高产超氧化物歧化酶菌株发酵条件的研究 被引量:5

摘要 从 1 348株芽孢杆菌 (Bacilluscereus)中分离和选育出高产超氧化物歧化酶 (Superoxidedismutase)菌株C32 8,并进行了产酶条件的研究。结果表明 ,该菌株最佳产酶的最适温度为 30~35℃ ,培养基最适起始 pH为 8.0~ 8.5。金属离子Cu2 + 、Mn2 + 、Zn2 + ,表面活性剂吐温 80、司班80、琥珀酸钠和无机盐CaCl2 、CaCO3 对产酶有抑制作用。适宜浓度的K2 HPO4、EDTA、金属离子Fe3 + 和表面活性剂NaAC对产酶有明显的促进作用。发酵培养用 2 5 0mL三角瓶以 30mL装液量酶活力最高。适宜种龄为 8~ 1 2h ,适宜接种量为 0 .9%~ 3% ,最佳接种量为 1 %~ 2 %。 A high yielding C328 strain for producing superoxide dismutase was selected from Bacillus cereus . The fermentation conditions for the SOD enzyme production of the strain were examined. The optimal temperature and pH for the enzyme formation were 30~35℃ and pH 8.0~8.5 respectively. The enzyme activity was obviously inhibited by metal ion Cu 2+ 、Mn 2+ 、 Zn 2+ ,sufactants Tween 80 and Span 80, sodium succinate and salts CaCl 2,CaCO 3. The enzyme formation was enhanced by proper concentration K 2HPO 4, EDTA, metal ion Fe 3+ and surfactants NaAC. The bacterium cultured in 250ml flask with 30ml of the medium in it was best.
