摘要
本试验以五个籼稻品种为材料,研究成熟种子经200Gy^(137)Csγ射线、2mM NaN_3单独和复合处理后的体细胞培养反应。试验结果表明,γ射线和NaN_3单独处理降低种子出愈率,愈伤组织生长率和分化率,γ射线+NaN_3复合处理明显降低种子出愈率、初代培养愈伤组织生长率和分化率,其处理效应高于单独处理效应。但第2、3次培养时,γ射线处理效应大于γ射线+NaN_3复合处理效应。根据本试验结果。认为在以水稻成熟种胚作外植体时,培养前采用200Gy^(137)Csγ射线+2mM NaN_3进行复合处理是可行的。
Mature rice seeds of 5 varieties were used as explants for in vitro culture to study the effects of 200 Gy ^(137) Cs γ—rays combined with NaN^3 on somatic cultures.Decreased callus induction rate, callus growth rate and callus differentiation rate (especially in the 1st culture) were observed in the combined treatments of γ-ray and NaN_3.The mutagenic effects of γ-rays treatment were much higher than those of NaN_3.However, mutagenic effects of treatments with γ-rays were much higher than those of combined treatment of γ—rays and NaN_3 in the 2nd and the 3rd culture.The results suggested that combined treatments of 200 Gy ^(137) Cs γ—rays and 2mM NaN_3 were suitable for explant treatment in rice before culture.
