背景与目的:肿瘤抑制基因PTEN、间隙连接蛋白43(connexin43,Cx43)及血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)的表达在肿瘤发生和进展中发挥重要的作用。本研究目的是观察肝细胞癌(hepatocellularcarcinoma,HCC)中PTEN、Cx43及VEGF蛋白的表达及其关系,探讨它们在HCC发生和发展中的作用。方法:免疫组化链酶素抗生物素-过氧化物酶(SP)法研究47例HCC组织中PTEN、Cx43及VEGF的表达情况。结果:PTEN、Cx43、VEGF在HCC组织中总的阳性率分别为76.6%、42.6%、70.2%。随着肿瘤病程进展,PTEN和Cx43阳性率降低(P均<0.05),而VEGF阳性率显著增高(P=0.001)。与肝内血管癌栓组相比,PTEN在无肝内血管癌栓组的阳性率显著增高(P=0.006)。肝硬化组Cx43阳性率明显低于无肝硬化组(χ2=4.7135,P=0.03)。PTEN阳性率在肿瘤大小、癌组织分化程度及肝硬化背景组无显著性差异。Cx43阳性率在肿瘤大小、癌组织分化程度及肝内血管瘤栓组无显著性差异。VEGF阳性率在肿瘤大小、癌组织分化程度、肝硬化及肝内血管癌栓组无显著性差异。相关分析表明:PTEN阳性率与Cx43阳性率正相关(CramersV=0.3949,P=0.023);PTEN阳性率与VEGF阳性率负相关(CramersV=0.3937,P=0.024);Cx43阳性率与VEGF阳性率无明显相关(CramersV=0.3072,P=0.
BACKGROUND &OBJECTIVE: Previous studies showed that expression of phosphatase and tensin homology deleted on chromosome ten (PTEN), gap junction p rotein connexin 43 (Cx43) and vascular endothelial growth factor (VEGF) may play an important role in tumor occurrence and progression. This study was designed to investigate the relationship among the protein expression of PTEN, Cx43, and VEGF in carcinogenesis and progression of hepatocellular carcinoma. METHODS: The expression of PTEN, Cx43 and VEGF were determined in 47 cases of HCC by strepta vidin peroxidase immunohistochemistry. RESULTS: In the hepatocellular carcinoma, the total positive rates of PTEN, Cx43 and VEGF were 76.4%, 42.6%, and 70.2% , respectively. With progression of tumor course, PTEN and Cx43 positive rates d ecreased (P< 0.05), but the VEGF positive rate significantly increased (P=0.001) . In comparison with the group of intrahepatic vascular embolism, PTEN positive rate increased apparently in the group without intrahepatic vascular embolism (P =0.006). Cx43 positive rate was lower in the group with cirrhosis background tha n that in the group without cirrhosis background (r size, differentiation degree in cancerous tissue and intrahepatic vascular embolism, VEGF positive rate in tumor size, differentiation degree in cancerous tissue, cirrhosis background and intrahepatic vascular embolism. Correlation ana lysis revealed that PTEN positive rate was positively correlated with Cx43 posit ive rate (Cramers V=0.394 9, P=0.023), negatively correlated with VEGF positiv e rate (Cramers V=0.393 7,P=0.024), but no significant association was observ ed between Cx43 positive rate and VEGF positive rate (Cramers V=0.307 2, P=0.0 64). CONCLUSION: The aberrant expression of PTEN, Cx43, and VEGF may play a role in occurrence, progression, and intrahepatic metastasis of the hepatocellular c arcinoma. Determining the expression of PTEN, Cx43, and VEGF may contribute to s ynthetically evaluating the biological behavior of hepatocellular carcinoma.
