摘要
用亚型选择性标记配基~3H-CGP_(12177)观察了急性缺血时大鼠心肌β_1AR的变化。急性缺血60分钟后,大鼠β_1AR密度为 97.5 fmol/mg蛋白,较正常对照组(49.6fmol/mg蛋白)增高 96.6%(P<0.01);较手术对照组(61.7fmol/mg蛋白)增高58.5%(P<0.05)。
The changes of β_1-adrenergic receptor (β_1 AR) during acute myocardial ischemia were determinedusing radioligand binding techniques (~3H-CGP 12177, β_1-selective antagonist) in Wistar rat hearts(male, 200-360 g). The Wista rats were divided into three groups.(1) Acute myocardial ischemia group (n=8):the rats were anesthetized with ether and the leftcoronary artery was ligated under left thoractomy. The rats sacrificed after electrocardiography showedS-T segment elevations characteristic of ischemia for 60 min. The left ventricle was isolated and car-diac membrane was prepared; (2) Sham-operated group (n=7): and (3) normal group (n=12) ratswere used as controls.The results showed that the β_1 AR density in the ischemic group was incressed (Bmax 97.5±13.1 fmol/mg protein)to a level much higher than that in the control groups (Bmax 61.7±8.9 fmol/mgprotein, P<0.05, sham-operated group, and Bmax 49.6±5.1 fmol/mg protein, P<0 .01,normal group).The Kd values were not significantly different among the three group (15.6±4.2, 12.2±2.7 and11.0±1. 6 nmol/L, respectively).The results indicate that β_1 AR levels are increased during acute myocardial ischemia, and thisfinding may have some significance in the clinic.
