We hypothesized that a chemically extracted acellular allogeneic nerve graft used in combination with bone marrow mesenchymal stem cell transplantation would be an effective treatment for long-segment sciatic nerve defects.To test this,we established rabbit models of 30 mm sciatic nerve defects,and treated them using either an autograft or a chemically decellularized allogeneic nerve graft with or without simultaneous transplantation of bone marrow mesenchymal stem cells.We compared the tensile properties,electrophysiological function and morphology of the damaged nerve in each group.Sciatic nerves repaired by the allogeneic nerve graft combined with stem cell transplantation showed better recovery than those repaired by the acellular allogeneic nerve graft alone,and produced similar results to those observed with the autograft.These findings confirm that a chemically extracted acellular allogeneic nerve graft combined with transplantation of bone marrow mesenchymal stem cells is an effective method of repairing long-segment sciatic nerve defects. 展开更多

Treatment for optic nerve injury by brain-derived neurotrophic factor or the transplantation of human umbilical cord blood stem cells has gained progress, but analysis by biomechanical indicators is rare. Rabbit models of optic nerve injury were established by a clamp. At 7 days after injury, the vitreous body received a one-time injection of 50 μg brain-derived neurotrophic factor or 1 × 10^6 human umbilical cord blood stem cells. After 30 days, the maximum load, maximum stress, maximum strain, elastic limit load, elastic limit stress, and elastic limit strain had clearly improved in rabbit models of optical nerve injury after treatment with brain-derived neurotrophic factor or human umbilical cord blood stem cells. The damage to the ultrastructure of the optic nerve had also been reduced. These findings suggest that human umbilical cord blood stem cells and brain-derived neurotrophic factor effectively repair the injured optical nerve, improve biomechanical properties, and contribute to the recovery after injury. 展开更多

BACKGROUND Gastric cancer(GC)is a common type of digestive cancer with high morbidity and mortality rates worldwide.Considerable effort has been expended in understanding the mechanism of GC development and metastasis.The current study therefore explores the involvement of microRNAs in the regulation of GC progression.AIM To explore the expression and function of miR-30e-3p in GC development.METHODS MiR-30e-3p was found to be downregulated in GC,with low levels thereof predicting poor outcomes among patients with GC.Functionally,we revealed that miR-30e-3p suppressed cell growth and metastatic behaviors of GC cells.Bioinformatics analysis predicted that THO complex 2(THOC2)was a direct target of miR-30e-3p,and the interaction between miR-30e-3p and THOC2 was further validated by a luciferase reporter assay.RESULTS Our findings revealed that knockdown of THOC2 inhibited the growth and metastatic behaviors of GC cells.After investigating signaling pathways involved in miR-30e-3p regulation,we found that the miR-30e-3p/THOC2 axis regulated the PI3K/AKT/mTOR pathway in GC.CONCLUSION Our findings suggest the novel functional axis miR-30e-3p/THOC2 is involved in GC development and progression.The miR-30e-3p/THOC2 axis could be utilized to develop new therapies against GC. 展开更多

Autoimmune pancreatitis(AIP)is a form of chronic pancreatitis that is categorized as type 1 or type 2according to the clinical profile.Type 1 AIP,which predominantly presents in a few Asian countries,is a hyper-IgG4-related disease.We report a case of IgG4-related AIP overlapping with Mikulicz’s disease and lymphadenitis,which is rare and seldom reported in literature.A 63-year male from Northeast China was admitted for abdominal distension lasting for one year.He presented symmetric swelling of the parotid and submandibular glands with slight dysfunction of salivary secretion for 6 mo.He had a 2-year history of bilateral submandibular lymphadenopathy without pain.He underwent surgical excision of the right submandibular lymph node one year prior to admission.He denied any history of alcohol,tobacco,or illicit drug use.Serological examination revealed high fasting blood sugar level(8.8 mmol/L)and high level of IgG4(15.2 g/L).Anti-SSA or anti-SSB were negative.Computed tomography of the abdomen showed a diffusely enlarged pancreas with loss of lobulation.Immunohistochemical stain for IgG4 demonstrated diffuse infiltration of IgG4-positive plasma cells in labial salivary gland and lymph node biopsy specimens.The patient received a dose of 30 mg/d of prednisone for three weeks.At this three-week follow-up,the patient reported no discomfort and his swollen salivary glands,neck lymph node and pancreas had returned to normal size.The patient received a maintenance dose of 10mg/d of prednisone for 6 mo,after which his illness had not recurred. 展开更多

AIM: To investigate the mechanism of calcyclin binding protein/Siah-1 interacting protein(Cacy BP/SIP) nuclear translocation in promoting the proliferation of gastric cancer(GC) cells. METHODS: The effect of Cacy BP/SIP nuclear translocation on cell cycle was investigated by cell cycle analysis. Western blot analysis was used to assess the change in expression of cell cycle regulatory proteins and proteasome-mediated degradation of p27Kip1. Coimmunoprecipitation(co-IP) analysis was performed to examine the binding of Cacy BP/SIP with Skp1. A Cacy BP/SIP truncation mutant which lacked the Skp1 binding site was constructed and fused to a fluorescent protein. Subsequently, the effect on Skp1 binding with the fusion protein was examined by co-IP, while localization of fluorescent fusion protein observed by confocal laser microscopy, and change in p27Kip1protein expression assessed by Western blot analysis.RESULTS: Cacy BP/SIP nuclear translocation induced by gastrin promoted progression of GC cells from G1 phase. However, while Cacy BP/SIP nuclear translocation was inhibited using si RNA to suppress Cacy BP/SIP expression, cell cycle was clearly inhibited. Cacy BP/SIP nuclear translocation significantly decreased the level of cell cycle inhibitor p27Kip1, increased Cyclin E protein expression whereas the levels of Skp1, Skp2, and CDK2 were not affected. Upon inhibition of Cacy BP/SIP nuclear translocation, there were no changes in protein levels of p27Kip1 and Cyclin E, while p27Kip1 decrease could be prevented by the proteasome inhibitor MG132. Moreover, Cacy BP/SIP was found to bind to Skp1 by immunoprecipitation, an event that was abolished by mutant Cacy BP/SIP, which also failed to stimulate p27Kip1 degradation, even though the mutant could still translocate into the nucleus.CONCLUSION: Cacy BP/SIP nuclear translocation contributes to the proliferation of GC cells, and Cacy BP/SIP exerts this effect, at least in part, by stimulating ubiquitin-mediated degradation of p27Kip1. 展开更多

The transplantation of polylactic glycolic acid conduits combining bone marrow mesenchymal stem cells and extracellular matrix gel for the repair of sciatic nerve injury is effective in some respects, but few data comparing the biomechanical factors related to the sciatic nerve are available. In the present study, rabbit models of 10-mm sciatic nerve defects were prepared. The rabbit models were repaired with autologous nerve, a polylactic glycolic acid conduit + bone marrow mesenchymal stem cells, or a polylactic glycolic acid conduit + bone marrow mesenchymal stem cells + extracellular matrix gel. After 24 weeks, mechanical testing was performed to determine the stress relaxation and creep parameters. Following sciatic nerve injury, the magnitudes of the stress decrease and strain increase at 7,200 seconds were largest in the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells + extracellular matrix gel group, followed by the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells group, and then the autologous nerve group. Hematoxylin-eosin staining demonstrated that compared with the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells group and the autologous nerve group, a more complete sciatic nerve regeneration was found, including good myelination, regularly arranged nerve fibers, and a completely degraded and resorbed conduit, in the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells + extracellular matrix gel group. These results indicate that bridging 10-mm conduit + bone marrow mesenchymal stem sciatic nerve defects with a polylactic glycolic acid cells + extracellular matrix gel construct increases the stress relaxation under a constant strain, reducing anastomotic tension. Large elongations under a constant physiological load can limit the anastomotic opening and shift, which is beneficial for the regeneration and functional reconstruction of sciatic nerve. Better regeneration was found with the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells + extracellular matrix gel grafts than with the polylactic glycolic acid conduit + bone marrow mesenchymal stem cells grafts and the autologous nerve grafts. 展开更多

Objective:To observe the differences in clinical therapeutic effect on acute attack of lumbar disc herniation(LDH) treated with electroacupuncture(EA) of different wave patterns so as to optimize EA wave pattern in the treatment of acute attack of LDH.Methods:A total of 108 patients were randomized into three groups,i.e.,a disperse-dense wave,an intermittent wave and a continuous wave group,36 cases in each one.In all of three groups,Shangliao(上髎BL31),Ciliao(次髎BL32),Zhongliao(中髎BL33) and Xialiao(下髎BL34) were selected bilaterally as the main acupoints,which is generally termed as Baliao(八髎BL31 to BL34,bilateral).In each group,the corresponding wave pattern was used in EA.The treatment lasted 20 min each time,once daily,totally for 10 days.Before and after treatment,the score of visual analogue scale(VAS) and the score of the lower back pain of Japanese orthopedic association(JOA) were observed in the patients of three groups and the overall therapeutic effect was evaluated.Results:After treatment,the VAS score was decreased and JOA score was increased in the patients of 3 groups,indicating statistical significance(all P <0.05).In comparison of VAS score before and after treatment among three groups,the score in the disperse-dense wave group was higher than the intermittent wave group and the continuous wave group,indicating statistical differences(both P <0.05).In comparison of JOA score before and after treatment among three groups,the score in the continuous wave group was higher than the intermittent wave group and the disperse-dense wave group,indicating statistical differences(both P <0.05).The total effective rate was 94.12% in the disperse-dense wave group,94.29% in the intermittent wave group and 97.14% in continuous wave group,without significant difference among groups(P> 0.05).Conclusion:Electroacupuncture of 3 different wave patterns all effectively relieves clinical symptoms of acute attack of lumbar disc herniation.Regarding pain release,electroacupuncture with disperse-dense wave is better than intermittent wave and continuous wave.Regarding the improvement of lumbar function,the effect of electroacupuncture with continuous wave is better than disperse-dense wave and intermittent wave. 展开更多

The fractures and kerogen,which generally exist in the shale,are signifcant to the CO_(2) huf-n-puf in the shale reservoir.It is important to study the efects of fractures and kerogen on oil recovery during CO_(2) huf-n-puf operations in the fracture-matrix system.In this study,a modifed CO_(2) huf-n-puf experiment method is developed to estimate the recovery factors and the CO_(2) injectivity in the fractured organic-rich shales and tight sandstones.The efects of rock properties,injection pressure,and injection time on the recovery factors and CO_(2) usage efciency in shales and sandstones are discussed,respectively.The results show that although the CO_(2) injectivity in the shale is higher than that in the sandstone with the same porosity;besides,the recovery factors of two shale samples are much lower than that of two sandstone samples.This demonstrates that compared with the tight sandstone,more cycles are needed for the shale to reach a higher recovery factor.Furthermore,there are optimal injection pressures(close to the minimum miscible pressure)and CO_(2) injection volumes for CO_(2) huf-npuf in the shale.Since the optimal CO_(2) injection volume in the shale is higher than that in the sandstone,more injection time is needed to enhance the oil recovery in the shale.There is a reference sense for CO_(2) huf-n-puf in the fractured shale oil reservoir for enhanced oil recovery(EOR)purposes. 展开更多

Clinical association of habitual breakfast skipping with cognitive decline and neurodegeneration among older adults

Background Unhealthy lifestyles have a considerable impact on the incidence of dementia.Skipping breakfast disturbs energy homeostasis and impairs brain function.In this study,we investigated the association between breakfast skipping and cognitive performance among community-dwelling adults.Methods We recruited 859 community-dwelling adults aged≥60 years from January 1 to December 31,2021.Participants’sociodemographic information and breakfast skipping habits were self-reported.Participants were followed up for 36 months and cognitive function was assessed using the Mini-Mental State Examination(MMSE)with an interval of 18 months.Trajectories of cognitive change were compared between individuals with and without breakfast skipping.To reduce the risk of bias owing to unmatched sample sizes between the groups,we conducted 1:1 propensity score matching(PSM)based on age,sex,education level,and ApoE genotype.Results At baseline and 18-month follow-up,no difference was found in MMSE scores between participants with and without breakfast skipping.However,those who habitually skipped breakfast had significantly lower MMSE scores than those who did not at 36-month follow-up.Individuals with habitual breakfast skipping had a steeper rate of cognitive decline than those without habitual breakfast skipping during follow-up.Breakfast skipping was a risk factor for longitudinal cognitive decline,defined as a decrease in MMSE scores of≥3,adjusted for age,sex,education,body mass index,ApoEε4 carrier status,hypertension,diabetes,and hyperlipidemia.At the last follow-up,participants who habitually skipped breakfast had significantly higher levels of ptau181 and NfL than those who did not.In the PSM cohort,similar findings were obtained regarding cognitive trajectories and plasma biomarkers.Conclusion Breakfast skipping was linked to an increased risk of long-term cognitive decline and neurodegeneration among older adults.The link between unhealthy dietary habits and cognitive decline may be attributed to a deficiency in neurorestoration resulting from inadequate energy consumption. 展开更多

Dear Editor, Triacylglycerols (triglycerides) (TAGs), as the major storage forms of energy, mainly are stored in adipocytes, myocytes, enterocytes, hepatocytes, and mammary epithelial cells in mammals, oilseeds in plants, and lipid droplets in microorgan- isms (Yen et al., 2008). Aside from energy storage, TAGs have essential functions in multiple physiological processes. In plants, TAGs are crucial for seed oil accumulation, germina- tion, and seedling development (Zhang et al., 2005, 2009). Notably, TAGs derived from plants and microorganisms could serve as the feedstock for biofuels production (Deng et al., 2009). Therefore, understanding of the molecular basis of TAGs bio- synthesis and storage is of considerable economic importance. 展开更多
