摘要
以紫薇叶片为材料,利用改进的酚—氯仿法,提取到了高质量的紫薇叶片DNA,并建立了20个紫薇品种和南紫薇的AFLP银染反应体系,首次在紫薇DNA提取、酶切连接、预扩增、选择性扩增等实验过程取得了成功,得到了清晰的紫薇品种的AFLP指纹图谱,为紫薇基因库的建立、优良品种选育以及亲缘关系的系列研究奠定了理论基础.
The study dealt with extraction of DNA with phenol-cholorofore method from L.indica leaves and established AFLP fingerprinting in 20 L.indica cultures and L.subcostata. indica Clear and high resolution DNA fingerprints were obtained,which means success in DNA extraction, restriction_ligase reaction, pre_amplification and staining techniques. The aim of this paper is to lay a foundation for the establishment of gene pool, selection and breeding of eminent cultivars and genetic relationship in L. indica.
