摘要
参照Hascall实验方法建立了牛关节软骨体外培养,通过组织学观察,S掺入率测定及已糖醛酸测定的方法证明,在加入20%小牛血清的DMEM培养液中,培养7天后,软骨蛋白多糖的代谢达到了稳态。这为研究关节软骨蛋白多糖代谢及其影响因素提供了一个较好的实验模型。
This paper reports the establishment of bovine articular cartilage explant culture in accord with the methods of Vincent C Hascall. The determination of 35 S sulfate incorporation and hexuronate content was employed to monitor the proteoglycan metabolism state in culture. The growth condition of cartilage pieces was observed with upside and light microscope. It was found that a steady-state metablism of proteoglycan was reached on the 7th day of culture. This provides a profitable experimental model for the study of proteoglycan metabolism and regulation of articular cartilage.
