The effect of human nerve growth factor(hNGF) on experimental diabetic neuropathy

摘要 目的 观察人神经生长因子 (hNGF)对早期糖尿病神经病变 (DN)大鼠的治疗作用。 方法 用链脲佐菌素 (STZ)一次性腹腔注射诱导建立糖尿病大鼠模型 ,在DN造模后 4周腹腔注射hNGF(90U·kg-1 ·d-1 ) ,共 4周。观察摆尾温度阈值 (TTT)、坐骨神经运动 (MNCV)、感觉神经传导速度 (sNCV )、形态和NGFmRNA表达。 结果 DN 8周大鼠TTT升高 ,MNCV和SNCV减慢 ,形态学明显异常 ,NGFmRNA表达减少 ;hNGF治疗 4周 ,DN大鼠TTT明显降低 (44 5℃± 0 5℃和46 0℃± 0 5℃ ,P <0 0 5) ,MNCV〔(41 6± 2 5)m /s和 (3 1 7± 3 5)m/s ,P <0 0 0 1〕和SNCV〔(43 7± 4 3 )m /s和 (3 4 8± 3 4)m /s ,P <0 0 1〕明显加快 ,形态学明显改善 ,并且NGFmRNA表达增加。 结论 hNGF能改善早期DN ,可能与其促进NGFmRNA表达有关。 Objective To observe the effect of human nerve growth factor(hNGF) on diabetic neuropathy rats. Methods Diabetic rats were induced by a single intraperitoneal injection of streptozoto cin. Four weeks after the inducement of diabetes, hNGF(90 U·kg -1 ·d -1 ·) was intraperitoneally injected for 4 weeks. Tailflick threshold tempera ture(TTT) was tested, motor and sensory nerve conduction velocities(MNCV and SN CV), morphology and level of NGF mRNA in sciatic nerve were measured. Results After 8 weeks of diabetes, increased tailflicting temperature, decreased nerve conduction velocity, morphological abnormalities and decreased NGF mRNA level in sciatic nerve were observed. After treatment with hNGF for 4 weeks, TTT was sig nificantly decreased(44 5℃±0 5℃ vs 46 0℃±0 5℃, P<0 05), MNCV and SNCV were significantly increased 〔(41 6±2 5 m/s vs 31 7±3 5 m/s, P<0 001 and 43 7±4 3) m/s vs (34 8±3 4) m/s,P<0 01〕, morpholo gical abnormalities in sciatic nerve were improved, and expression of NGF mRNA w as significantly increased. Conclusions Diabetic neuropathy is improved by hNGF. The increased expression of NGF mRNA i n sciatic nerve might contribute to the improvement.
