摘要
目的 探讨p73基因在儿童急性淋巴细胞白血病 (ALL)中的表达及临床意义。方法 RT-PCR法检测了 45例儿童ALL患儿骨髓单个核细胞内p73mRNA的表达 ,并分析了p73mRNA阴性表达与影响儿童ALL预后的临床特点的关系。结果 儿童ALL患者中存在较高频率的p73mRNA阴性表达 ,且与高危年龄、高白细胞数、高肿瘤负荷、髓外浸润、高危临床分型显著相关。结论 p73mRNA阴性表达在儿童ALL的发病过程中可能起着重要作用 ,p73mRNA阴性表达者与预后不良因素相关 ,p73mR NA检测可能对评估儿童ALL预后、指导临床治疗有重要意义。
Objective To investigate the expression of p73 gene in childhood acute lymphoblastic leukemic cells and the clinical significance in childhood ALL. Methods The level of p73 transcripts was detected by using reverse transcriptase polymerase chain reaction (RT-PCR) technique . And their correlation with the clinical characteristics including age, sex, the leucocyte count, extramedullary infiltration, the value of the leukemic cell mass risk factor (RF), clinical phenotype was analyzed. Results Of the 45 primary childhood ALL samples, 32 showed expression of p73 mRNA, with a higher negative rate of 28.9% (13/45), and negative expression for p73 mRNA was significantly associated with age(<2 years old or>10 years old), the higher leucocyte count (≥25×10 9/L) , the higher value of the leukemic cell mass risk factor (RF≥1.2), extramedullary infiltration and high-risk ALL (P= 0.019, 0.008, 0.004, 0.027, 0.011, respectively). Conclusions These findings suggested that p73 gene inactivation might play an important role in the genesis of childhood ALL. Examination of p73 mRNA might have clinical significance in predicting prognosis of childhood ALL.
