家蚕核型多角体病毒四角形多角体的研究 被引量:3

STUDIES ON BOMBYX MORI NUCLEAR POLYHEDROSIS VIRUS WITH TETRAGONAL POLYHEDRON

摘要 BmNPVt是从BmNPVh中筛选出的一株四角形多角体病毒突变株。研究表明:BmNPVt的潜伏期、致病力、寄生部位、细胞病变同BmNPVh无甚差异;BmNPVt和BmNPVh之间存在干扰现象;Bm-NPVt的增殖类似于BmNPVh,但囊膜的形成方式两者之间不同。四角形多角体表面粗糙,具有小孔;它的外层电子密度较高,有一厚度为1500A的中间层。多角体蛋白晶格间距离为48A,分子量为29000dolt。同六角形多角体蛋白相比,四角形多角体蛋白的Glu含量较低,Tyr含量相对较高,胰酶酶切图谱两多角体蛋白存在差异。此外,多角体蛋白中Cu、Fe的含量和对酸碱溶解性,两多角体间也存在差异。BmNPVt为杆状,大小为330—340×85nm,纯病毒具有典型的杆状病毒紫外吸收特性,BmNPVt—DNA的变性温度为83℃,分子量为6.94×10~7dolt。镇江株、苏州株、日本株之间病毒DNA的EcoRI酶切图谱存在差异。 BmNPVt was a virus mutant with tetragonal polyhedron isolated from the wild-typed Bombyx mori nuclear polyhedrosis virus (BmNPVh). The results showed that its occulted stage, pathogenicity to the silkworm, the target tissue and the cell pathological changes were no diverse from BmNPVh. Interference was observed between BmNPV, and BmNPVh. In the multiplication, BmNPV, was similar to BmNPVh, but different from each other in formation of the envelope. The tetragonal polyhedron's surface was very rough where little neck pores can be found,the electron density of tetragonal polyhedral exteral membrance was higher, there was a intermediate layer being of about 1 500 A ?The distance between polyhedral protein crystal lattile was 48A, The polyhedral protein molecular weight was 29 000 dolt. In comparison to hexagonal polyhedron, the content of Glu residue of the tetgagonal polyhedron was lower, the Tyr was higher. There were differences in both polyhedron trypsin peptide-maps. In the contents of copper and iron elements, and stablization to acidic and alkali solution, there were also differences between both polyhedrons. BmNPV, was baculiform with size about 330 - 340 × 85nm, pured virions had the typical ultra-violet absorption of baculoviruses. The melting temperature of BmNPVt-DNA was 83 ℃ , the molecular weight of viral DNA was 6. 94 × 107 dolt. BmNPV in EcoRI restriction pattern among Suzhou strain, Zhen-jiang strain and Japan strain was slighgtly diverse.
