Shear-sliding mode(mode Ⅱ)fracture of rocks is a vital failure form in deep underground engineering.To gain deep insight into the anisotropic shear fracture behaviors of a typical shale under high normal stress condi...Shear-sliding mode(mode Ⅱ)fracture of rocks is a vital failure form in deep underground engineering.To gain deep insight into the anisotropic shear fracture behaviors of a typical shale under high normal stress conditions,a series of direct shear tests were conducted on double-notched specimens in three typical bedding orientations(i.e.,the arrester,divider,short-transverse orientations)and under five normal stresses.The modeⅡfracture toughness(K_(Ⅱc))is found to exhibit a significant 3D anisotropy.The maximum K_(Ⅱc)is obtained in the divider orientation,followed by those in the arrester and short-transverse orientations.In contrast,the 3D anisotropy in the critical modeⅡenergy release rate(G_(Ⅱc))is not as significant as that in K_(Ⅱc),and G_(Ⅱc)in the arrester orientation is quite close to that in the divider orientation.The anisotropy in the prepeak input energy accumulated during shearing is found to be exactly consistent with that in G_(Ⅱc),which has not been noted before.Furthermore,the anisotropies in the modeⅡfracture resistances will,unexpectedly,not be weakened by the high normal stress.Owing to the layered structures,tensile cracks are involved during the modeⅡfracture process,resulting in the formation of rough fracture surfaces.展开更多

By using MTS815 rock mechanics test system,a series of acoustic emission(AE) location experiments were performed under unloading confining pressure,increasing the axial stress.The AE space-time evolution regularities ...By using MTS815 rock mechanics test system,a series of acoustic emission(AE) location experiments were performed under unloading confining pressure,increasing the axial stress.The AE space-time evolution regularities and energy releasing characteristics during deformation and failure process of coal of different loading rates are compared,the influence mechanism of loading rates on the microscopic crack evolution were studied,combining the AE characteristics and the macroscopic failure modes of the specimens,and the precursory characteristics of coal failure were also analyzed quantitatively.The results indicate that as the loading rate is higher,the AE activity and the main fracture will begin earlier.The destruction of coal body is mainly the function of shear strain at lower loading rate and tension strain at higher rate,and will transform from brittleness to ductility at critical velocities.When the deformation of the coal is mainly plasticity,the amplitude of the AE ringing counting rate increases largely and the AE energy curves appear an obvious ''step'',which can be defined as the first failure precursor point.Statics of AE information shows that the strongest AE activity begins when the axial stress level was 92-98%,which can be defined as the other failure precursor point.As the loading rate is smaller,the coal more easily reaches the latter precursor point after the first one,so attention should be aroused to prevent dynamic disaster in coal mining when the AE activity reaches the first precursor point.展开更多
OBJECTIVE:To investigate the impact of Yemazhui(Herba Eupatorii Lindleyani,HEL)against lipopolysaccharide(LPS)-induced acute lung injury(ALI)and explore its underlying mechanism in vivo.METHODS:The chemical constituen...OBJECTIVE:To investigate the impact of Yemazhui(Herba Eupatorii Lindleyani,HEL)against lipopolysaccharide(LPS)-induced acute lung injury(ALI)and explore its underlying mechanism in vivo.METHODS:The chemical constituents of HEL were analyzed by ultra-high performance liquid chromatographyquadrupole time-of-flight mass spectrometry method.Then,HEL was found to suppress LPS-induced ALI in vivo.Six-week-old male Sprague-Dawley rats were randomly divided into 6 groups:control,LPS,Dexamethasone(Dex),HEL low dose 6 g/kg(HEL-L),HEL medium dose 18 g/kg(HEL-M)and HEL high dose 54 g/kg(HEL-H)groups.The model rats were intratracheally injected with 3 mg/kg LPS to establish an ALI model.Leukocyte counts,lung wet/dry weight ratio,as well as myeloperoxidase(MPO)activity were determined followed by the detection with hematoxylin and eosin staining,enzyme linked immunosorbent assay,quantitative real time polymerase chain reaction,western blotting,immunohistochemistry,and immunofluorescence.Besides,to explore the effect of HEL on ALI-mediated intestinal flora,we performed 16s rRNA sequencing analysis of intestinal contents.RESULTS:HEL attenuated LPS-induced inflammation in lung tissue and intestinal flora disturbance.Mechanism study indicated that HEL suppressed the lung coefficient and wet/dry weight ratio of LPS-induced ALI in rats,inhibited leukocytes exudation and MPO activity,and improved the pathological injury of lung tissue.In addition,HEL reduced the expression of tumor necrosis factoralpha,interleukin-1beta(IL-1β)and interleukin-6(IL-6)in bronchoalveolar lavage fluid and serum,and inhibited nuclear displacement of nuclear factor kappa-B p65(NF-κBp65).And 18 g/kg HEL also reduced the expression levels of toll-like receptor 4(TLR4),myeloid differentiation factor 88,NF-κBp65,phosphorylated inhibitor kappa B alpha(phospho-IκBα),nod-like receptor family pyrin domain-containing 3 protein(NLRP3),IL-1β,and interleukin-18(IL-18)in lung tissue,and regulated intestinal flora disturbance.CONCLUSIONS:In summary,our findings revealed that HEL has a protective effect on LPS-induced ALI in rats,and its mechanism may be related to inhibiting TLR4/NF-κB/NLRP3 signaling pathway and improving intestinal flora disturbance.展开更多
Mesenchymal stem cells (MSCs) are multipotent stem cells capable of differentiating into various cell types,including osteocytes,chondrocytes,adipocytes,myocytes,and tenocytes.However,the difficulty or failure in ex...Mesenchymal stem cells (MSCs) are multipotent stem cells capable of differentiating into various cell types,including osteocytes,chondrocytes,adipocytes,myocytes,and tenocytes.However,the difficulty or failure in expanding the mouse MSCs in vitro greatly hampered important research in animal models.The OP9,a stromal cell line from mouse bone marrow,has hematopoietic supportive capacity.Here,we report that the OP9 has the immunophenotype (CD45-,CD11b-,FLK-1-,CD31-,CD34-,CD44+,CD29+,Sca-1+,CD86-,and MHCII-) identical to canonical mouse MSCs.The expression of CD140a+,CD140b+,α-SMA+ and Calponin+ suggested the perivascular origin of OP9.Functionally,the OP9 had strong clonogenic ability and could be induced into osteocytes,chondrocytes and adipocytes.The lymphocyte transformation test (LTT) and mixed leukocyte reaction (MLR) showed that the OP9 could suppress T lymphocyte proliferation stimulated by nonspecific mitogens (PHA) or allogeneic lymphocytes (BALB/c T cells).Finally,the migration of OP9 could be efficiently induced by bFGF,IGF-1,IL-3,PDGF-BB,TGF-β1 and TGF-β3.In conclusion,the OP9 were bona fide MSCs,and such homogenous cell line will be helpful to delineate biological features of MSCs at the stem cell level.展开更多

The corona characteristic of the conductor test was done, and the corona onset voltage increase after the spraying of the anti-corona layer, the corona loss in the same voltage decrease, which could prove the excellent effect of improving the corona characteristic of the conductor.
