摘要
目的 探讨丙烯腈对雄性小鼠生精过程的影响。方法 用腹腔注射染毒的方法 ,对 5 0只雄性未成年小鼠及 2 5只成年小鼠分别给丙烯腈 1 2 5、2 5、5mg kg ,染毒 5天 ,于初次染毒后 35天处死动物 ,应用流式细胞术检测睾丸生精细胞DNA含量及细胞周期的变化。结果 (1)染毒组未成年小鼠的单倍体细胞 (1C)的百分含量与阴性组比较有减少 ,但无统计学差异 ;染毒组成年小鼠的单倍体细胞 (1C)百分含量与阴性组比较有减少 (P <0 0 5 ) ;成年和未成年小鼠的凋亡细胞的百分含量与阴性组比较有增加 (P <0 0 5 )。 (2 )细胞周期分析显示 ,染毒组未成年小鼠的S期细胞比例增加 (P <0 0 5 ) ,G0 G1 期细胞比例减少 (P <0 0 5 ) ;染毒组成年小鼠的G2 M期细胞比例增加 (P <0 0 5 ) ,G0 G1 期细胞比例减少 (P <0 0 5 )。结论 丙烯腈对雄性小鼠的生精过程有抑制作用。
Objective To observe the effect of acrylonitrile on the spermatogenesis in mice. Methods 50 male pubertal mice and 25 male adult mice were exposed to acrylonitrile (1.25,2.5,5mg/kg) by intraperitoneal injection for 5 days, and detected the DNA content and cell period of spermatogenic cells by flow cytometry after 35 days. Results (1) The percentage of 1C cell in the exposed pubertal mice groups was lower than in the control group, respectively. The percentage of 1C cell in the exposed adult mice groups was lower than in the control group, respectively (P<0.05). The percentage of apoptotic cell in pubertal and adult mice was higher than in the control group, respectively (P<0.05). (2) The percentage of cells in S-phase in the exposed pubertal mice groups was higher than in the control group, respectively(P<0.05), G 0/G 1 phase were lower(P<0.05). The percentage of cells in G 2/M phase in the exposed adult mice groups was higher than in the control group, respectively(P<0.05), G 0/G 1 phase was lower(P<0.05). Conclusion There are restrained effects of acrylonitrile on the spermatogenesis in mice.
