基质金属蛋白酶-10在结肠癌侵袭转移中作用的研究 被引量:1

Study of the effects of matrix metalloproteinase 10 on invasion and metastasis in colon carcinoma

摘要 目的探讨基质金属蛋白酶-10(MMP-10)在结肠癌中的表达特点及其与临床病理的关系。方法采用免疫组织化学SP法检测结肠腺癌MMP-10、MMP-9、血管内皮生长因子(VEGF)、Ⅳ型胶原及FⅧAg的表达情况。结果结肠癌MMP-10、MMP-9和VEGF的阳性率分别为64%、71%和80%。3者呈正相关共同作用于肿瘤间质血管,致微血管密度(MVD)增多。MMP-10的阳性表达强度较MMP-9弱,MMP-10具有激活MMP-9的功能。MMP-10的表达与肿瘤的侵袭、分化程度及淋巴结转移有关,但与患者的发病部位、性别和组织分型无关。结论MMP-10、VEGF和MMP-9的表达与结肠癌的侵袭转移密切相关。 Objective To investigate the expression of matrix metalloproteinase 10 (MMP 10) and its relationship with clinicopathological features in colon carcinoma. Methods Immunohistochemistry(streptomycin avidin biotinperoxidase complex,SP) was used to detect the expression of MMP 10, MMP 9, vascular endothelial growth factor (VEGF), collagen Ⅳand FⅧAg in colon carcinoma. Results The positive rates of MMP 10, MMP 9, VEGF in colon carcinoma were 64%,71%,80%respectively. MMP 10, MMP 9, VEGF were positively correlated with each other, leading to MVD increase. The expression of MMP 10 was correlated with tumor invasion, differentiation and lymph node metastasis, but not with patients age, sex and histotype. Conclusions MMP 10 can degrade extracellular matrix, and play a role in occurrence and development of colon carcinoma. The expression of MMP 10 and MMP 9 is significantly correlated with metastasis and invasion of colon carcinoma.
