Diterpenoids and lignans from fossil Chinese medicinal succinum and their activity against renal fibrosis

Five previously undescribed diterpenoids,named succipenoids D‒H(1‒5),along with four undescribed lignans,named succignans A‒D(6‒9),were isolated from the dichloromethane extract of Chinese medicinal succinum.Compounds 1‒5 were characterized as nor-abietane diterpenoids,while compounds 6‒9 were identified as lignans polymerized from two groups of phenylpropanoid units.The structures of these novel compounds,including their absolute configurations,were determined through spectroscopic and computational methods.Biological assessments of renal fibrosis demonstrated that compounds 6 and 7 effectively reduce the expression of proteins associated with renal fibrosis,includingα-smooth muscle actin(α-SMA),collagen I,and fibronectin in transforming growth factor-β1(TGF-β1)induced normal rat kidney proximal tubular epithelial cells(NRK-52e). 展开更多

Retrograde adeno-associated viruses(AAVs)are capable of infecting the axons of projection neurons and serve as a powerful tool for the anatomical and functional characterization of neural networks.However,few retro-grade AAV capsids have been shown to offer access to cor-tical projection neurons across different species and enable the manipulation of neural function in non-human primates(NHPs).Here,we report the development of a novel retro-grade AAV capsid,AAV-DJ8R,which efficiently labeled cortical projection neurons after local administration into the striatum of mice and macaques.In addition,intrastriatally injected AAV-DJ8R mediated opsin expression in the mouse motor cortex and induced robust behavioral alterations.Moreover,AAV-DJ8R markedly increased motor cortical neuron firing upon optogenetic light stimulation after viral delivery into the macaque putamen.These data demonstrate the usefulness of AAV-DJ8R as an efficient retrograde tracer for cortical projection neurons in rodents and NHPs and indicate its suitability for use in conducting functional interrogations. 展开更多

目的:探究黄芪多糖(astragalus polysaccharide,APS)在缺氧条件下促进血管新生的作用机制与靶点。方法:于体外鼠尾胶基质上种植胸主动脉血管段构建新生血管三维培养模型,随机分为对照组、APS25 mg·L^(-1)组,利用倒置显微镜观察新... 目的:探究黄芪多糖(astragalus polysaccharide,APS)在缺氧条件下促进血管新生的作用机制与靶点。方法:于体外鼠尾胶基质上种植胸主动脉血管段构建新生血管三维培养模型,随机分为对照组、APS25 mg·L^(-1)组,利用倒置显微镜观察新生血管生长情况。体外培养大鼠脑微血管内皮细胞,以氯化钴400μmol·L^(-1)诱导24 h,形成可复制的缺氧模型,随机分为7组(n=6):对照组、缺氧模型组、APS25 mg·L^(-1)组、自噬激动剂雷帕霉素10 mg·L^(-1)组,阳性对照药巴弗洛霉素组20mg·L-1,APS加雷帕霉素组,APS加巴弗洛霉素组。然后利用CCK-8比色法检测各组细胞的增殖活性;蛋白印迹法检测各组细胞自噬相关蛋白Becline-1、LC3B、P62的表达。结果:(1)血管新生实验:与对照组相比,经APS干预新生血管数量明显增多(P<0.05)。(2)细胞实验:与模型组相比,经APS治疗内皮细胞增殖能力提高(P<0.05),Becline-1、LC3B蛋白表达量下调(P<0.01)。结论:在缺氧条件下,黄芪多糖可通过促进血管内皮细胞增殖,促进血管新生,其机制可能与下调内皮细胞Becline-1、LC3B的表达以抑制自噬发生有关。 展开更多

Methamphetamine(MA),an extremely addictive synthetic stimulant,has quickly spread to become the most frequently used illicit drug in China.People with a history of chronic and heavy MA use have a high possibility of exhibiting schizophrenia-like psychotic symptoms,mainly delusions of reference,auditory hallucinations and cognitive deficits.These emerging findings suggest MA use increases aggression and violence and that there is a correlation between MA use and violence.However,it is unclear how to assess the capacity of criminal responsibility in“MA-induced”psychosis and how to set clear boundaries between schizophrenia and MA-induced psychosis when only limited and inconsistent evidence is available.Furthermore,a final persuasive differential diagnostic method based on improved understanding of schizophrenia and MA-induced psychotic disorders has yet to be developed.This paper will evaluate the epidemiology,social harm,and forensic psychiatric assessment of MA users,propose a future direction for the differential diagnosis between MA-induced psychotic disorders and schizophrenia,and put forward some practical solutions to assess the capacity of criminal responsibility of defendants with drug-induced psychotic disorder. 展开更多
