摘要
目的:高效液相色谱法测定二氢齐墩果酸在大鼠血浆中的稳定性以及血浆蛋白结合率,以期为二氢齐墩果酸的临床药物开发研究提供依据并建立出相应检测方法。方法:以高效液相为检测手段,采用超滤法考察二氢齐墩果酸与大鼠血浆蛋白的结合情况。结果:二氢齐墩果酸在大鼠血浆中质量浓度为1~20 μg/ml时,血浆稳定性好,大鼠血浆蛋白结合率较高且无浓度依赖性。结论:建立的超滤法能够满足血浆蛋白结合率测定的要求,建立的高效液相检测方法满足二氢齐墩果酸的体外试验检测。
Objective: To determine the stability of dihydrogen oleanolic acid in rat plasma as well as the plasma protein binding rate in order to provide the basis for the development of clinical research on dihydro oleanolic acid. Methods: The HPLC method was used to detect the binding of dihydrogen oleanolic acid to rat plasma proteins by ultrafiltration. Results: Dihydro-oleanolic acid showed good plasma stability when the concentration of dihydroxyaluminum in rat plasma was 1 - 20 μg/ml, while plasma protein binding rate was high and concentration-independent in rats. Conclusion: The established ultrafiltration method can meet the requirement of determination of plasma protein binding rate. The established HPLC method meets the requirement of dihydromyric acid assay in vitro.
