摘要
本文应用热处理与茎尖分生组织培养相结合等方法,成功地脱除了枣疯病类菌原体(MLO),并筛选出适合茎尖分生组织培养的培养基,在国内外未见报导.同时,首次应用了迪纳氏染色、苯胺蓝染色、DAPI染色等组织化学方法检测枣疯病类菌原体(MLO),试验证明DAPI染色法灵敏度高.特异性强,对枣树苗木的检疫、抗枣疯病品种的筛选具有重要意义.
Heat treatment and apical meristem culture are used in eliminating Mycoplasma-Like Organism (MLO) from Jujube Whitches'Broom (JWB) infecting Zizyphus Jujuba Mill and Pathogen-free Plantlets are obtained.The optimal medium for the apical meristem culture is WPM or MS medium without any plant regulators.Later,the plantlet was examined under electron microscope and by cytochemical methods to be MLO-free.By fluoresecnce microscopy DAPI(4!6-diamidino-2-phenylindole-2 HCl) Stain is a more specific and sensitive method than aniline bule stain,and better than Dienes' stain by light microscopy.Both Jujube apical meristem culture and applying of cytochemical methods in detecting JWB MLO are reported for the first time.
