摘要
目的:探讨蛋白酪氨酸磷酸酶SHP-1蛋白在γ射线体外诱发的白血病小鼠胸腺细胞中的变化特点。方法:建立γ射线体外诱发BALB/c小鼠白血病模型。实验分为癌变组、未癌变组及对照组,从各组小鼠获取胸腺细胞,应用Western印迹、免疫沉淀法及生物化学方法检测各组胸腺细胞SHP-1蛋白质的表达、酪氨酸磷酸化水平及酶活性。结果:癌变组SHP-1蛋白的表达明显高于对照组及未癌变组(P<0.01),但各组SHP-1的酪氨酸磷酸化水平及酶催化活性差异不显著(P>0.05),且其酪氨酸磷酸化水平与酶活性间存在明显相关性。结论:在γ射线诱发的白血病小鼠的胸腺细胞中,SHP-1蛋白表达明显升高,但可能存在酪氨酸磷酸化障碍,导致其活性降低,即表现出SHP-1蛋白表达与其活性分离现象,提示辐射致癌中SHP-1蛋白可能存在功能障碍。
Objective:To investigate the expression of SH2 domain containing protein tyrosine phosphatase SHP-1 in thymus cells of γ-ray irradiation-induced leukemia mice. Methods : BALB/c mice leukemia model was developed by γ irradiation and mice were randomly divided into cancerization groups, uncancerization groups and controls. The expression, tyrosine phosphorylation and catalysis activity of SHP-1 were detected with Western blotting, immunoprecipitation and biochemistry methods in thymus cells. Results: The expressions of SHP-1 in cancerization groups were significantly higher than those in uncancerization groups and controls, but the tyrosine phosphorylation and catalysis activity of SHP-1 had no significant difference among them(P>0. 05). There were close correlation between tyrosine phosphorylation and catalysis activity of SHP-1. Conclusion: The expression of SHP-1 are significantly increased in thymus cells of leukemia mice induced by γ-ray irradiation, but there may be an obstacle to tyrosine phosphorylation of SHP-1 in cancerization mice which results in a decline of catalysis activity of SHP-1. The separated activities between expression of protein and catalysis activity suggest the dysfunction of SHP-1.
