摘要
目的 建立脑卒中后抑郁状态动物模型。方法 采用Wistar成年大鼠 ,去除大鼠脑左侧皮层血管建立脑缺血模型 ,在此基础上加用孤养法和慢性不可预见的温和性应激方法 ,造成脑卒中后抑郁状态动物模型。结果 造模后即予行为学测试 ,模型组大鼠蔗糖水饮用量与正常组相比明显减少 ,有显著差异 (P <0 0 1) ,反映模型组大鼠兴趣和快感缺乏。旷野试验中模型组大鼠水平及垂直得分均小于正常组 ,有显著性差异 (P <0 0 1) ,反映模型组大鼠活动性减少 ,兴趣缺乏。模型组大鼠游到终点的时间延长 ,错误次数增加 ,与正常组相比 ,有统计学意义 (P <0 0 5或 0 0 1) ,反映模型组大鼠的学习和记忆能力下降。结论 脑卒中后抑郁状态动物模型快感缺乏 ,活动减少 ,能充分模拟脑卒中后抑郁患者的核心症状 ,实验重复性较好 ,是研究脑卒中后抑郁的理想模型。
Objective To establish the animal model of post-stroke depression. Method Adult Wistar rats were adopted in the experiment. First, the model of cerebral ischemia was set up by removing the blood vessels in the left cortex of the rat, and then the model rats were separately raised and put into chronic unpredictable mild stress, in order to induce the model of cerebral ischemia into the model of post-stroke depression. Results After undergoing the model-inducing procedures, the rats were examined by behavioral tests. It was found that the amount of sugar-water consumption by the rats in the model group was much less than that by the rats in the normal group (P<0.01), implying the absence of interest and anhedonia of the rats in the model group. In the open-field test, both the crossing time and rearing time were found to be much less in the model group than those in the normal group (P<0.01), implying the underactivity and absence of interest of the rats in the model group. In the water maze test, it was found that the rats in the model group needed more time to reach the end-point and had more errors than the rats in the normal group did (P<0.05 or 0.01), implying the decreased ability to learn and memorize of the rats in the model group. Conclusion With good repeatability, the anhedonia and underactivity found in the rats with post-stroke depression may represent the core symptoms in the patients of post-stroke depression, and the rat model of post-stroke depression is an ideal model for post-stroke depression research.
