SAG3基因在造血干细胞移植患者弓形虫感染诊断中的应用 被引量:4

摘要 目的 探讨造血干细胞移植患者弓形虫感染的快速、特异、敏感的诊断方法。方法 以弓形虫SAG3基因为靶基因 ,采用聚合酶链反应法 (PCR)对 5 6例造血干细胞移植受者及 2 0名正常对照者进行检测 ,同时以酶联免疫吸附试验 (ELISA)进行弓形虫抗体IgG、IgM的血清学检测。 结果 以PCR和ELISA检测弓形虫SAG3基因片段和其抗原 ,阳性分别为 10例及 7例 ,阳性率分别为17.9%和 12 .5 % ,其中PCR检测阳性、而ELISA检测阴性的 3例 ,经病原学检测 ,均证实有弓形虫感染 ;2 0名正常对照者的检测结果均为阴性。结论 体外扩增弓形虫SAG3基因的方法诊断弓形虫感染具有准确、快速、特异和敏感的优点 ,可作为造血干细胞移植患者弓形虫感染的诊断方法之一。 Objective To establish a rapid, specific and sensitive diagnostic technique for the human T. gondii infection with hematopoietic stem cell transplantation and discuss its clinical significance. Me- thods Fifty-six patients subject to hematopoietic stem cell transplantation were detected with ELISA and PCR. Results Among 56 recipients of hematopoietic stem cell transplantation, 7 were positive for T. gondii antigen and 10 were positive for SAG3 gene fragment respectively with the positive rate being 14.3 % and 17.8 % in the ELISA and PCR screening respectively. Twenty healthy people were negative for anti-Toxo antibody.Conclusion PCR is an accurate, relatively rapid, sensitive and specific method for detecting SAG3 gene of T. gondii, and can be considered a valuable additional tool for identification of T. gondii infections.
