摘要 目的 :探讨过量氟对大鼠破骨细胞 (osteoclasts,OCs)活性的影响。方法 :对经饮水投氟 2 0周的大鼠胫骨进行常规组织切片 ,并对其进行 HE及抗酒石酸酸性磷酸酶 (TRAP)染色 ,观察染氟大鼠胫骨 OCs的变化。同时提取股骨干骺端的总蛋白质 ,应用蛋白质印迹法 (Western blotting)观察长骨干骺端基质金属蛋白酶 9(MMP- 9) (又称明胶酶 B)的变化。结果 :染氟组大鼠胫骨各个包被及干骺端 OCs增多 ,骨皮质密质骨松质化。常食加氟组、偏食对照组 (低钙偏食饲料组 )、偏食加氟组 TRAP阳性细胞数较对照组均有增加 ,但差异无显著性 (P>0 .0 5 )。Western blotting结果显示 ,常食加氟组大鼠干骺端 MMP- 9蛋白表达增高 ,与常食对照组比较差异有显著性 (P<0 .0 5 )。偏食加氟组与偏食对照组比较差异也有显著性 (P<0 .0 5 )。且二者与常食对照组差异均有显著性 (P<0 .0 5 )。结论 :过量氟造成骨转换增高 ,破骨性骨吸收增强 ,低钙偏食是氟骨症的促发因素。氟可通过增强 MMP- 9的蛋白质表达来加强 Objective To investigate the effects of excessive fluoride on osteoclastic activity in rats.Methods The tibia slices of chronic fluorotic rats were stained with HE and tartrate resistant acid phosphatase (TRAP),and the changes of osteoclasts were observed;At the same time,the protein was extracted from metaphysis of femurs in chronic fluorotic rats,then the expression of matrix metalloproteinase 9 (MMP 9) was investigated using Western blotting technique. Results The number of osteoclasts in tibia envelope and metaphysis increased in fluoride groups and compact bone of the cortex became porosity. Compared with stock diet group,the number of TRAP positive osteoclasts increased in stock diet group treated with fluoride, low calcium diet group,and low calcium diet group treated with fluoride. But there was no much statistical difference ( P >0 05). Compared with stock diet group, the increase of MMP 9 expression in stock diet group treated with fluoride was statistically significant( P <0 05). Statistical difference also showed between low calcium diet group and low calcium diet group treated with fluoride ( P <0 05),and there were significant diffiences when both above were compared with stock diet group ( P <0 05). Conclusion The results prove that excessive fluoride can induce accelerative bone turnover and active bone resorption, and low calcium diet is an important promotive factor of skeletal fluorosis. Fluoride can enhance the activity of osteoclastic resorption by increasing the expression of MMP 9.
