乙型肝炎肝硬化是由乙型肝炎病毒(hepatitis B virus,HBV)长期感染引起的一种慢性肝脏疾病,通常是乙型肝炎发展的严重后果。本研究通过分析乙型肝炎肝硬化患者的尿液代谢组学特征,旨在寻找潜在的代谢标志物,为乙型肝炎肝硬化患者的早诊...乙型肝炎肝硬化是由乙型肝炎病毒(hepatitis B virus,HBV)长期感染引起的一种慢性肝脏疾病,通常是乙型肝炎发展的严重后果。本研究通过分析乙型肝炎肝硬化患者的尿液代谢组学特征,旨在寻找潜在的代谢标志物,为乙型肝炎肝硬化患者的早诊早治提供依据。采用液相色谱-串联质谱代谢组学技术检测乙型肝炎肝硬化患者和健康体检人群的尿液代谢物,研究代谢组学特征,鉴定差异表达的代谢物,并分析其与疾病进展的关联。共筛选出229种组间差异代谢物,其中218种表达上调以及11种表达下调;结合生物学角度,利用京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析得到5条显著差异的代谢通路:抗坏血酸与醛酸代谢、氨基糖和核苷酸糖代谢、戊糖和葡萄糖醛酸的相互转化、丁酸代谢、萜类化合物和甾体化合物的生物合成(P<0.05);与健康体检人群相比,乙型肝炎肝硬化患者尿液中的苯腈磷、五氯苯酚、4-氯苯甲酸、辛二酸、氢化肉桂酸、2'-脱氧鸟苷5'-二磷酸含量显著增加,结合十折交叉验证后曲线下面积(area under the curve,AUC)均值不低于0.90。综合以上结果,通过非靶向代谢组学技术发现乙型肝炎肝硬化患者具有独特的尿液代谢组学特征。辛二酸、氢化肉桂酸、2'-脱氧鸟苷5'-二磷酸有望成为乙型肝炎肝硬化潜在的内源性代谢标志物。该方法也为尿液无创代谢标志物的发现提供方法参考。展开更多

Scanning electron microscope(SEM) images show that the surface of virgin sample is smooth, while the outdoor aged samples are all rough. According to the S(E) curves obtained by slow positron beam, in a depth of more than 1 μm, the S parameter of the sample aged at low-potential side keeps the same value with the virgin one;while the S values of the highpotential side aged sample remain rather low in a depth of about 5 μm. Thermo gravimetric analysis(TGA) results show that the sample in high potential side contains more inorganic constituents than that of other samples. The results are attributed to the strong electric field induced corona aging at high potential side of the composite insulator.展开更多
