摘要
目的 建立测定血清中苯巴比妥(PB)、苯妥英(PT)、卡马西平(CBZ)、氯硝西泮(CZP)和10,11-环氧卡马西平(ECBZ)浓度的高效液相色谱法。方法 血清中的这几种药物在弱碱性条件下经叔丁基甲醚提取,以α-溴苯乙酮为内标,流动相为甲醇-乙腈-正己烷(55:10:35),紫外检测波长230 nm的RP-HPLC测定。结果 在一定的浓度范围内,被检测药物(PB,PT,CBZ,CZP,ECBZ)分别和内标的峰面积之比与浓度呈良好的线性关系,回收率均高于95%,日内和日间RSD均小于5%。结论 本法准确、灵敏、选择性强、重现性好、快速、简便,适用于抗癫痫药物和茶碱血药浓度的临床和科研测定。
OBJECTIVE: To determine concentrations of phenobarbital (PB), phenytoin (PT), carbamazepine (CBZ), 10, 11-exoxy carbamazepine (ECBZ) and clonazepam (CZP) in serum by high performance liquid chromatography. METHODS: These drugs in serum were extracted by tertbutylmathyl ether under alkaline condition. α-bromoacetophenone (α-BrP) was adopted as the internal standard (IS). The mobile phase was composed of methanol-acetonitrile-n-hexane (55:10:35). The analysis was taken at 230 nm. RESULTS: The peak area ratios of every drug vs. internal standard-concentration curves were linear within a certain range. The recoveries were more than 95%. The relative standard deviations of within-day and between-day were less than 5%. CONCLUSION: The method is sensitive, accurate, selective, rapid and simple. It was suitable for the clinical research on antiepileptic drugs.
