摘要
以乙酰丙酮 (1)为原料 ,经溴代、酯化反应得到 3 苯甲酰氧基 2 ,4 戊二酮 (2 ) ,2与甲酸乙酯缩合、脱水闭环、酯水解得麦芽酚 (3) ,总收率 5 0 .4 %。 3与α 溴代四乙酰化葡萄糖 (4 )经糖甙化、脱乙酰基得麦芽酚 3 O β D 葡萄糖甙 (5 ) ,收率 6 0 .8% ,IR ,1HNMR和13 CNMR确证 5的葡萄糖为 β 构型。中间产物 2无须纯化 ,可实现“一锅”反应合成
Maltol(3) was synthesized by using 'one-pot' method under mild reaction conditions from acetylacetone(1) in an overall yield of 50.4%. Acetylacetone was at first brominated and esterificated to give 3-penzoyloxy-2, 4-pentanedione(2), which condensated with ethyl formate, then dehydrated and hydrolyzed to obtain 3. Maltol-3-O-β-D-glucoside(5) was also synthesized by glucosidation reaction of 3 with α-bromotetraacetylglucose(4) and then deacetylation in 60.8% yield. Configurati of 5 was confirmed by IR, 1H NMR and 13C NMR to be β-anomer .
