野生小豆种质资源遗传多样性的RAPD分析 被引量:9

Study on Germplasm Resources of Wild Adzuki Bean( V.angularis var.nipponensis)Based on RAPD Analysis

摘要 利用RAPD分子标记技术对16份不同来源地的野生小豆种质进行基因组DNA多态性分析,25个随机引物共扩增出240个DNA片段,其中197条谱带表现出多态性。对基于数字化处理后的多态性谱带进行聚类,16个野生小豆种质可划分为4个类群:日本类群、中国类群、不丹类群和尼泊尔类群,材料的归组与其来源地有显著的相关性。 It is very important to detect the genetic diversity of wild adzuki bean (V. angularis var.nipponensis)for understanding the origin, evolution and diffusion of adzuki bean and giving references to the development and utilization of wild adzuki bean germplasm. Random amplified polymorphic DNA (RAPD) was used to analyze the genetic diversity of 16 wild adzuki bean accessions in the present study. Twenty-five random primers was used to amplify the total accessions. Among the total 240 RAPD bands identified, 197 bands(82. 08%)were polymorphic. Four groups(Chinese group ,Japanese group, Bhutanese group and Nepal group) were clustered by the digital data of polymorphic bands. There was obvious association between RAPD variation and different adzuki bean geographical regions.
