摘要
本文报道自行合成了睾酮-羧甲基肟-异鲁米诺作为睾酮的化学发光标记物,发光检测限为0.21pg/tube.并用此标记物建立了睾酮的化学发光免疫测定法.本法的线性范围为12.5~200pg/tube,灵敏度为7.4pg/tube,平均回收率为101.63%,批内变异系数(CV)为5.86%(Man,n=8)及13.86%(woman,n=7),批间为13.64%(Man,n=5)及10.02%(Woman,n=7).免疫法标准曲线的各项指标都经测定,变异系数在2.3%~21.6%范围(n=7~9).用本法与放射免疫法同时对22份正常男女样品进行测定,睾酮含量相符.
Testosterone is an important hormone in human. The measurement of testosterone may provide a useful index in assessment of gonadal function in men and the androgenic status of women. An immunoassay procedure for determination of testosterone in human serum is described which utilizes lightemitting tracer synthesized by coupling aminobutylethylisoluminol(ABEI) co-valently to testosterone 3-carboxymethyloxime (T-3-CMO) with an anhydroreaction. The assay utilizes dextran coated charcoal for the separation of bound and free fraction of the ligand. The sensitivity of the assay is 7. 4pg/tube. The intra and interassay variations are 5. 86%(Man,n = 8) or 13. 86% (Woman, n = 7) and 13. 64% (Man,n = 5) or 10. 02%(Woman,n = 7) respectively. The linearity is covered from 12. 5 to 200 pg/tube for serum sample. The cross-reactivity is similar to that in RIA. The correlation for CIA and RIA is CIA = 1. 048 RIA + 51. 44,r=0. 943 for measuring 22 serum samples, but health hazards and radioactive waste disposal problems can be avoided in CIA.
