摘要
目的复制碘缺乏病大鼠模型,研究胚胎期及新生期缺碘、甲状腺激素不足对海马神经细胞原癌基因c鄄fos表达的影响,并探讨其在缺碘性学习记忆低下中可能的机制。方法以碘缺乏病病区粮食加去离子水复制碘缺乏病动物模型,以免疫组化方法研究第一代第二、三胎大鼠海马神经细胞原癌基因c鄄fos表达。结果缺碘组子代大鼠尿碘及血清T3、T4均较同龄正常对照组大鼠明显降低,海马神经细胞原癌基因c鄄fos阳性表达百分率较对照组大鼠海马明显增多(33.74±7.51)/(18.56±4.97),P<0.05。结论胚胎期与新生期缺碘、甲状腺激素不足导致海马神经细胞原癌基因c鄄fos表达明显增加,这种增加的原癌基因c鄄fos表达可能参与了缺碘、甲状腺激素不足,导致海马神经元损伤及其功能、代谢的变化乃至学习记忆低下。
Objective To study the expression of oncogene c-fos in hippocampus of rats suffered iodine deficiency (ID) and hypothyroidism. Methods Using foods from iodine deficiency area to make animal model. Immunohistochemistry method was used to study the expression of oncogene c-fos in hippocampus. Results Urine iodine and serum T3, T4 were significantly decreased in ID rats. Oncogene c-fos expression was significantly higher in hippocampus of ID group than that in control group(P < 0.05). Conclusions Iodine deficiency and hypothyroidism during their fetus period could result in high oncogene c-fos expression in hippocampus of rats. These changes may be involved in the dysfunction of hippocampus and impair of learning and memory in ID rats.
