测定,发现当青稞麸皮β-葡聚糖浓度为10.0 mg/mL时,对胰脂肪酶抑制率可达76.70%。综上,优化后的青稞麸皮β-葡聚糖水提工艺简单、稳定,得到的β-葡聚糖产品有良好的体外降脂活性。 展开更多

2024—2025年西宁市生鲜果蔬农药残留快速检测分析

目的 调查研究2024—2025年西宁市生鲜果蔬农产品的农药残留情况。方法 本研究采用酶抑制法对西宁市某农产品交易市场和产地采购的样品, 185个生鲜蔬菜和125个生鲜水果,其中生鲜蔬菜13464批次,生鲜水果5712批次,共计310个品种19176批次... 目的 调查研究2024—2025年西宁市生鲜果蔬农产品的农药残留情况。方法 本研究采用酶抑制法对西宁市某农产品交易市场和产地采购的样品, 185个生鲜蔬菜和125个生鲜水果,其中生鲜蔬菜13464批次,生鲜水果5712批次,共计310个品种19176批次样品,进行有机磷和氨基甲酸酯类农药残留快速检测与分析。结果 共有30个蔬菜品种、7个水果品种, 60批次样品检测出农药残留超标。其中,超标率前5的果蔬是黄瓜、青笋、香笋、旱瓜和线茄。农药残留超标样品的检测值在51.30%~99.89%之间,总体定性初筛阳性率为0.33%,合格率在98.92%~99.94%之间,整体样品合格率为99.67%,合格率较高。结论 2024—2025年西宁地区生鲜果蔬农产品中农药残留情况处于相对安全的水平,但仍存在农药残留超标的情况。建议持续加强检测与监测,进一步保障市民的食品安全问题。 展开更多

疏松砂岩有水气藏气井井口增压开发制度研究及应用

为了解决S气田井口增压工艺因地面设备与地层能量不匹配导致的抽吸失衡问题,以S气田出砂出水气井为研究对象,在分析增压开采过程中控砂、携液与携砂的动态关系基础上,指出了多因素联动机制在气井稳定运行中的关键作用,并通过控砂产量与... 为了解决S气田井口增压工艺因地面设备与地层能量不匹配导致的抽吸失衡问题,以S气田出砂出水气井为研究对象,在分析增压开采过程中控砂、携液与携砂的动态关系基础上,指出了多因素联动机制在气井稳定运行中的关键作用,并通过控砂产量与携液流量的协同耦合,建立了“控砂—排液一体化”井口增压制度优化技术。研究结果表明:(1)明确了最佳增压时机,积液井油压降至1.1~1.3倍外输压力、非积液井油压降至1.1倍外输压力时实施增压;(2)构建了井口增压开发制度,以临界出砂流量为上限、以临界携液与携砂流量较大值为下限,取最大允许产量为合理产量,基于合理产量采取节点分析方法计算井口压力,当临界出砂流量不足时,以其为基准并辅以气举协同排液;(3)优选了关键计算模型,临界出砂预测采用PT模型,井筒压力计算选用Hagedorn-Brown模型,临界携液流量依据水气比情况选择李闽模型或谭晓华模型。结论认为:(1)现场应用符合率为82%,实际进气压力、产气量的平均偏差分别为10%、8%,可靠性较高;(2)出水气井增产效果随产水量的增加呈递减趋势,不出水气井则受动态储量与渗透率的共同影响,在渗透率高于40 mD时,单位剩余动态储量下的增产量与渗透率呈正相关关系,低于40 mD时该值主要受渗流能力制约;(3)该项研究成果为同类气田低压井开发提供了参考。 展开更多

Inhibitory Effect of Buddlejasaponin Ⅳ on Hepatocarcinoma 22(H_(22)) in Mice 被引量:1

[Objective] This study aimed to investigate the inhibitory effect of buddle- jasaponin IV on growth of hepatocarcinoma 22 (H22) tumor in mice. [Method] The H= tumor cells were transplanted in the right axillary skins of mice. The tumor- bearing mice were randomly divided into five groups, including control group, CTX group (20.0 mg/kg) and buddlejasaponin IV treatment groups (0.25, 0.50 and 1.00 mg/kg). There were 10 mice in each group. During the treatment, the body weights and survivals of mice in all groups were recorded. The buddlejasaponin IV was in- jected into the abdominal cavities of mice, which lasted for 10 consecutive days. All the mice were slaughtered the next day. The tumors in the abdominal cavities were tanked out and weighed. The tumor inhibition rate, spleen index and thymus index, as well as SOD activity, MDA content, GGT activity and AKP activity in serum were determined. [Result] Compared with the control group, the high- and middle- dosage buddlejasaponin IV treatment groups all showed significant (P〈0.01) inhibito- ry effects on transplanted H22 tumor in mice with tumor inhibition rates of 56.96% and 50.63%, respectively. Compared with those in the control group, the SOD ac- tivity of mice in the high-dosage buddlejasaponin IV treatment group was significant- ly increased (P〈0.05), and the MDA contents, GGT and AKP activities in mice in the high-, middle- and low-dosage buddlejasaponin IV treatment groups were all sig- nificantly reduced (P〈0.01). There were no significant differences in all the indexes, except SOD activity, between the CTX and control groups. [Conclusion] Buddlejas- aponin IV has certain inhibitory effect on H22 tumor, of which the mechanism might be related to antioxidation capacity in body. 展开更多

In the report, the model of Murine Ehrlich Ascites Cells cultured in vivo and vitro is elected for a series of experiments. It has been proved that Na 211At and 211At-Te colloid injections possess an inhibition effect on tumor cells. The inhibition effect was expressed in surviving of the mice and inhibiting growth of tumor as well as the changes of enzyme activity. [Objectives]To fully understand the quality of commercial enzyme inhibition-colorimetric pesticide residue rapid detection kits,so that they can play a greater role in the detection and supervision of agricultural products.[Methods]The sensitivity of 28 kinds of pesticides was determined by using the commercially available enzyme inhibition colorimetric rapid detection kit with Hendu brand.[Results]There was a significant difference in the sensitivity of the kit to each pesticide,and the kit was more sensitive to dichlorvos among the 28 pesticides tested.The sensitivity to methyl isosalifos,dimethoate,isocarbophos,fenthion and phorate was poor,and the sensitivity to quinalphos was different between 3.0 and 2.5 mL.[Conclusions]The large difference of the sensitivity of the enzyme inhibition-colorimetric rapid detection kit for pesticide residues to different kits is a reason for the false positive and false negative test results of the kit,which needs to be considered by relevant personnel. 展开更多

Correlation between Hemagglutination Inhibition Titer and Protection against Infectious Bronchitis Virus Challenge in SPF Layers

[ Objective] To study the correlation between HI titer and protection against IBV challenge in SPF layers. [ Method ] SPF layers were randomly divided into four groups, namely group A1, A2, B1 and B2. The group A1 was immunized with H120 live vaccine. The group A2 was first immunized with H120 live vaccine and later boosted with ND-IB-EDS trivalent inactivated vaccine. The group B1 was used as unimmunized chal- lenge control. The group B2 was kept as unimmunized unchallenged control. The blood samples were taken prior and post-vaccination at intervals and HI tests were conducted. At the laying peak, the group A1, A2 and B1 were challenged with IBV M4t virulent strain. The clinical features and egg production of layers were monitored and recorded. [Result] After 30 d post vaccination with H120 live vaccine, the HI titer reached 4.45 log2; after 30 days boosting with ND-IB-EDS trivalent inactivated vaccine, the HI titer reached to 7.35 log2. Before challenge, HI antibody titer in group A1, A2, B1 and B2 were respectively 4.24 log2, 7.40 Iog2, 2.10 log2 and 2.10 log2. After challenge, chickens in unimmunized challenge control group B1 showed respiratory symptoms, egg production dropped by 30.9%, and they produced more soft-shelled, no-shelled or abnormal eggs. In the group A1, some chickens had light respiratory symptoms and egg production dropped by 11.7%. In the group A2, the egg production of all chickens was as normal as the group B2. [ Conclusion] When the HI titer was over 6 log2, challenge by virulent virus had no impact on egg produc- tion; when the HI titer was 5 log2, 4 log2 and less 3 log2, egg production dropped by 6.0%, 11.3% and 29.6%, respectively. Thus, the HI anti- body level in chickens has close correlation with protection against IBV challenge. 展开更多

响应面优化超声辅助双水相提取无花果叶多糖及生物活性研究

以无花果叶为原料,通过用双水相体系(Aqueous two-phase system,ATPS)结合超声技术对无花果叶多糖的提取工艺进行优化以提高其多糖得率,并对其体外抗氧化活性分析。首先考察了不同ATPS的分配行为得出最佳ATPS溶剂,再采用单因素对料液比... 以无花果叶为原料,通过用双水相体系(Aqueous two-phase system,ATPS)结合超声技术对无花果叶多糖的提取工艺进行优化以提高其多糖得率,并对其体外抗氧化活性分析。首先考察了不同ATPS的分配行为得出最佳ATPS溶剂,再采用单因素对料液比、超声处理时间、温度和功率进行了考察,在此基础上用响应面法对无花果叶多糖的提取工艺优化,并分析其体外抗氧化活性和降血脂能力。最佳ATPS溶剂为质量分数12%聚乙二醇6000(PEG6000)和16%(NH_(4))_(2)SO_(4),最佳提取工艺条件为料液比1:22 g·mL^(-1),超声提取功率360W,超声提取时间25 min,超声波提取温度50℃。在此条件下,多糖得率最高可达13.36±0.08%,与预测值接近。无花果叶多糖抗氧化能力及胰脂肪酶抑制能力随着浓度提高而增强,其清除DPPH·和·OH的IC_(50)值分别为0.33和0.32 mg·L^(-1),其对胰脂肪酶抑制率的IC_(50)值分别为0.94 mg·mL^(-1),说明具有较好的抗氧化性能和体外降血脂活性。双水相法提取工艺稳定、可行,可用于无花果叶多糖的提取,为进一步纯化植物多糖奠定了基础,也为植物多糖的提取提供了一种有价值的替代方法。 展开更多

外施褪黑素对本生烟抗东亚西番莲病毒的效果及作用分析

【目的】明确褪黑素在寄主植物本生烟抵抗东亚西番莲病毒(East Asian Passiflora Virus,EAPV)中的效果和作用方式,阐明褪黑素抗EAPV的作用方式及机制。【方法】以不同浓度(0、50、100、200、400μmol/L)褪黑素处理接种EAPV前后的本生烟... 【目的】明确褪黑素在寄主植物本生烟抵抗东亚西番莲病毒(East Asian Passiflora Virus,EAPV)中的效果和作用方式,阐明褪黑素抗EAPV的作用方式及机制。【方法】以不同浓度(0、50、100、200、400μmol/L)褪黑素处理接种EAPV前后的本生烟幼苗,以无褪黑素处理也不接种病毒的健康植株为对照(CK),通过测定各处理本生烟株高等外部症状、病毒分子检测结果及水杨酸(SA)和茉莉酸(JA)浓度变化,筛选适宜的褪黑素浓度;通过酶联免疫反应(ELISA)测定本生烟OD_(405)值,量化评价褪黑素的保护作用(抗侵染)和抑制作用(抗复制)。【结果】接种EAPV后,本生烟幼苗株高显著变矮,未喷施褪黑素(T0)本生烟感染EAPV后株高为6.4(±0.20)cm,仅为CK的52.2%。喷施不同浓度褪黑素对植株变矮情况有一定缓解作用,其中200μmol/L褪黑素处理(T3)效果最为显著。200μmol/L褪黑素叶面喷施本生烟后1、4、7 d接种EAPV,21 d后染病本生烟株高分别为T0的1.66、1.27和1.06倍。200μmol/L褪黑素对本生烟的保护率均小于0,表明该浓度褪黑素对本生烟未起到保护作用,但该浓度褪黑素对染病本生烟体内EAPV有一定抑制作用,平均抑制率约为26.9%。本生烟激素水平变化方面,褪黑素处理后1~7 d,本生烟体内JA浓度从0 d的10.38 ng/g显著上升到7 d的123.37 ng/g,而SA水平则无明显变化。【结论】本生烟接种EAPV后,植株显著矮化,外施褪黑素可明显缓解本生烟矮化状况,最佳浓度为200μmol/L;喷施褪黑素后接种EAPV,本生烟体内JA水平显著上升而SA水平未发生明显变化,提示褪黑素抗病毒作用方式可能并非保护寄主免受EAPV侵染,而是本生烟在外源褪黑素诱导下,激活JA介导的诱导系统抗性(ISR),从而对EAPV有一定抑制作用。 展开更多
