摘要
采用PCR克隆技术对黑熊四川亚种线粒体DNA cytb基因进行克隆、测序。黑熊四川亚种线粒体DNA cytb基因的全序列为1140 bp,CG含量为46.23%。与AB020910和U23558两个黑熊cytb序列的变异共有61处.其中T-C转换共有36处.A—G转换共有23处。仅有2处发生T-A颠抉,碱基突变率为5.35%.其中转换率96.72%,颠换率为3.28%。密码子第3个碱基发生变异的有48个。
An Asian black bear (Ursus thibetanu mupinensis) mtDNA Cytb gene was amplified, cloned and sequenced. The full length of the black bear mitochondrial DNA Cytb gene (1140bp) is obtained, which C + G content accounts for 46.25 % . The comparison of the nucleotide sequence obtained in this experiment among the sequences downloaded from GenBank( AB020910 and U235S8) showed 61 mutations , including 36 transpositions of T - C, 23 A - G and 2 T - A. The rate of base mutation and transposition of T-C ,A-G&T-Ais5.35% and 96.72 % , 3.28 % respectively. 48 mutation sites were found at the 3rd of base of codons.
