摘要
目的 观察重组防御素 5 (r HD- 5 )对 6株肠道常驻细菌的杀菌效果。方法 通过冷冻干燥获得含有 r HD- 5的细胞培养上清 ( 液 )、5倍浓缩液 ( 液 )和 10倍浓缩液 ( 液 ) ,以不含 r HD- 5的培养上清 (O液 )为对照 ,观察了它们对大肠埃希菌 (ATCC 2 5 92 2 )、金黄色葡萄球菌 (ATCC 2 5 92 3)、铜绿假单胞菌 (ATCC 2 785 3) ,表皮葡萄球菌、铜绿假单胞菌、鲍氏不动杆菌野生株的抑菌环直径大小。结果 随着 r HD- 5浓度的逐渐增加 ,O、 、 、 液对大肠埃希菌的抑菌环直径分别为 0 mm、(15 .78± 0 .6 7) m m、(31.11± 0 .89) m m、(36 .94± 0 .77) mm ;对金黄色葡萄球菌 (ATCC 2 5 92 3)的抑菌环直径分别为 0 mm、 (18.6 7± 0 .35 ) mm、(2 7.11± 0 .5 5 ) mm、(30 .72± 0 .4 4 )mm ;仅 液对铜绿假单胞菌 (ATCC 2 785 3)产生了抑菌环 (16 .6 7± 0 .79mm ) ;O、 、 、 液对表皮葡萄球菌野生株的抑菌环直径分别为 0 m m、0 mm、(15 .6 7± 0 .5 0 ) m m、(30 .5 6± 0 .5 8) mm ;而仅有 液对鲍氏不动杆菌野生株产生了抑菌环 (14 .4 4± 0 .77) m m ,对铜绿假单胞菌野生株均没有抑菌环产生。结论 r HD- 5对 6种肠道常驻细菌具有不同程度的杀菌活性 ,提示其在防治肠源性感染中具有潜在的临床应用价?
OBJECTIVE To observe bactericidal effects of recombinant human defensin 5 (rHD-5) on six intestinal resident bacteria. METHODS One-fold-,5-fold-,10-fold-condensed solutions (Solution Ⅰ,Ⅱ,Ⅲ with different rHD-5 concentration) were obtained by lyophilization from CHO-K1 cell culture supernatant,which were transferred by recombinant plasmids(pcDNA3.1 (+) -DEFA5). The untransferred CHO-K1 cell culture supernatant(solution O) was a control. By determining the diameter of antibacterial annulus,bactericidal effects of the four above-mentioned solutions were observed in the bactericidal test of six bacteria,such as Escherichia coli (ATCC 25922),Staphylococcus aureus (ATCC 25923),Pseudomonas aeruginosa (ATCC27853),wild S. epidermidis ,wild P. aeruginosa and wild Acinetobacter baumanii . RESULTS Diameters of antibacterial annulus of the solutions(O,Ⅰ,Ⅱ,Ⅲ) to E. coli (0,15.78±0.67,31.11±0.89,36.94±0.77) and S. aureus (0,18.67± 0.35,27.11±0.55,30.72±0.44) increased along with the increasing in rHD-5 concentration. Although there was no such tendency of them to P. aeruginosa (ATCC 27853),the diameter of antibacterial annulus of the solution Ⅲ was (16.67±0.79) mm. The antibacterial diameter(mm) of the four solutions to S. epidermidis were 0,0,15.67±0.50,30.56±0.58,respectively,while the bactericidal effect was observed only in solution Ⅲ to wild A. baumanii and no bactericidal effect of the four solutions to wild P. aeruginosa was found. CONCLUSIONS Different bactericidal activity of rHD-5 to six intestinal resident bacteria shows that potential clinical application value of rHD-5 would be beneficial to the prevention and treatment of enterogenic infection.
