摘要
目的:建立端粒酶反转录酶(humantelomerasereversetranscriptasehTERT)基因稳定转染的成骨细胞,构建组织工程骨。方法:克隆hTERTcDNA编码区,构建hTERT表达载体,培养并鉴定成骨细胞,确定成骨细胞G418耐受含量,hTERT基因稳定转染成骨细胞并传代培养,制备组织工程骨,植入动物体内,观察骨折愈合的情况和进程。结果:成功的克隆了hTERTcDNA编码区,构建了hTERT表达载体并将hTERT稳定转染于成骨细胞中。经hTERT稳定转染的成骨细胞其构建的组织工程骨治愈骨折愈合的情况和进程明显好于未经转染的成骨细胞组织工程骨。结论:经hTERT稳定转染的成骨细胞,能有效的延长成骨细胞的寿命防止细胞老化,并保持成骨细胞的功能特性。
AIM:To stably transfect osteoblasts with human telomerase reverse transcriptase (hTERT) so as to constitute tissue engineered bone. METHODS:cDNA encoding hTERT was cloned and hTERT expression vector was constituted for culture and differentiation of osteoblasts.Concentration of osteoblast G418 resistance to was measured.Tissue engineered bone was made on the basis of hTERT transfected osteoblasts,and then implanted into animal for observation of fracture healing. RESULTS:After successful cloning of cDNA encoding hTERT, construction of hTERT expression vector and stable transfection of hTERT into osteoblast,tissue engineered bone based on hTERT transfected osteoblast had a better effect on fracture healing than that without transfecton. CONCLUSION:hTERT is effective in prolonging life span of transfected osteoblast,preventing cell aging and maintaining the functional characteristics of osteoblast.
