摘要
本实验应用分散大鼠垂体前叶细胞体外灌流培养方法,观察了β-内啡肽及其拮抗剂——纳洛酮对垂体细胞 LH 分泌的影响。结果表明:(1)β-内啡肽抑制了 LHRH(GnRH)-诱发的 LH 分泌,使LH 分泌峰值降低,净释放量/脉冲减少。(2)纳洛酮不仅能阻断β-内啡肽的抑制作用,还可直接增加GnRH-诱发的 LH 分泌,使 LH 峰值升高,净释放量/脉冲增加。(3)β-内啡肽及纳洛酮对 LH 的基础分泌无显著影响。本实验通过从预培养到灌流期间均用含β-内啡肽的培养液以延长作用时间,与灌流时才用β-内啡肽处理的细胞相对比,作用无明显的时间依赖关系。本研究结果为β-内啡肽在垂体水平的直接作用提供了更多的证据。
Using perifusion culture of dispersed rat anterior
pituitary cells,the effect of/3-endorphin,and naloxone
on LH secretion was investigated.The results showed
that β-endorphin reduced GnRH-induced peak LH and
total LH release/pulse.Naloxone,counteracted the in-
hibitory effect of β-endorphin,and a direct stimulatory
effect of naloxone on GnRH-induced LH secretion was
also observed.β-endorphin and naloxone did not signif-
icantly affect basal LH secretion.There was no signifi-
cant difference in LH secretion between pituitary cells,
preincubeted with β-endorphin for 48 hours and then
followed by perifusion with medium containing β-en-
dorphin,and those without preincubation.The result
suggested that there is no apparent time-dependent in-
hibitory effect of β-endorphin.Our data demonstreted
that β-endorphin can exert a direct inhibitory effect on
GnRH-induced LH secretion on pituitary cells in vitro.
