摘要
实验选择了不同浓度的rhG-CSF(0.015~0.50μg/ml)体外作用于HL-60细胞5天,检测其细胞数、~3H-TdR掺入量和细胞内cAMP水平。结果表明,细胞数和~3H-TdR掺入量都随rhG-CSF浓度增加而明显下降。在浓度达0.125μg/ml时,这种抑制作用基本达坪值;而细胞内cAMP水平随rhG-CSF浓度增加明显增高,呈剂量依赖性。推测rhG-CSF是通过诱导细胞分化来实现对HL-60细胞增殖的抑制作用。
The HL-60 cells were cultured in the presence of recombinant human granulocyte colony-stimulating factor (rhG-CSF) (0. 015-0. 50μg/ml) for 5 days and then the cell numbers, 3H-TdR incorporation and cAMP levels in the cells were estimated. The results showed marked decrease in cell numbers and 3HTdR incorporation along with the increasing rhG-CSF concentration. The inhibitory effect reached a plateau at 0. 125μg/ml of rhG-CSF. Moreover, the cAMP content in HL-60 cells increased significantly with the increase of rhG-CSF concentration in a dose - dependent manner. It is speculated that rhG-CSF suppresses the HL-60 cell proliferation through inducing cell differentiation.
