低氧预适应增加小鼠海马组织内神经细胞黏附分子的表达 被引量:4

摘要 观察重复性低氧对小鼠海马组织内神经细胞黏附分子 (neuralcelladhesionmolecule ,NCAM)表达的影响 ,复制低氧 0次 (H0 ) ,1次 (H1 ) ,4次 (H4 )组模型小鼠 ,用半定量反转录 -聚合酶链反应 (semi quantitativereversetranscriptionandpolymerasechainreaction ,SqRT PCR)和WesternBlot技术 ,检测小鼠海马组织内NCAM的mRNA和蛋白表达变化。实验发现H1组海马组织内NCAMmRNA显著降低 ,(P <0 0 5 ,n =8) ;H4组海马组织内NCAM的mRNA表达水平回升显著高于H1组 (P <0 0 5 ,n =8)。NCAM 1 80ku亚型蛋白在H1组显著下降 (P <0 .0 5 ,n =8) ;H4组显著回升 ,高于H0和H1组 (P <0 0 5 ,n =8)。 The effect of repetitive hypoxia on expression of NCAM in the hippocampus was observed in mice. The level of NCAM expression in the hippocampus was determined and compared among the groups H0(normal control,without hypoxic exposure), H1(exposure to hypoxia once)and H4(exposure to hypoxia for 4 runs) by semi quantitative reverse transcription and polymerase chain reaction ( SqRT PCR)as well as Western Blotting.NCAM mRNA in the hippocampus was significantly decreased( vs H0, P <0 05,n=8) in group H1, while the decrease was significantly relieved( vs H1, P <0 05,n=8)in group H4. NCAM 180kD isoform protein was significantly decresed in group H1 ( vs H0, P <0 05 ,n=6) and increased in H4( vs H0,H1, P <0 05 ,n=6).The increase of NCAM may play a role in the hypoxic preconditioning.
