摘要
目的建立一种膜片钳技术(patchclamptechnique,PCT)的成年大鼠海马CAI区锥体细胞(adultrathippocampalCAIpyramidalneuron,ARHCPN)的急性分离方法。方法采用酶加机械分离的方法制备ARHCPN,用相差显微镜观察其形态特征,并用PCT的细胞贴附模式记录L-型Ca2+通道的单通道电话动。结果所分离的ARHCPN不仅形成学特征保存完好,而且保存了L型Ca2+通道的电生理学特性。结论成功地建立了一种简单、快速、可靠的适用于膜片钳单通道记录的ARHCPN的急性分离方法。
Objective:To establish a method of acute isolation of adult rat hippocampal CAI pyramidal neurons(ARHCPN)suitable for patch clamp technique(PCT).Methods:ARHCPN were obtained by a combi-nation of mechanical and enzymatic means.The isolated neurons were identified under a phasic contrast mi-croscope,and cell-attached mode was used to record L-type calcium channel currents.Results:The isolated ARHCPN exhibited the well-kept proximal dendritic structures with clean surface,and preserved electrophysi-ological properties of L-type calcium channel.Conclusions:A simple,fast and credible method of acute iso-lation of ARHCPN suitable for single channel recording is successfully founded.
